IJERPH, Vol. 8, Pages 1126-1140: Assessment of Water Quality in a Subtropical Alpine Lake Using Multivariate Statistical Techniques and Geostatistical Mapping: A Case Study

Posted: 15 Apr 2011 12:00 AM PDT

Concerns about the water quality in Yuan-Yang Lake (YYL), a shallow, subtropical alpine lake located in north-central Taiwan, has been rapidly increasing recently due to the natural and anthropogenic pollution. In order to understand the underlying physical and chemical processes as well as their associated spatial distribution in YYL, this study analyzes fourteen physico-chemical water quality parameters recorded at the eight sampling stations during 2008–2010 by using multivariate statistical techniques and a geostatistical method. Hierarchical clustering analysis (CA) is first applied to distinguish the three general water quality patterns among the stations, followed by the use of principle component analysis (PCA) and factor analysis (FA) to extract and recognize the major underlying factors contributing to the variations among the water quality measures. The spatial distribution of the identified major contributing factors is obtained by using a kriging method. Results show that four principal components i.e., nitrogen nutrients, meteorological factor, turbidity and nitrate factors, account for 65.52% of the total variance among the water quality parameters. The spatial distribution of principal components further confirms that nitrogen sources constitute an important pollutant contribution in the YYL.

IJMS, Vol. 12, Pages 2518-2542: Functioning Nanomachines Seen in Real-Time in Living Bacteria Using Single-Molecule and Super-Resolution Fluorescence Imaging

Posted: 15 Apr 2011 12:00 AM PDT

Molecular machines are examples of "pre-established" nanotechnology, driving the basic biochemistry of living cells. They encompass an enormous range of function, including fuel generation for chemical processes, transport of molecular components within the cell, cellular mobility, signal transduction and the replication of the genetic code, amongst many others. Much of our understanding of such nanometer length scale machines has come from in vitro studies performed in isolated, artificial conditions. Researchers are now tackling the challenges of studying nanomachines in their native environments. In this review, we outline recent in vivo investigations on nanomachines in model bacterial systems using state-of-the-art genetics technology combined with cutting-edge single-molecule and super-resolution fluorescence microscopy. We conclude that single-molecule and super-resolution fluorescence imaging provide powerful tools for the biochemical, structural and functional characterization of biological nanomachines. The integrative spatial, temporal, and single-molecule data obtained simultaneously from fluorescence imaging open an avenue for systems-level single-molecule cellular biophysics and in vivo biochemistry.

JLPEA, Vol. 1, Pages 131-149: Path Specific Register Design to Reduce Standby Power Consumption

Posted: 15 Apr 2011 12:00 AM PDT

A methodology is proposed to design low leakage registers by considering the type of timing path, i.e., short or long, and type of register, i.e., launching or capturing. Three different dual threshold voltage registers are developed where each register trades, depending upon the timing path, a different timing constraint for reducing the leakage current. For example, the first proposed register is used as a launching register in a noncritical path, trading clock-to-Q delay for leakage current. Other timing constraints such as setup and hold times are maintained the same not to introduce any timing violations. Alternatively, the second and third registers, trade, respectively, setup time and hold time for leakage current while maintaining clock-to-Q delay constant. The effect of the proposed methodology on leakage current is investigated for four technology nodes. The overall reduction in the leakage current of a register can exceed 90% while maintaining the clock frequency and other design parameters such as area and dynamic power the same. Three ISCAS 89 benchmark circuits are utilized to evaluate the methodology, demonstrating, on average, 23% reduction in the overall leakage current.

Materials, Vol. 4, Pages 801-815: Structural Consideration in Designing Organotin Polyethers to Arrest the Growth of Breast Cancer Cells In Vitro

Posted: 15 Apr 2011 12:00 AM PDT

The ability to inhibit cancer is inherent in organotin materials yet the structural relationships that regulate/direct this activity remains unknown. We measured antitumor activity using a matched pair of cell lines MDA-MB-231 cells that are estrogen-independent, estrogen receptor negative and MCF-7 cells, a cell line that is estrogen receptor (ER) positive. Those polyethers that contained a O-phenyl unit were able to significantly inhibit the non-estrogen sensitive cell line but were much less effective against the estrogen sensitive cell line; that is, the human breast cancer cell line MDA-MB-231 showed better test results for polymers derived from diols containing the O-phenyl moiety than the breast cancer cell line MCF-7, a well-characterized estrogen receptor positive control cell line. Those polyethers that did not contain the O-phenyl unit inhibited both cell lines approximately the same. The differential activity of the O-phenyl-containing polyethers is likely due to the estrogen-sensitive cells combining with some of the organotin polyethers minimizing their ability to inhibit cell growth.

Micromachines, Vol. 2, Pages 129-149: Mori-Tanaka Based Estimates of Effective Thermal Conductivity of Various Engineering Materials

Posted: 15 Apr 2011 12:00 AM PDT

The purpose of this paper is to present a simple micromechanics-based model to estimate the effective thermal conductivity of macroscopically isotropic materials of matrix-inclusion type. The methodology is based on the well-established Mori-Tanaka method for composite media reinforced with ellipsoidal inclusions, extended to account for imperfect thermal contact at the matrix-inclusion interface, random orientation of particles and particle size distribution. Using simple ensemble averaging arguments, we show that the Mori-Tanaka relations are still applicable for these complex systems, provided that the inclusion conductivity is appropriately modified. Such conclusion is supported by the verification of the model against a detailed finite-element study as well as its validation against experimental data for a wide range of engineering material systems.

Molecules, Vol. 16, Pages 3197-3211: Antioxidant Properties and Antioxidant Compounds of Various Extracts from the Edible Basidiomycete Grifola Frondosa (Maitake)

Posted: 15 Apr 2011 12:00 AM PDT

Grifola frondosa is an edible mushroom currently available in Taiwan. Ethanolic, cold-water and hot-water extracts were prepared and their antioxidant properties were investigated. At 1 mg/mL, G. frondosa T1 and T2 cold-water extracts showed high reducing powers of 1.02 and 0.50, respectively. Chelating abilities on ferrous ions of G. frondosa T1 and T2 were higher for cold-water extracts than for ethanolic and hot-water extracts. For the scavenging ability on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, G. frondosa T1 and T2 extracts were effective in the following order: ethanolic > hot-water > cold-water. The G. frondosa hot-water extract showed high scavenging ability on superoxide anions. Total phenols, flavonoids, ascorbic acid and α-tocopherol are the major antioxidant components found in the various G. frondosa extracts. Based on EC50 values (

Molecules, Vol. 16, Pages 3179-3196: Induction of Intracellular Ca2+ and pH Changes in Sf9 Insect Cells by Rhodojaponin-III, A Natural Botanic Insecticide Isolated from Rhododendron molle

Posted: 15 Apr 2011 12:00 AM PDT

Many studies on intracellular calcium ([Ca2+]i) and intracellular pH (pHi) have been carried out due to their importance in regulation of different cellular functions. However, most of the previous studies are focused on human or mammalian cells. The purpose of the present study was to characterize the effect of Rhodojaponin-III (R-III) on [Ca2+]i and pHi and the proliferation of Sf9 cells. R-III strongly inhibited Sf9 cells proliferation with a time- and dose-dependent manner. Flow cytometry established that R-III interfered with Sf9 cells division and arrested them in G2/M. By using confocal scanning technique, effects of R-III on intracellular free calcium ([Ca2+]i) and intracellular pH (pHi) in Sf9 cells were determined. R-III induced a significant dose-dependent (1, 10, 100, 200 μg/mL) increase in [Ca2+]i and pHi of Sf9 cells in presence of Ca2+-containing solution (Hanks) and an irreversible decrease in the absence of extra cellular Ca2+. We also found that both extra cellular Ca2+ and intracellular Ca2+ stores contributed to the increase of [Ca2+]i, because completely treating Sf9 cells with CdCl2 (5 mM), a Ca2+ channels blocker, R-III (100 μg/mL) induced a transient elevation of [Ca2+]i in case of cells either in presence of Ca2+ containing or Ca2+ free solution. In these conditions, pHi showed similar changes with that of [Ca2+]i on the whole. Accordingly, we supposed that there was a certain linkage for change of [Ca2+]i, cell cycle arrest, proliferation inhibition in Sf9 cells induced by R-III.
