A new mixed-anion crystal composed of BEDT-TTF radical cation salt [BEDT-TTF]2(CuBr2)0.4(CuCl2)0.6 with an α'-type donor arrangement with a formal charge of +0.5 per BEDT-TTF was prepared by using a chemical oxidation method and characterized by using X-ray diffraction, four-probe electrical resistivity measurements (semiconductor: ρrt = 2 × 102 Ω cm, Ea = 0.2 eV), and energy band calculations. The results showed that this system had a quasi-one dimensional Fermi surface.

Molecules, Vol. 17, Pages 4755-4769: Dynamic Action of Carotenoids in Cardioprotection and Maintenance of Cardiac Health

Posted: 23 Apr 2012 12:00 AM PDT

Oxidative stress has been considered universally and undeniably implicated in the pathogenesis of all major diseases, including those of the cardiovascular system. Oxidative stress activate transcriptional messengers, such as nuclear factor—κB, tangibly contributing to endothelial dysfunction, the initiation and progression of atherosclerosis, irreversible damage after ischemic reperfusion, and even arrhythmia, such as atrial fibrillation. Evidence is rapidly accumulating to support the role of reactive oxygen species (ROS) and reactive nitrogen species (RNS) as intracellular signaling molecules. Despite this connection between oxidative stress and cardiovascular disease (CVD), there are currently no recognized therapeutic interventions to address this important unmet need. Antioxidants that provide a broad, "upstream" approach via ROS/RNS quenching or free radical chain breaking seem an appropriate therapeutic option based on epidemiologic, dietary, and in vivo animal model data. Short-term dietary intervention trials suggest that diets rich in fruit and vegetable intake lead to improvements in coronary risk factors and reduce cardiovascular mortality. Carotenoids are such abundant, plant-derived, fat-soluble pigments that functions as antioxidants. They are stored in the liver or adipose tissue, and are lipid soluble by becoming incorporated into plasma lipoprotein particles during transport. For these reasons, carotenoids may represent one plausible mechanism by which fruits and vegetables reduce the risk of chronic diseases as cardiovascular disease (CVD). This review paper outlines the role of carotenoids in maintaining cardiac health and cardioprotection mediated by several mechanisms including redox signaling.

Molecules, Vol. 17, Pages 4733-4754: Simultaneous Determination of Salidroside and Its Aglycone Metabolite p-Tyrosol in Rat Plasma by Liquid Chromatography-Tandem Mass Spectrometry

Posted: 23 Apr 2012 12:00 AM PDT

Salidroside and its aglycone p-tyrosol are two major phenols in the genus Rhodiola and have been confirmed to possess various pharmacological properties. In our present study, p-tyrosol was identified as the deglycosylation metabolite of salidroside after intravenous (i.v.) administration to rats at a dose of 50 mg/kg, but was not detectable after intragastric gavage (i.g.) administration through HPLC-photodiode array detection (PDA) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. Next, an accurate and precise LC-MS/MS method was developed to quantitatively determine salidroside and p-tyrosol in rat plasma samples. Samples were analyzed by LC-MS/MS on a reverse-phase xTerra MS C18 column which was equilibrated and eluted with an isocratic mixture of acetonitrile-water (1:9, v/v) at a flow rate of 0.3 mL/min. The analytes were monitored by multiple reaction monitoring (MRM) under the negative electrospray ionization mode. The precursor/product transitions (m/z) were 299.0→118.8 for salidroside, 137.0→118.9 for p-tyrosol and 150.1→106.9 for the internal standard (IS), paracetamol, respectively. The calibration curve was linear over the concentration ranges of 50–2,000 ng/mL for salidroside and 20–200 ng/mL for p-tyrosol. The inter- and intra-day accuracy and precision were within ±15%. The method has been successfully applied to the pharmacokinetic study and the oral bioavailability was calculated.

Cells, Vol. 1, Pages 35-50: Cytomegalovirus (CMV)-Specific Perforin and Granzyme B ELISPOT Assays Detect Reactivation of CMV Infection in Inflammatory Bowel Disease

Posted: 23 Apr 2012 12:00 AM PDT

The role of cytomegalovirus (CMV) infection in the pathogenesis and exacerbation of Inflammatory Bowel Disease (IBD) has been unresolved. Typically, the CMV genome remains dormant in infected cells, but a breakdown of immune surveillance can lead to re-activation of viral replication in the gut mucosa, which is not necessarily associated with viremia or changes in antibody titers. We hypothesized that the detection of CMV-specific CD8 effector T cells should permit the distinction between dormant and active CMV infection. As CD8 effector T cells, unlike memory CD8 T cells, have perforin (PFN) and granzyme B (GzB) preformed in their cytoplasmic granules, we employed single cell resolution ELISPOT assays to measure the CMV antigen-triggered release of these molecules by CD8 T cells isolated from subjects with IBD, and age-matched healthy controls. The frequencies of CMV-specific (GzB) and PFN-producing CD8 T cells were increased in IBD patients compared to healthy controls. Furthermore, the increased CMV reactivity was associated with active IBD disease and with longer disease duration. Notably, PCR on serum frequently failed to detect CMV DNA during flares. The data show that during active IBD there is a flare of CD8 T cell activity against CMV in a substantial proportion of IBD patients, suggesting CMV reactivation that serum PCR does not detect. While it remains open whether CMV reactivation is a cause or consequence of IBD, our data suggest that monitoring CMV antigen-specific effector CD8 T cells with GzB and PFN ELISPOT analysis can provide novel insights into the role of CMV infection in IBD. Additionally, our data have implications for the fields of transplantation, HIV, cancer, and autoimmune diseases, in all of which patient care critically depends on sensitive and reliable detection of a reactivation of CMV infection.

Polymers, Vol. 4, Pages 1084-1108: Microfluidic-Based Synthesis of Hydrogel Particles for Cell Microencapsulation and Cell-Based Drug Delivery

Posted: 23 Apr 2012 12:00 AM PDT

Encapsulation of cells in hydrogel particles has been demonstrated as an effective approach to deliver therapeutic agents. The properties of hydrogel particles, such as the chemical composition, size, porosity, and number of cells per particle, affect cellular functions and consequently play important roles for the cell-based drug delivery. Microfluidics has shown unparalleled advantages for the synthesis of polymer particles and been utilized to produce hydrogel particles with a well-defined size, shape and morphology. Most importantly, during the encapsulation process, microfluidics can control the number of cells per particle and the overall encapsulation efficiency. Therefore, microfluidics is becoming the powerful approach for cell microencapsulation and construction of cell-based drug delivery systems. In this article, I summarize and discuss microfluidic approaches that have been developed recently for the synthesis of hydrogel particles and encapsulation of cells. I will start by classifying different types of hydrogel material, including natural biopolymers and synthetic polymers that are used for cell encapsulation, and then focus on the current status and challenges of microfluidic-based approaches. Finally, applications of cell-containing hydrogel particles for cell-based drug delivery, particularly for cancer therapy, are discussed.

IJMS, Vol. 13, Pages 5138-5162: Identification of Important Chemical Features of 11β-Hydroxysteroid Dehydrogenase Type1 Inhibitors: Application of Ligand Based Virtual Screening and Density Functional Theory

Posted: 23 Apr 2012 12:00 AM PDT

11ß-Hydroxysteroid dehydrogenase type1 (11ßHSD1) regulates the conversion from inactive cortisone to active cortisol. Increased cortisol results in diabetes, hence quelling the activity of 11ßHSD1 has been thought of as an effective approach for the treatment of diabetes. Quantitative hypotheses were developed and validated to identify the critical chemical features with reliable geometric constraints that contribute to the inhibition of 11ßHSD1 function. The best hypothesis, Hypo1, which contains one-HBA; one-Hy-Ali, and two-RA features, was validated using Fischer's randomization method, a test and a decoy set. The well validated, Hypo1, was used as 3D query to perform a virtual screening of three different chemical databases. Compounds selected by Hypo1 in the virtual screening were filtered by applying Lipinski's rule of five, ADMET, and molecular docking. Finally, five hit compounds were selected as virtual novel hit molecules for 11ßHSD1 based on their electronic properties calculated by Density functional theory.

IJMS, Vol. 13, Pages 5125-5137: A DNA Network as an Information Processing System

Posted: 23 Apr 2012 12:00 AM PDT

Biomolecular systems that can process information are sought for computational applications, because of their potential for parallelism and miniaturization and because their biocompatibility also makes them suitable for future biomedical applications. DNA has been used to design machines, motors, finite automata, logic gates, reaction networks and logic programs, amongst many other structures and dynamic behaviours. Here we design and program a synthetic DNA network to implement computational paradigms abstracted from cellular regulatory networks. These show information processing properties that are desirable in artificial, engineered molecular systems, including robustness of the output in relation to different sources of variation. We show the results of numerical simulations of the dynamic behaviour of the network and preliminary experimental analysis of its main components.

Default Folder X 4.4.11 - Enhances Open and Save dialogs with bountiful options.. (Shareware)

Posted: 23 Apr 2012 02:24 AM PDT

Default Folder X attaches a toolbar to the right side of the Open and Save dialogs in any Mac OS X-native application. The toolbar gives you fast access to various folders and commands. You just click on the buttons to go to your favorite and recently used folders, manage the folders and files shown in the list, and make changes to your settings.

Default Folder also fixes a number of problems in Open and Save dialogs, "rebounding" back to the last selected file, putting the path listing back in the top menu, and correcting bugs in scrolling column views.

Check out the screencast by clicking here!

Features:

  • Directory assistant: You can access the contents of your recent, favorite, and open folders right from Default Folder X's toolbar. Its hierarchical pop-up menus expand as you mouse over them, letting you navigate your folders to get to your files. To work faster, you can assign a keyboard shortcut for all of your favorite folders. If an Open and Save dialog isn't open, then you can use Default Folder X's menu bar, Dock icon, or keyboard shortcuts.
  • Real time saving: If you're constantly saving things inside the same folder, Default Folder X can remember that folder for you. You can even set a different default folder for each application you use. That way, you'll already be in the right folder when you're working.
  • Finder management: Have you ever wanted to save a file to an open Finder window? Default Folder X lets you open up or save to any Finder window with a single click. It also lets you get info on, rename, and delete existing files, putting the Finder's power in every Open and Save dialog.
  • Instant Recall: To help you quickly return to folders you've recently used, Default Folder X remembers them for you. It can even rebound to the last file you used in all your recent folders. With Default Folder X, you'll never forget where you put something.
  • Spotlight on the spot: Mac OS X delivers amazing search capabilities with Spotlight, but adding your own search keywords requires a trip to the Finder's Get Info window or to an application like Tags, Fresh, or Tag Folders. No more! Default Folder X lets you add Spotlight keywords, OpenMeta tags, and Finder labels on the spot, while you're still in the Save dialog. It even remembers keywords you've used before and automatically completes them as you type.
  • Information central: Leopard delivers amazing previews with QuickLook, but in Open dialogs they're small and you can only see them in column view. With Default Folder X you get previews below every Open dialog, expanded to fill the available space. Need to see more detail? Click on the image and it zooms up to full size. And if you'd rather have words instead of pictures, Default Folder X delivers those details too. The file's size, the date you created and modified it, the Finder label, Spotlight comments, OpenMeta tags, access permissions - they're all there too.
  • Workflow without work: Rather than make you learn and set up a new application, Default Folder X blends into Mac OS X. Its pop-up menus and Finder integration will start saving you time immediately, and it will put your pre-existing Favorite folders in its toolbar right away. When you do decide to start customizing it, Default Folder X lets you set default folders and assign shortcuts right from its toolbar. Finally, effortless workflow.

Version 4.4.11:
  • Fixed a problem with Default Folder X not working in sandboxed applications (Preview and TextEdit) in the developer release of Mountain Lion.
  • Worked around a bug in OS X's codesign utility that created an empty file named "/-" on your system drive.
  • Added support for CodeKit.

  • Mac OS X 10.5 or later

Games, Vol. 3, Pages 78-96: What Behaviors are Disapproved? Experimental Evidence from Five Dictator Games

Posted: 23 Apr 2012 12:00 AM PDT

The literature on social norms has often stressed that social disapproval is crucial to foster compliance with norms and promote fair and cooperative behavior. With this in mind, we explore the disapproval of allocation decisions using experimental data from five dictator games with a feedback stage. Our data suggests that subjects are heterogeneous in their disapproval patterns, distinguishing two main groups: (1) Subjects who only disapprove choices that harm them, and (2) subjects who disapprove socially inefficient choices.
