Viruses, Vol. 4, Pages 1104-1115: Legume Lectins Inhibit Human Parainfluenza Virus Type 2 Infection by Interfering with the Entry

Three lectins with different sugar binding specificities were investigated for anti-viral activity against human parainfluenza virus type 2 (hPIV-2). The lectins, concanavalin A (Con A), lens culinaris agglutinin (LCA) and peanut agglutinin (PNA), inhibited cell fusion and hemadsorption induced by hPIV-2. Virus nucleoprotein (NP) gene synthesis was largely inhibited, but fusion (F) and hemagglutinin-neuraminidase (HN) gene syntheses were not. An indirect immunofluorescence study showed that Con A inhibited virus NP, F and HN protein syntheses, but LCA did not completely inhibit them, and that PNA inhibited only NP protein synthesis. Using a recombinant green fluorescence protein-expressing hPIV-2, without matrix protein (rghPIV-2ΔM), it was found that virus entry into the cells was not completely prevented. The lectins considerably reduced the number of viruses released compared with that of virus infected cells. The lectins bound to cell surface within 10 min, and many aggregates were observed at 30 min. Con A and LCA slightly disrupted actin microfilaments and microtubules, but PNA had almost no effect on them. These results indicated that the inhibitory effects of the lectins were caused mainly by the considerable prevention of virus adsorption to the cells by the lectin binding to their receptors.

Chronic bee paralysis virus (CBPV) infection causes chronic paralysis and loss of workers in honey bee colonies around the world. Although CBPV shows a worldwide distribution, it had not been molecularly detected in Japan. Our investigation of Apis mellifera and Apis cerana japonica colonies with RT-PCR has revealed CBPV infection in A. mellifera but not A. c. japonica colonies in Japan. The prevalence of CBPV is low compared with that of other viruses: deformed wing virus (DWV), black queen cell virus (BQCV), Israel acute paralysis virus (IAPV), and sac brood virus (SBV), previously reported in Japan. Because of its low prevalence (5.6%) in A. mellifera colonies, the incidence of colony losses by CBPV infection must be sporadic in Japan. The presence of the (−) strand RNA in dying workers suggests that CBPV infection and replication may contribute to their symptoms. Phylogenetic analysis demonstrates a geographic separation of Japanese isolates from European, Uruguayan, and mainland US isolates. The lack of major exchange of honey bees between Europe/mainland US and Japan for the recent 26 years (1985–2010) may have resulted in the geographic separation of Japanese CBPV isolates.

Elongation factor (EF) is a key regulation factor for translation in many organisms, including plants, bacteria, fungi, animals and insects. To investigate the nature and function of elongation factor 1β' from Spodoptera exigua (SeEF-1β'), its cDNA was cloned. This contained an open reading frame of 672 nucleotides encoding a protein of 223 amino acids with a predicted molecular weight of 24.04 kDa and pI of 4.53. Northern blotting revealed that SeEF-1β' mRNA is expressed in brain, epidermis, fat body, midgut, Malpighian tubules, ovary and tracheae. RT-PCR revealed that SeEF-1β' mRNA is expressed at different levels in fat body and whole body during different developmental stages. In RNAi experiments, the survival rate of insects injected with SeEF-1β' dsRNA was 58.7% at 36 h after injection, which was significantly lower than three control groups. Other elongation factors and transcription factors were also influenced when EF-1β' was suppressed. The results demonstrate that SeEF-1β' is a key gene in transcription in S. exigua.

Synthesis and spectral evaluation of new zinc and copper unsymmetrical mesoporphyrinic complexes are reported. Zn(II)-5-(4-acetoxy-3-methoxyphenyl)-10,15,20-tris-(4-carboxymethylphenyl)porphyrin, Zn(II)-5-[(3,4-methylenedioxy)phenyl]-10,15,20-tris-(4-carboxymethylphenyl)porphyrin, Cu(II)-5-(4-acetoxy-3-methoxyphenyl)-10,15,20-tris-(4-carboxymethylphenyl)porphyrin and Cu(II)-5-[(3,4-methylenedioxy)phenyl]-10,15,20-tris-(4-carboxymethylphenyl)porphyrin were synthesized using microwave-assisted synthesis. The complexes were characterized by elemental analysis, FT-IR, UV-Vis, EPR and NMR spectroscopy, which fully confirmed their structure. The spectral absorption properties of the porphyrinic complexes were studied in solvents with different polarities. Fluorescence emission and singlet oxygen formation quantum yields were evaluated for the compounds under study, revealing high yields for the zinc derivatives. The copper complexes are not emissive and only display residual capacity for singlet oxygen formation.

Monitoring high latitude wetlands is required to understand feedbacks between terrestrial carbon pools and climate change. Hydrological variability is a key factor driving biogeochemical processes in these ecosystems and effective assessment tools are critical for accurate characterization of surface hydrology, soil moisture, and water table fluctuations. Operational satellite platforms provide opportunities to systematically monitor hydrological variability in high latitude wetlands. The objective of this research application was to integrate high temporal frequency Synthetic Aperture Radar (SAR) and high spatial resolution Light Detection and Ranging (LiDAR) observations to assess hydroperiod at a mire in northern Sweden. Geostatistical and polarimetric (PLR) techniques were applied to determine spatial structure of the wetland and imagery at respective scales (0.5 m to 25 m). Variogram, spatial regression, and decomposition approaches characterized the sensitivity of the two platforms (SAR and LiDAR) to wetland hydrogeomorphology, scattering mechanisms, and data interrelationships. A Classification and Regression Tree (CART), based on random forest, fused multi-mode (fine-beam single, dual, quad pol) Phased Array L-band Synthetic Aperture Radar (PALSAR) and LiDAR-derived elevation to effectively map hydroperiod attributes at the Swedish mire across an aggregated warm season (May–September, 2006–2010). Image derived estimates of water and peat moisture were sensitive (R2 = 0.86) to field measurements of water table depth (cm). Peat areas that are underlain by permafrost were observed as areas with fluctuating soil moisture and water table changes.

Medium Spectral Resolution Imaging Spectrometer (MERIS) oxygen A band measurements were used as a proxy for the Earth Polychromatic Imaging Camera (EPIC),to be launched on NASA's Deep Space Climate Observatory (DSCOVR). The high spatial resolution of MERIS (1 × 1 km2) is exploited to study the effects of subscale spatialheterogeneity of clouds on the cloud-top pressure retrieved at the coarser spatial resolutionof EPIC (10 × 10 km2). In general, for a sub-scale cloud fraction less than 1, a shift of cloud-top pressure toward the middle atmosphere is found, with a low-bias for highclouds and a high-bias for low clouds. In addition, the deviation is found to be a function of surface reflectance. The subscale variability of fully clouded EPIC pixels causes a weak underestimation of cloud-top pressure, when compared to averaged high-resolution retrievals.

Woody plant encroachment into grasslands and rangelands is a world-wide phenomenon but detailed descriptions of changes in geographical distribution and infilling rates have not been well documented at large land scales. Remote sensing with either aerial or satellite images may provide a rapid means for accomplishing this task. Our objective was to compare the accuracy and utility of two types of images with contrasting spatial resolutions (1-m aerial and 30-m satellite) for classifying woody and herbaceous canopy cover and determining woody infilling rates in a large area of rangeland (800 km2) in north Texas that has been invaded by honey mesquite (Prosopis glandulosa). Accuracy assessment revealed that the overall accuracies for the classification of four land cover types (mesquite, grass, bare ground and other) were 94 and 87% with kappa coefficients of 0.89 and 0.77 for the 1-m and 30-m images, respectively. Over the entire area, the 30-m image over-estimated mesquite canopy cover by 9 percentage units (10 vs. 19%) and underestimated grass canopy cover by the same amount when compared to the 1-m image. The 30-m resolution image typically overestimated mesquite canopy cover within 225 4-ha sub-cells that contained a range of mesquite covers (1–70%) when compared to the 1-m image classification and was not suitable for quantifying infilling rates of this native invasive species. Documenting woody and non-woody canopy cover on large land areas is important for developing integrated, regional-scale management strategies for rangeland and grassland regions that have been invaded by woody plants.

Fires modify the structure of vegetation communities, the carbon and water cycles, the soil's chemistry, and affect the climate system. Within this context, this work aimed to understand the distribution patterns of burned areas in Brazil, during the period of 2002 to 2010, taking into consideration each one of the six Brazilian biomes (Amazon, Caatinga, Cerrado, Atlantic Forest, Pampa and Pantanal) and the respective major land cover classes. Data from the MODIS MCD45A1 product (burned area), as well as thermal anomalies (MOD14 and MYD14) and precipitation (TRMM), were analyzed according to the 2002 Brazilian official land cover and land use map (PROBIO). The Brazilian savanna biome, known as Cerrado, presented the largest concentration of burned areas detected by MODIS (73%), followed by the Amazon (14%), Pantanal (6%), Atlantic Forest (4%), Caatinga (3%), and Pampa (0,06%) biomes. Indeed, in the years of 2007 and 2010, 90% and 92% of Brazil's burned areas were concentrated in the Cerrado and Amazon biomes, respectively. TRMM data indicated that during these two years there was a significant influence of La Niña, causing low rainfall in the Amazon, Cerrado, Caatinga, and Atlantic Forest biomes. Regarding the land cover classes, approximately 81% of the burned areas occurred over remnant vegetation areas. Although no unequivocal correlation can be established between burned areas and new land conversions, the conspicuous concentration of fire scars, particularly in Amazon–Cerrado transition (i.e., the Arc of Deforestation) is certainly not a simple coincidence. Such patterns and trends corroborate the need of improved territorial governance, in addition to the implementation of systematic fire warning and preventive systems.

South Korea's shutdown law under fire

Korean game companies like NCsoft and Neowiz are leading the charge against the country's Shutdown Law.

 

Parents and game companies are starting to throw lawsuits at the South Korean government concerning the country's Shutdown Law. The law itself prohibits minors under sixteen from playing games for six hours starting at 12 midnight in an effort to use their time more productively.

According to Korean site This Is Game (via Kotaku), game makers that have currently thrown down the gauntlet are NCsoft, Neowiz, and Nexon.

An unverified Korean parent and teenager organization representing culture (possibly the group MoonHwaYunDae) also filed for a lawsuit against the government. On a related note, South Korea's Ministry of Gender Equality and Family said that the law is the "bare minimum" required to protect teenagers from managing their time badly and making unsound decisions based on their upbringing.

Recently, the PSN Store in Korea was affected by the law.

Square Enix has highest salary average among game companies in Japan

Final Fantasy publishers hold top spot, above Sony, Nintendo, and Sega.

 

Japanese salary and income analysing company Nensyu Labo recently revealed a ranking chart for the companies with the highest average salary for its employees (via Andriasang). Square Enix is at the top, with employees getting an average salary of 21.68 million yen peryear ($273,057).

This was followed up by Sony, with an average of 9.23 million yen ($116,253), and Nintendo with 9.14 million yen ($115,117). Sega Sammy Corporation was placed fourth, with a salary average of 8.22 million yen, while Konami and Namco Bandai were just behind with 6.78 and 6.40 million, respectively.

The top ten list is just below:

The nerve tissue mini-hemoglobin from Cerebratulus lacteus (CerHb) displays an essential globin fold hosting a protein matrix tunnel held to allow traffic of small ligands to and from the heme. CerHb heme pocket hosts the distal TyrB10/GlnE7 pair, normally linked to low rates of O2 dissociation and ultra-high O2 affinity. However, CerHb affinity for O2 is similar to that of mammalian myoglobins, due to a dynamic equilibrium between high and low affinity states driven by the ability of ThrE11 to orient the TyrB10 OH group relative to the heme ligand. We present here the high resolution crystal structures of CerHb in the unligated and carbomonoxy states. Although CO binds to the heme with an orientation different from the O2 ligand, the overall binding schemes for CO and O2 are essentially the same, both ligands being stabilized through a network of hydrogen bonds based on TyrB10, GlnE7, and ThrE11. No dramatic protein structural changes are needed to support binding of the ligands, which can freely reach the heme distal site through the apolar tunnel. A lack of main conformational changes between the heme-unligated and -ligated states grants stability to the folded mini-Hb and is a prerequisite for fast ligand diffusion to/from the heme.

Glutamine synthetase (GS) is the key enzyme involved in the assimilation of ammonia derived either from nitrate reduction, N2 fixation, photorespiration or asparagine breakdown. A small gene family is encoding for different cytosolic (GS1) or plastidic (GS2) isoforms in legumes. We summarize here the recent advances carried out concerning the quaternary structure of GS, as well as the functional relationship existing between GS2 and processes such as nodulation, photorespiration and water stress, in this latter case by means of proline production. Functional genomic analysis using GS2-minus mutant reveals the key role of GS2 in the metabolic control of the plants and, more particularly, in carbon metabolism.

This paper describes the adsorption of sodium dodecyl sulfate (SDS) molecules in a low polar solvent on Ge substrate by using Fourier transform infrared-attenuated total reflection (FTIR-ATR) spectroscopy and atomic force microscopy (AFM). The maximum SDS amount adsorbed is (5.0 ± 0.3) × 1014 molecules cm−2 in CHCl3, while with the use of CCl4 as subphase the ability of SDS adsorbed is 48% lower. AFM images show that depositions are highly disordered over the interface, and it was possible to establish that the size of the SDS deposition is around 30–40 nm over the Ge surface. A complete description of the infrared spectroscopic bands for the head and tail groups in the SDS molecule is also provided.

Poly(L-lactic acid) (PLLA) fibers were fabricated by electrospinning. The effects of various electrospinning process parameters on the thermal properties, especially the crystallinity of the electrospun fibers were investigated. Thermal analysis of the fibers revealed that they exhibited degree of crystallinity ranging from 23% to 46% while that for the as-received granules was approximately 37%, suggesting that the crystallinity of electrospun PLLA fibres can be controlled by optimizing the electrospinning process. This finding is very important because crystallinity affects polymer properties such as degradation, stiffness, yield stress, modulus and tensile strength, solubility, optical and electrical properties which will in turn affect the behavior of these materials when they are utilized in energy, environment, defense and security applications. The results presented in this paper show that the degree of crystallinity of the electrospun fibers decreased with increasing the polymer solution concentration. Furthermore, an optimum electrospinning voltage at which maximum degree of crystallinity can be obtained was observed. At voltages higher or lower than the optimum electrospinning voltage, the degree of crystallinity will decrease or increase, respectively. The effect of the needle tip to collector distance (NTCD) on the degree of crystallinity follows no predictable and consistent pattern.

In vitro high-throughput screening was carried out in order to detect new activities for old drugs and to select compounds for the drug development process comprising new indications. Tebrophen, a known antiviral drug, was found to inhibit activities on inflammation and cancer related targets. In primary screening this semisynthetic halogenated polyphenol was identified to inhibit the activities of kinases ZAP-70 and Lck (IC50 0.34 µM and 16 µM, respectively), as well as hydrolase DPPIV (at 80 µM 41% inhibition). Next, it showed no cytotoxic effects on standard cell lines within 24 h. However, tebrophen slowed propagation of breast cancer (MDA-MB-231), osteosarcoma (U2OS) and cervical carcinoma (HeLa), through at least 35 population doublings in a dose-dependent manner. It completely stopped the division of the prostate cancer (PC3) cell line at 50 µM concentration and the cells entered massive cell death in less than 20 days. On the other hand, tebrophen did not influence the growth of normal fibroblasts. According to the measured oxidative burst and estimated in silico parameters its direct antioxidative ability is limited. The obtained results indicate that tebrophen can be considered a promising lead molecule for generating more soluble derivatives with specific anticancer efficacy.

Big in Japan June 18-24: Pokemon Black/White 2

New iteration to Pokemon Black/White bumps PS Vita RPG from top spot; PS Vita sales cool down.

 

Nintendo's Pokemon series never fail to grab Japan by storm sales-wise, and the latest update to the Media Creates sales chart proves to be no exception.

Pokemon Black 2 and Pokemon White 2 for the Nintendo DS took first place on the top 10, bumping down former top dog Persona 4: Golden with 1,561,738 units combined. The new versions of Pokemon Black and Pokemon White introduce new locations in the southwest part of the Unova region, a Pokemon World Tournament that lets players fight every gym leader and champion from past generations of the series, and a Pokewood scenario that lets players pit Pokemon in movies.

Persona 4: Golden was bumped down to third place with 29,293 units sold. Tokyo Jungle clung onto fifth place with 18,715 units sold to slot in at five, while The PS3 versions of Dragon's Dogma and Lollipop Chainsaw capped off the end of the top 10 list with 9,162 units and 7,898 units sold, putting them at nine and ten respectively.

New titles that made their mark for the week were Kenka Bancho Bros. Tokyo Battle Royale for the PSP which sold 21,797 units and took fourth spot, and K-On After School Live!! HD version for the PS3 which sold 13,581 units to place at six. The former is an action game styled like a classic beat-em-up where players control one of the two brothers who has to clean up Tokyo of crime. The latter is an HD update of a PSP rhythm game based on the hit anime series.

On the hardware front, the PlayStation Vita lost its momentum from its previous sales record and dropped down to 13,589 units as a result. The 3DS got a slight increase in sales to 68,056 units.
