Molecules, Vol. 17, Pages 6705-6715: Calculation of the Stabilization Energies of Oxidatively Damaged Guanine Base Pairs with Guanine

DNA is constantly exposed to endogenous and exogenous oxidative stresses. Damaged DNA can cause mutations, which may increase the risk of developing cancer and other diseases. G:C-C:G transversions are caused by various oxidative stresses. 2,2,4-Triamino-5(2H)-oxazolone (Oz), guanidinohydantoin (Gh)/iminoallantoin (Ia) and spiro-imino-dihydantoin (Sp) are known products of oxidative guanine damage. These damaged bases can base pair with guanine and cause G:C-C:G transversions. In this study, the stabilization energies of these bases paired with guanine were calculated in vacuo and in water. The calculated stabilization energies of the Ia:G base pairs were similar to that of the native C:G base pair, and both bases pairs have three hydrogen bonds. By contrast, the calculated stabilization energies of Gh:G, which form two hydrogen bonds, were lower than the Ia:G base pairs, suggesting that the stabilization energy depends on the number of hydrogen bonds. In addition, the Sp:G base pairs were less stable than the Ia:G base pairs. Furthermore, calculations showed that the Oz:G base pairs were less stable than the Ia:G, Gh:G and Sp:G base pairs, even though experimental results showed that incorporation of guanine opposite Oz is more efficient than that opposite Gh/Ia and Sp.

In capacitance-voltage (C-V) measurements, frequency dispersion in high-k dielectrics is often observed. The frequency dependence of the dielectric constant (k-value), that is the intrinsic frequency dispersion, could not be assessed before suppressing the effects of extrinsic frequency dispersion, such as the effects of the lossy interfacial layer (between the high-k thin film and silicon substrate) and the parasitic effects. The effect of the lossy interfacial layer on frequency dispersion was investigated and modeled based on a dual frequency technique. The significance of parasitic effects (including series resistance and the back metal contact of the metal-oxide-semiconductor (MOS) capacitor) on frequency dispersion was also studied. The effect of surface roughness on frequency dispersion is also discussed. After taking extrinsic frequency dispersion into account, the relaxation behavior can be modeled using the Curie-von Schweidler (CS) law, the Kohlrausch-Williams-Watts (KWW) relationship and the Havriliak-Negami (HN) relationship. Dielectric relaxation mechanisms are also discussed.

This study focused on phase separation of activated sludge mixed liquor by flat-sheet membranes of polytetrafluoroethylene (PTFE). A 20 liter working volume lab-scale MBR incorporating immersed PTFE flat-sheet membrane modules with different pore sizes (0.3, 0.5 and 1.0 μm) was operated for 19 days treating a synthetic wastewater. The experiment was interrupted twice at days 5 and 13 when the modules were removed and cleaned physically and chemically in sequence. The pure water permeate flux of each membrane module was measured before and after each cleaning step to calculate membrane resistances. Results showed that fouling of membrane modules with 0.3 μm pore size was more rapid than other membrane modules with different pore sizes (0.5 and 1.0 μm). On the other hand, it was not clear whether fouling of the 0.5 μm membrane module was more severe than that of the 1.0 μm membrane module. This was partly because of the membrane condition after chemical cleaning, which seemed to determine the fouling of those modules over the next period. When irreversible resistance (Ri) i.e., differences in membrane resistance before use and after chemical cleaning was high, the transmembrane pressure increased quickly during the next period irrespective of membrane pore size.

Metal Gear Rising: Revengeance scheduled for early 2013

Platinum Games' action title hitting store shelves early next year; demo to be included with Zone of the Enders HD Collection.

 

Metal Gear Rising: Revengeance has been dated for early 2013.

The news came from Konami's pre-E3 press conference, in which the publisher also unveiled a new gameplay trailer for the Platinum Games-developed action title.

The game's story has changed places in the Metal Gear timeline. Originally, Metal Gear Solid: Rising was intended to take place before the events of Metal Gear Solid 4: Guns of the Patriots, showing how the character of Raiden made his transformation into the cyborg ninja of the 2007 PlayStation 3 hit. However, when Platinum Games was attached to the project, the time frame was moved to several years after Metal Gear Solid 4, to a time when cyborgs are becoming more commonplace in the world.

While Metal Gear Solid: Rising was originally intended to showcase a mix of stealth and action gameplay, Revengeance drops the stealth aspect to focus exclusively on action. Konami also dropped the "Solid" from the title to distance itself from the particular brand of stealth action people associate with the Metal Gear Solid series.

Mirroring a tactic that was used for Metal Gear Solid 2: Sons of Liberty and the original Zone of the Enders, a demo for Metal Gear Rising: Revengeance will be bundled with Zone of the Enders HD Collection when it launches later this year. The demo will also be released on Xbox Live and PlayStation Network at a later date, Konami revealed.

Series creator Hideo Kojima gave a teaser about the future of the Metal Gear series in the Konami presentation, talking about the new Fox game engine. "We're also very excited about the new Fox Engine, which I can't say too much about for now, but please stay tuned." It was recently announced that the next generation of Pro Evolution Soccer will run on the engine.

Dota 2 will be free to play

Valve confirms online battle game will be free; introduces Dota Store and Steam Workshop Integration.

 

Valve today confirmed Dota 2 will be free to play.

The publisher also announced the inclusion of an in-game store, where players will be able to buy gear and customise heroes--however, all items in the store will be cosmetic, and will not affect gameplay.

All heroes will be available free of charge, with Valve writing on the Dota 2 blog: "We believe restricting player access to heroes could be destructive to game design, so it's something we plan to avoid."

Upon opening, the Dota Store will offer items created by Valve and members of the community that may be used immediately and carried over after the public launch of the game. Early access to the game will not be required to purchase items before launch.

However, those wishing to get into the game now can do so by purchasing the Dota 2 Early Access Bundle of items through Steam. The bundle costs $39.99, and includes some items that can be traded.

Valve also announced that Dota 2 is now part of the Steam Workshop, a place for fan-created content that can be voted on by other players and translated to the official game by the Valve development team (with the original creator of the content receiving a slice of the sales from the in-game store).

Valve said over $3.5 million dollars were paid out to Team Fortress 2 fans whose creations became part of the game in the first year.

"The opening of the Dota Store is a big part of our final push to launch," Dota 2 design lead IceFrog said in a statement. "By making the game free to play, we hope to give gamers the ability to decide how--and how much--they want to invest in the game."

Dota 2 is scheduled for PC and Mac release this year. The game combines action and real-time strategy elements in multiplayer matches that pit teams of player-controlled heroes against one another. The game will include integrated voice chat and computer-controlled characters that will take over for players who disconnect during play sessions, as well as a coaching system, where veteran players will have the chance to tutor newcomers.

LucasArts unveils Star Wars 1313

New third-person action adventure franchise set in the Star Wars universe; players to control bounty hunter in criminal underworld beneath Coruscant; no platforms announced; more details at E3 2012.

LucasArts has unveiled a new Star Wars games franchise, making the announcement tonight during Spike's Game Trailers TV show.

Titled Star Wars 1313, the internally-developed third-person action adventure game will see players take on the role of a bounty hunter navigating a subterranean metropolis Level 1313, beneath the surface of the planet Coruscant.

While the game will make its formal debut at next week's Electronic Entertainment Expo (E3), LucasArts announced the game is being development with the help of artists across the Lucasfilm organization, including LucasArts, Industrial Light & Magic, Lucasfilm Animation Ltd. and Skywalker Sound. The game will be based on the Unreal engine and feature full-body performance capture.

Star Wars 1313 will see players utilise "exotic" weapons in a bid to uncover the truth surrounding a criminal conspiracy beneath Coruscant. According to LucasArts, the game will emphasize fast-paced combat--the game's hero will use human skills and gadgets rather than supernatural Force powers. In addition, the game will feature a combination of cover-based combat, platforming and "playable cinematic gameplay".

"Star Wars 1313 dives into a part of the Star Wars mythos that we've always known existed, but never had a chance to visit," LucasArts president Paul Meegan said in a statement.

No details have yet been given on platforms or release date.

The latest entry in the Star Wars franchise was Kinect Star Wars, which was released in April and received a chilly reception by critics. That game is set primarily during the time of Star Wars Episode II: Attack of the Clones, with overlapping parts of the other prequel trilogy films and nods to the original Star Wars trilogy.

LucasArts will reveal more details about Star Wars 1313 at next week's E3 in Los Angeles.
