The peroxisome proliferator-activated receptor-γ coactivators (PGC-1) are transcriptional coactivators with an important role in mitochondrial biogenesis and regulation of genes involved in the electron transport chain and oxidative phosphorylation in oxidative tissues including cardiac tissue. These coactivators are thought to play a key role in the development of obesity, type 2 diabetes and the metabolic syndrome. In this study we have used a combined metabolomic and lipidomic analysis of cardiac tissue from the PGC-1β null mouse to examine the effects of a high fat diet on this organ. Multivariate statistics readily separated tissue from PGC-1β null mice from their wild type controls either in gender specific models or in combined datasets. This was associated with an increase in creatine and a decrease in taurine in the null mouse, and an increase in myristic acid and a reduction in long chain polyunsaturated fatty acids for both genders. The most profound changes were detected by liquid chromatography mass spectrometry analysis of intact lipids with the tissue from the null mouse having a profound increase in a number of triglycerides. The metabolomic and lipodomic changes indicate PGC-1β has a profound influence on cardiac metabolism.

Human beings inherit an informational culture transmitted through spoken and written language. A growing body of empirical work supports the mutual influence between language and categorization, suggesting that our cognitive-linguistic environment both reflects and shapes our understanding. By implication, artifacts that manifest this cognitive-linguistic environment, such asWikipedia, should represent language structure and conceptual categorization in a way consistent with human behavior. We use this intuition to guide the construction of a computational cognitive model, situated in Wikipedia, that generates semantic association judgments. Our unsupervised model combines information at the language structure and conceptual categorization levels to achieve state of the art correlation with human ratings on semantic association tasks including WordSimilarity-353, semantic feature production norms, word association, and false memory.

    Hosts add a preference pane to your system preferences which lets you toggle your host file entries on and off, as well as add and remove them.

    Hosts.prefpane likes to keep your hosts file clean. When an entry is toggled off, it is not commented out in the hosts file, but instead stored in the app's preferences storage. This means you will have to uncomment all your unused host entries and untoggle them in the prefpane if you want to use them. Comments are treated as comments and will never show up in the list of host entries!

    Ofcourse Hosts.prefpane will detect edits you made to the /etc/hosts directly and merge them into the listing.

    Backups of your hosts file is made in ~/Library/Application Support/Hosts. The backup or your hosts file from before you started using Hosts.prefpane is called hosts.orig and each session a backup is made called hosts.session.

    In spite of their favourable chemical characteristics, using AB5 alloys as fixed bed for hydrogen storage devices requires proper management of a number of technological aspects. Among these, the mechanical stability of metal particle grains under hydrogen cycling and the overall thermal conductivity of the material bed constitute crucial features. We developed by High Energy Ball Milling HEBM a mechanically stable silica-based AB5 composite with enhanced thermal conductivity. Here, focusing on the material's physical-chemical properties, we report on the silica-AB5 composite development and characterization. Particularly, we studied the material consolidation process, the resulting composite morphology and the system behaviour under hydrogen loading/unloading cycling.

    1-Amino-3-nitroguanidine (ANQ, 1) was synthesized by hydrazinolysis of nitroguanidine (NQ) with hydrazine hydrate. Four different amino-nitroguanidinium salts (chloride (2), bromide (3), iodide (4) and sulfate (5)) were synthesized and structurally characterized by low-temperature X-ray diffraction. The halides 2–4 could only be obtained crystalline as monohydrates. In addition, they were characterized by NMR and vibrational spectroscopy, elemental analysis and the sensitivities towards impact, friction and electrostatic discharge were determined. The compounds can be used in silver (AgX, X = Cl, Br, I) and barium (BaSO4) based metathesis reactions in order to form more complex salts of 1-amino-nitroguanidine.

    Cationic lipids have been used in the development of non-viral gene delivery systems as lipoplexes. Stearylamine, a cationic lipid that presents a primary amine group when in solution, is able to compact genetic material by electrostatic interactions. In dispersed systems such as nanoemulsions this lipid anchors on the oil/water interface confering a positive charge to them. The aim of this work was to evaluate factors that influence DNA compaction in cationic nanoemulsions containing stearylamine. The influence of the stearylamine incorporation phase (water or oil), time of complexation, and different incubation temperatures were studied. The complexation rate was assessed by electrophoresis migration on agarose gel 0.7%, and nanoemulsion and lipoplex characterization was done by Dynamic Light Scattering (DLS). The results demonstrate that the best DNA compaction process occurs after 120 min of complexation, at low temperature (4 ± 1 °C), and after incorporation of the cationic lipid into the aqueous phase. Although the zeta potential of lipoplexes was lower than the results found for basic nanoemulsions, the granulometry did not change. Moreover, it was demonstrated that lipoplexes are suitable vehicles for gene delivery.

    The expression levels of genes involved in drug and nutrient absorption were evaluated in the Madin-Darby Canine Kidney (MDCK) in vitro drug absorption model. MDCK cells were grown on plastic surfaces (for 3 days) or on Transwell® membranes (for 3, 5, 7, and 9 days). The expression profile of genes including ABC transporters, SLC transporters, and cytochrome P450 (CYP) enzymes was determined using the Affymetrix® Canine GeneChip®. Expression of genes whose probe sets passed a stringent confirmation process was examined. Expression of a few transporter (MDR1, PEPT1 and PEPT2) genes in MDCK cells was confirmed by RT-PCR. The overall gene expression profile was strongly influenced by the type of support the cells were grown on. After 3 days of growth, expression of 28% of the genes was statistically different (1.5-fold cutoff, p < 0.05) between the cells grown on plastic and Transwell® membranes. When cells were differentiated on Transwell® membranes, large changes in gene expression profile were observed during the early stages, which then stabilized after 5–7 days. Only a small number of genes encoding drug absorption related SLC, ABC, and CYP were detected in MDCK cells, and most of them exhibited low hybridization signals. Results from this study provide valuable reference information on endogenous gene expression in MDCK cells that could assist in design of drug-transporter and/or drug-enzyme interaction studies, and help interpret the contributions of various transporters and metabolic enzymes in studies with MDCK cells.

    Skin cancer is an increasingly common disease, particularly among young adult women. Sunburn early in life is a risk factor for skin cancer. Few studies have reported on psychosocial correlates of sunburn. The current study consisted of an online survey of undergraduate women from a university in the northeastern part of the USA. A logistic regression demonstrated that young women who reported a history of four or more sunburns were significantly more likely to report fair skin, higher perceived susceptibility to skin cancer, greater perceived benefits of tanning (e.g., appearance enhancement), lower perceived control over skin protection, and more frequent sunscreen use. Sunbathing was not associated with a greater number of sunburns. These results suggest that young women who sunburn more often possess other skin cancer risk factors, are aware of their susceptibility to skin cancer, and try to use sunscreen, but feel limited control over their skin protection behavior and are not less likely to sunbathe than others. Therefore, interventions are needed to assist high risk young women in asserting more control over their sun protection behavior and perhaps improve the effectiveness of the sunscreen or other skin protection methods they do employ.

    Astaxanthin captured peroxynitrite to form nitroastaxanthins. 15-Nitroastaxanthin was a major reaction product of astaxanthin with peroxynitrite. Here, the anti-oxidative, anti-tumor-promoting, and anti-carcinogensis activities of 15-nitroastaxanthin were investigated. In addition to astaxanthin, 15-nitroastaxanthin showed excellent singlet oxygen quenching activity. Furthermore, 15-nitroastaxanthin showed inhibitory effects of in vitro Epstein-Barr virus early antigen activation and two-stage carcinogensis on mouse skin papillomas. These activities were slightly higher than those of astaxanthin. Similar results were obtained for the 15-nitrolutein, a major reaction product of lutein with peroxynitrite.

    The grading entropy concept can be adapted to the field of geotechnics, to establish criteria for phenomena such as particle packing, particle migration and filtering, through a quantified expression of the order/disorder in the grain size distribution, in terms of two entropy-based parameters. In this paper, the grading entropy theory is applied in some geotechnical case studies, which serve as benchmark examples to illustrate its application to the characterisation of piping, softening and dispersive soils, and to filtering problems in the context of a leachate collection system for a landfill site. Further, since unstable cohesive (dispersive) soils are generally improved by lime, the effect of lime addition is also considered, on the basis of some measurements and a further application of the grading entropy concept, which allows evolutions in the entropy of a soil to be considered as its grading is modified. The examples described support the hypothesis that the potential for soil erosion and particle migration can be reliably identified using grading entropy parameters derived from grading curve data, and applied through an established soil structure stability criteria and a filtering rule. It is shown that lime modification is not necessarily helpful in stabilizing against particle migration.

    A filtering algorithm is proposed that accurately extracts ground data from airborne light detection and ranging (LiDAR) measurements and generates an estimated digital terrain model (DTM). The proposed algorithm utilizes planar surface features and connectivity with locally lowest points to improve the extraction of ground points (GPs). A slope parameter used in the proposed algorithm is updated after an initial estimation of the DTM, and thus local terrain information can be included. As a result, the proposed algorithm can extract GPs from areas where different degrees of slope variation are interspersed. Specifically, along roads and streets, GPs were extracted from urban areas, from hilly areas such as forests, and from flat area such as riverbanks. Validation using reference data showed that, compared with commercial filtering software, the proposed algorithm extracts GPs with higher accuracy. Therefore, the proposed filtering algorithm effectively generates DTMs, even for dense urban areas, from airborne LiDAR data.

The recent GEO-BON European Biodiversity Observation Network project (EBONE) established a framework for an integrated biodiversity monitoring system. Underlying this framework is the idea of integrating in situ with EO and a habitat classification scheme based on General Habitat Categories (GHC), designed with an Earth Observation-perspective. Here we report on EBONE work that explored the use of NDVI-derived phenology metrics for the identification and mapping of Forest GHCs. Thirty-one phenology metrics were extracted from MODIS NDVI time series for Europe. Classifications to discriminate forest types were performed based on a Random Forests™ classifier in selected regions. Results indicate that date phenology metrics are generally more significant for forest type discrimination. The achieved class accuracies are generally not satisfactory, except for coniferous forests in homogeneous stands (77–82%). The main causes of low classification accuracies were identified as (i) the spatial resolution of the imagery (250 m) which led to mixed phenology signals; (ii) the GHC scheme classification design, which allows for parcels of heterogeneous covers, and (iii) the low number of the training samples available from field surveys. A mapping strategy integrating EO-based phenology with vegetation height information is expected to be more effective than a purely phenology-based approach.
