Posted: 05 Jun 2012 12:00 AM PDT The sequence polymorphism and population structure of Tomato chlorotic dwarf viroid (TCDVd) (isolate Trust) and Potato tuber spindle viroid (PSTVd) (isolate FN) in tomato plants were investigated. Of the 9 and 35 TCDVd clones sequenced from 2 different TCDVd-infected plants, 2 and 4 sequence variants were identified, respectively, leading to a total of 4 sequence variants of 360 nucleotides in length. Variant I was identical to AF162131, the first TCDVd sequence to be reported, and the rest exhibited 1 to 3 nucleotide differences, all in the TR domain, from AF162131/variant I. Of the 33 and 29 PSTVd clones sequenced from 2 different PSTVd-infected plants, 8 and 9 sequence variants were found, respectively, leading to a total of 15 variants ranging in length from 356 to 359 nucleotides. The variant I was identical to EF044303, a PSTVd reported in Russia. The rest exhibited 1 to 11 nucleotide differences scattering in all five domains from EF044303/variant I. The results demonstrated for the first time that TCDVd, like many other viroids including PSTVd, exists in host plants as a collective group comprised of various sequence variants. However, in comparison to PSTVd, TCDVd is less polymorphic in tomato plants as fewer variants and lower haplotype/nucleotide diversities were observed. |
Posted: 05 Jun 2012 12:00 AM PDT This paper describes a simple, sensitive and environmentally benign method for the direct determination of aniline and its derivatives in environmental water samples by capillary zone electrophoresis (CZE) with field-enhanced sample injection. The parameters that influenced the enhancement and separation efficiencies were investigated. Surprisingly, under the optimized conditions, two linear ranges for the calibration plot, 1–50 ng/mL and 50–1000 ng/mL (R > 0.998), were obtained. The detection limit was in the range of 0.29–0.43 ng/mL. To eliminate the effect of the real sample matrix on the stacking efficiency, the standard addition method was applied to the analysis of water samples from local rivers. |
Posted: 05 Jun 2012 12:00 AM PDT In the present study, the 26-residue amphipathic α-helical peptide A12L/A20L (Ac-KWKSFLKTFKSLKKTVLHTLLKAISS-amide) with strong anticancer activity and specificity was used as the framework to study the effects of helicity of α-helical anticancer peptides on biological activities. Helicity was systematically modulated by introducing D-amino acids to replace the original L-amino acids on the non-polar face or the polar face of the helix. Peptide helicity was measured by circular dichroism spectroscopy and was demonstrated to correlate with peptide hydrophobicity and the number of D-amino acid substitutions. Biological studies showed that strong hemolytic activity of peptides generally correlated with high hydrophobicity and helicity. Lower helicity caused the decrease of anti-HeLa activity of peptides. By introducing D-amino acids to replace the original L-amino acids on the non-polar face or the polar face of the helix, we improved the therapeutic index of A12L/A20L against HeLa cells by 9-fold and 22-fold, respectively. These results show that the helicity of anticancer peptides plays a crucial role for biological activities. This specific rational approach of peptide design could be a powerful method to improve the specificity of anticancer peptides as promising therapeutics in clinical practices. |

Yet, as many cities continue to grow and develop in ways deemed typical or standard, these transitions can be difficult to acknowledge. Narratives of continued growth and permanence become accepted and expected while the understanding of urban dynamics becomes lost. In many parts of the world, the shrinking cities phenomenon has given rise to a new awareness of urban transition that provides a laboratory of new conditions at the intersection of urbanism and ecology. With property vacancy rates easily exceeding 50% in certain locations, cities in the American Rust Belt look more like successional woodlands than bustling metropolises, yet these cities still contain significant numbers of urban residents. A central question that arises from this phenomenon is: how can vacant land, through the provision of ecosystem services, become a resource as opposed to a liability? This paper looks to recent studies in urban ecology as a lens for understanding the land use potential of shrinking cities, while discussing unconventional solutions for sustainable development of urban land. |
Metals, Vol. 2, Pages 136-142: Molding of Aluminum Foams by Using Hot Powder Extrusion Posted: 05 Jun 2012 12:00 AM PDT In order to form aluminum foams directly from powder, a combined process of hot powder extrusion and molding is proposed. Aluminum powder mixed with a foaming agent is extruded into the mold through the die heated to a temperature higher than the melting point, and the mold is filled with the aluminum foam. When a stainless steel pipe is used for a simple mold, an aluminum foam bar is obtained of which the relative density varies between 0.2 and 0.3. The molding of aluminum foam by using three types of mold shape shows the influence of gravity and friction. The effect of gravity is significant when a large step exists at the connection between the mold inlet and the die outlet, and friction is dominant in cases where foam is mold in a narrow space. |
