Uno 1.0.9 - Simple, basic text editor.. (Free)

Uno is a simple, yet basic text editor for Mac: it has almost all the base functions you would expect from an editor like TextEdit. Starting from a blank window you can do practically anything with your characters. As it is a pure text editor it doesn't work with images or photos you'd like to put in your text. It does open Rich Text Format files (.rtf) and Unicode-formatted Plain Text files (.txt): it saves both as .rtf and exports as Plain text.

Note for Mac OS X 10.5 Leopard users: although this is a Universal app, it has not been tested carefully on Leopard thus it could run with some issues.



Version 1.0.9 (Jan.9, 2012):
  • Now the user can export either the full text or just a selection of it directly as Plain text file; also just the selection can be saved as RTF file.
  • 'Resume Text' feature has been improved: starting from now, if the last closed window was left empty, it allows the user to retrieve text from the last saved window.

JFB, Vol. 3, Pages 61-78: Monodisperse 130 kDa and 260 kDa Recombinant Human Hemoglobin Polymers as Scaffolds for Protein Engineering of Hemoglobin-Based Oxygen Carriers

A recombinant 130 kDa dihemoglobin which is made up of a single-chain tetra-α globin and four β globins has been expressed as a soluble protein in E. coli. The sequence of the single chain tetra-α is: αI-Gly-αII-(SerGlyGly)5Ser-αIII-Gly-αIV. This dihemoglobin has been purified and characterized in vitro by size exclusion chromatography, electrospray mass spectroscopy, equilibrium oxygen binding, and analytical ultracentrifugation. The observed values of P50 and nmax for the dihemoglobin are slightly lower than those observed for the recombinant hemoglobin rHb1.1 (a "monohemoglobin" comprised of two β globins and an αI-Gly-αII diα-globin chain). Titration of the deoxy form of dihemoglobin with CO shows that all eight heme centers bind ligand. In vivo, dihemoglobin showed increased circulating halflife and a reduced pressor response in conscious rats when compared to rHb1.1. These observations suggest that dihemoglobin is an oxygen carrying molecule with desirable in vivo properties and provides a platform for an isooncotic hemoglobin solution derived solely from a recombinant source. A 260 kDa tetrahemoglobin has also been produced by chemical crosslinking of a dihemoglobin that contains a Lys16Cys mutation in the C-terminal α-globin subunit. Tetrahemoglobin also shows reduced vasoactivity in conscious rats that is comparable to that observed for dihemoglobin.

Google's Kenyan ripoff

Mocality is an African startup that has a Kenya-wide business directory. There is no Kenyan yellow pages, so the directory was crowdsourced, paying thousands of Kenyans to help create and validate its database.

When the businesses in Mocality's database started asking them about the premium service they were offering with Google, Mocality was puzzled. They had no joint venture with Google, and they had never charged any business for inclusion in their database. When they examined their server logs, they saw a large number of hits to the records for the businesses that had been cold-called from the same IP range.

So Mocality laid a trap: when that IP range next visited the Mocality site, they fed it fake phone numbers that went to Mocality's own call center, where a Mocality operator pretended to be a business-owner and recorded the conversation. In that conversation, the caller identified himself as a Google employee, calling about a joint Google-Mocality venture, and asking the business to pay Google for a Kenya Business Online website with its own domain on that basis. This was, of course, absolutely fraudulent. There was and is no Google-Mocality joint venture.

Shortly after, that IP range stopped visiting Mocality's servers, but another range, this one registered to Google's Mountain View headquarters, began to query its database. Again, Mocality served a fake result with its own call-center number, and an hour later, they received a call from someone identifying herself as working on Google's behalf, asking for money for a joint Google-Mocality product.

The conclusion is hard to escape: Google -- or people working on its behalf, with its knowledge and cooperation -- took the numbers of tens of thousands of Kenyan businesses from Mocality's database, then fraudulently solicited money from them by claiming to be in a joint venture with Mocality. This seems to me to be outright criminal activity, and Google has a lot of explaining to do.

Since October, Google's GKBO appears to have been systematically accessing Mocality's database and attempting to sell their competing product to our business owners. They have been telling untruths about their relationship with us, and about our business practices, in order to do so. As of January 11th, nearly 30% of our database has apparently been contacted.

Furthermore, they now seem to have outsourced this operation from Kenya to India.

When we started this investigation, I thought that we'd catch a rogue call-centre employee, point out to Google that they were violating our Terms and conditions (sections 9.12 and 9.17, amongst others), someone would get a slap on the wrist, and life would continue.

I did not expect to find a human-powered, systematic, months-long, fraudulent (falsely claiming to be collaborating with us, and worse) attempt to undermine our business, being perpetrated from call centres on 2 continents.

Direct catalytic valorization of bioethanol to 1-butanol over different alumina supported catalysts was studied. Thirteen (13) heterogeneous catalysts were screened in search for the optimal material composition for direct one-pot conversion of ethanol to 1-butanol. For the most promising catalyst, a 25% ethanol conversion with 80% selectivity (among liquid carbon products) to 1-butanol could be reached at 250 °C. Additionally, the reaction kinetics and mechanisms were further investigated upon use of the most suitable catalyst candidate.

Physical properties of commercial carbamazepine (CBZ) samples can significantly influence drug release and thereby jeopardize bioequivalence of the final dosage form. The aim of this study was to reduce variability in commercial CBZ samples by recrystallization. CBZ samples of four different suppliers were recrystallized in ethanol solution containing 1% polyvinylpyrrolidone (PVP). CBZ samples were analyzed by disk intrinsic dissolution rate (DIDR), X-ray powder diffraction (XRPD), differential scanning calorimetry (DSC), and scanning electron microscopy (SEM). Recrystallized CBZ samples showed strongly reduced variability in DIDR compared to the untreated CBZ samples. Moreover, transformation process to CBZ dihydrate was inhibited; no dihydrate crystals were visible on compact surfaces after 8 h intrinsic dissolution measurement. Recrystallized CBZ samples showed no change in polymorphic form, however, particle size and shape was inhomogenous. In binary mixtures with microcrystalline cellulose, recrystallized CBZ samples again showed difference in drug release. This difference was associated with the inhomogenous particle size in the recrystallized CBZ samples. The results show that a controlled grinding step is required after recrystallization. We suggest the recrystallization in presence of 1% PVP followed by a controlled grinding step as a strategy to reduce dissolution variability in commercial CBZ samples.

Polyethylene (PE) is one of the most widely used polymers in many industrial applications. Biomedical uses seem to be attractive, with increasing interest. However, PE it prone to infections and its additional surface treatment is indispensable. An increase in resistance to infections can be achieved by treating PE surfaces with substances containing antibacterial groups such as triclosan (5-Chloro-2-(2,4-dichlorophenoxy)phenol) and chlorhexidine (1,1'-Hexamethylenebis[5-(4-chlorophenyl)biguanide]). This work has examined the impact of selected antibacterial substances immobilized on low-density polyethylene (LDPE) via polyacrylic acid (PAA) grafted on LDPE by low-temperature barrier discharge plasma. This LDPE surface treatment led to inhibition of Escherichia coli and Staphylococcus aureus adhesion; the first causes intestinal disease, peritonitis, mastitis, pneumonia, septicemia, the latter is the reason for wound and urinary tract infections.

Then they are scrambled by rotating the rows and columns without the player's knowledge. The objective of the torus puzzle is to rearrange the pieces in ascending order by rotating the rows and columns. We provide a solution to this puzzle. In addition, we provide lower and upper bounds on the number of steps for solving the puzzle. Moreover, we consider a variant of the torus puzzle in which each piece is colored either black or white, and we present a hardness result for solving it.

Electrochemical aptasensors, which are based on the specificity of aptamer-target recognition, with electrochemical transduction for analytical purposes have received particular attention due to their high sensitivity and selectivity, simple instrumentation, as well as low production cost. Aptamers are functional nucleic acids with specific and high affinity to their targets, similar to antibodies. However, they are completely selected in vitro in contrast to antibodies. Due to their stability, easy chemical modifications and proneness to nanostructured device construction, aptamer-based sensors have been incorporated in a variety of applications including electrochemical sensing devices. In recent years, the performance of aptasensors has been augmented by incorporating novel nanomaterials in the preparation of better electrochemical sensors. In this review, we summarize the recent trends in the use of nanomaterials for developing electrochemical aptasensors.

Sampling of small molecules from both porous and non-porous surfaces poses a significant challenge across biological agents. Particle sizes of toxins are smaller than living organisms and can be extremely toxic at low level concentrations. A small number of studies evaluating sampling efficiencies of commercial off the shelf (COTS) materials have been performed with toxins and proteins. However, they have been limited to non-ricin stimulants with drastically different physical properties than their native counterparts. We have identified a commercially available non-toxic recombinant ricin, complete with both A and B subunits present, which can be recognized by antibodies commonly used to assay native ricin. In evaluating recovery efficiency, we deposited the recombinant ricin by both liquid deposition, and as a dry aerosol. Our studies demonstrated a significant difference in recovery efficiencies from liquid deposited ricin, ranging between 30% and 70%, than from an aerosol generated deposition ranging from below detectable levels to 22%, depending on the contaminated surface and swab material being used. This study demonstrates the necessity for accurate dissemination techniques of sampling technologies for the consideration of use in an environment where suspected toxin contamination is being evaluated.

Okara is a byproduct generated during tofu or soymilk production processes. Crude polysaccharide (yield 56.8%) was isolated by removing fat, protein and low molecular weight carbohydrates from initial okara. Crude okara polysaccharide was further divided into four soluble fractions and an insoluble residue fraction by extracting with 0.05 M EDTA + NH4 oxalate, 0.05 M NaOH, 1 M NaOH and 4 M NaOH, with yields of 7.7%, 3.6%, 20.7%, 16.0% and 27.9%, respectively. Arabinose, galactose, galacturonic acid, xylose and glucose (only for the insoluble fraction) were the major constituent sugars. The primary sugar residues of okara polysaccharides were 1,4-linked β-galactopyranose, 1,5- and 1,3-linked α-arabinofuranose, 1,5-linked α-xylofuranose, 1,2-linked, 1,2,4-linked and terminal α-rhamnopyranose (or fucopyranose), and 1,4-linked β-glucopyranose (only for the insoluble fraction), indicating okara polysaccharides might contain galactan, arabinan, arabinogalactan, xylogalacturonan, rhamnogalacturonan, xylan, xyloglucan and cellulose.

Ganoderma lucidum and Ganoderma sinense, known as Lingzhi in Chinese, are commonly used Chinese medicines with excellent beneficial health effects. Triterpenes and polysaccharides are usually considered as their main active components. However, the content of triterpenes differs significantly between the two species of Ganoderma. To date, a careful comparison of polysaccharides from the two species of Ganoderma has not been performed. In this study, polysaccharides from fruiting bodies of two species of Lingzhi collected from different regions of China were analyzed and compared based on HPSEC-ELSD and HPSEC-MALLS-RI analyses, as well as enzymatic digestion and HPTLC of acid hydrolysates. The results indicated that both the HPSEC-ELSD profiles and the molecular weights of the polysaccharides were similar. Enzymatic digestion showed that polyshaccharides from all samples of Lingzhi could be hydrolyzed by pectinase and dextranase. HPTLC profiles of their TFA hydrolysates colored with different reagents and their monosaccharides composition were also similar.

Arenaviruses comprise a diverse family of enveloped negative-strand RNA viruses that are endemic to specific rodent hosts worldwide. Several arenaviruses cause severe hemorrhagic fevers in humans, including Junín and Machupo viruses in South America and Lassa fever virus in western Africa. Arenavirus entry into the host cell is mediated by the envelope glycoprotein complex, GPC. The virion is endocytosed on binding to a cell-surface receptor, and membrane fusion is initiated in response to physiological acidification of the endosome. As with other class I virus fusion proteins, GPC-mediated membrane fusion is promoted through a regulated sequence of conformational changes leading to formation of the classical postfusion trimer-of-hairpins structure. GPC is, however, unique among the class I fusion proteins in that the mature complex retains a stable signal peptide (SSP) as a third subunit, in addition to the canonical receptor-binding and fusion proteins. We will review the curious properties of the tripartite GPC complex and describe evidence that SSP interacts with the fusion subunit to modulate pH-induced activation of membrane fusion. This unusual solution to maintaining the metastable prefusion state of GPC on the virion and activating the class I fusion cascade at acidic pH provides novel targets for antiviral intervention.

We have demonstrated the efficacy of a microfluidic medium exchange method for single cells using passive centrifugal force of a rotating microfluidic-chip based platform. At the boundary of two laminar flows at the gathering area of two microfluidic pathways in a Y-shape, the cells were successfully transported from one laminar flow to the other, without mixing the two microfluidic mediums of the two laminar flows during cell transportation, within 5 s with 1 g (150 rpm) to 36.3 g (900 rpm) acceleration, with 93.5% efficiency. The results indicate that this is one of the most simple and precise tools for exchanging medium in the shortest amount of time.
