Lethal toxin (LT) of Bacillus anthracis reduces the production of a number of inflammatory mediators, including transcription factors, chemokines and cytokines in various human cell lines, leading to down-regulation of the host inflammatory response. Previously we showed that the reduction of interleukin-8 (IL-8) is a sensitive marker of LT-mediated intoxication in human neutrophil-like NB-4 cells and that IL-8 levels are restored to normality when therapeutic monoclonal antibodies (mAb) with toxin-neutralising (TN) activity are added. We used this information to develop cell-based assays that examine the effects of TN therapeutic mAbs designed to treat LT intoxication and here we extend these findings. We present an in vitro assay based on human endothelial cell line HUVEC jr2, which measures the TN activity of therapeutic anti-LT mAbs using IL-8 as a marker for intoxication. HUVEC jr2 cells have the advantage over NB-4 cells that they are adherent, do not require a differentiation step and can be used in a microtitre plate format and therefore can facilitate high throughput analysis. This human cell-based assay provides a valid alternative to the mouse macrophage assay as it is a more biologically relevant model of the effects of toxin-neutralising antibodies in human infection.

With increasing economic and environmental costs associated with fertilizer use, the need for low-input agroecological systems is on the rise. It is well documented that intercropping legumes can increase the supply of nutrients, through N2-fixation and P mobilization. Concurrently, the integration of trees in the agricultural landscape shows increasing evidence of nutrient inputs through efficient biogeochemical cycles. However, little is known about the effects shade has on legume-crop performance. This research aims to determine whether intercropping of the legumes soybean (Glycine max L. Merr.) and alfalfa (Medicago sativa) with wheat (Triticum turgidum durum) is beneficial for performance, particularly under shady conditions associated with tree-based intercropping. Test species were cultivated in hydroponics with a broad nutrient solution and steady state addition of N for 3 weeks. Individual plants were transferred to rhizoboxes with a 2 mm zone of soil for 16 days under (i) full sun or (ii) shade to mimic light levels at the tree-crop interface. Under monocropping, shading was found to significantly decrease wheat biomass. Intercropping wheat with alfalfa under full sun had no negative effect on growth but did increase wheat P uptake as compared to monocropped wheat. In contrast, intercropping wheat with soybean under full sun decreased wheat biomass, suggesting competition. However, under shade, this competitive effect was mitigated, as wheat exhibited similar biomass and higher N and P shoot concentration when associated with soybean as compared to monocropped wheat under lower light levels. This effect may be attributed to reduced biomass of soybean combined with higher soybean N2-fixation under shade. Legume-based intercrops may increase nutrient supply and growth but these beneficial effects will be dependent on matching species selection to light levels under tree-based intercropping.

The increased activity in the field of Battery Electric Vehicles (BEVs) and Hybrid Electric Vehicles (HEVs) have led to an increase in standardization work, performed by both world-wide organizations like the IEC or the ISO, as by regional and national bodies such as CEN, CENELEC, SAE or JEVA. The issues of these standards cover several topics: safety, performance and operational/dimension issues. This paper reports a brief overview of current standardization activities of lithium batteries based on IEC 62660-1/2 and ISO 12405-1/2. Furthermore, in this paper, a series of innovative test procedures for lithium-ion batteries are presented. Thanks to these tests, the general characteristics of a battery such as charge and discharge capabilities, power performances and life cycle can be determined. Then, a new approach for extracting the life cycle of a battery in function of depth of discharge has been developed.

Bone-seeking (osteotropic) drug delivery systems (ODDS) represent an interesting solution for targeting different types of drugs to the bones. In particular, anticancer and antibacterial agents could take advantage of such therapeutic strategy. We have recently developed an innovative approach to this aim: a new osteotropic biomaterial was prepared, based on the conjugation of a poly(lactide-co-glycolide) (PLGA) with the bisphosphonate drug alendronate (PLGA-ALE); its hemo- and cytocompatibility were verified. Starting with this copolymer, an osteotropic nanoparticle system (NP) was produced for the targeted delivery of antineoplastic drugs to osteolytic bone metastases; in particular, doxorubicin was tested as a model drug. The in vitro and in vivo results of the new ODDS are validated in this article. All the experimental data confirmed that the drug retained its activity after loading in the PLGA-ALE NP; they can be thus considered a new promising strategy for active targeting of drugs to bone tissues in different pathological situations.

A new type of brushless double rotor machine (BDRM) is proposed in this paper. The BDRM is an important component in compound-structure permanent-magnet synchronous machine (CS-PMSM) systems, which are promising for power-split hybrid electric vehicle (HEV) applications. The BDRM can realize the speed adjustment between claw-pole rotor and permanent-magnet rotor without brushes and slip rings. The structural characteristics of the BDRM are described and its magnetic circuit model is built. Reactance parameters of the BDRM are deduced by an analytical method. It is found that the size characteristics of the BDRM are different from those of conventional machines. The new sizing and torque equations are analyzed and the theoretical results are used in the optimization process. Studies of the analytical magnetic circuit and finite element method (FEM) model show that the BDRM tends to have high leakage flux and low power factor, and then the method to obtain high power factor is discussed. Furthermore, a practical methodology of the BDRM design is developed, which includes an analytical tool, 2D field calculation and performance evaluation by 3D field calculation. Finally, different topologies of the BDRM are compared and an optimum prototype is designed.

Due to the very complex sets of component systems, interrelated thermodynamic processes and seasonal change in operating conditions, it is relatively difficult to find an accurate model for turbine cycle of nuclear power plants (NPPs). This paper deals with the modeling of turbine cycles to predict turbine-generator output using an adaptive neuro-fuzzy inference system (ANFIS) for Unit 1 of the Kuosheng NPP in Taiwan. Plant operation data obtained from Kuosheng NPP between 2006 and 2011 were verified using a linear regression model with a 95% confidence interval. The key parameters of turbine cycle, including turbine throttle pressure, condenser backpressure, feedwater flow rate and final feedwater temperature are selected as inputs for the ANFIS based turbine cycle model. In addition, a thermodynamic turbine cycle model was developed using the commercial software PEPSE® to compare the performance of the ANFIS based turbine cycle model. The results show that the proposed ANFIS based turbine cycle model is capable of accurately estimating turbine-generator output and providing more reliable results than the PEPSE® based turbine cycle models. Moreover, test results show that the ANFIS performed better than the artificial neural network (ANN), which has also being tried to model the turbine cycle. The effectiveness of the proposed neuro-fuzzy based turbine cycle model was demonstrated using the actual operating data of Kuosheng NPP. Furthermore, the results also provide an alternative approach to evaluate the thermal performance of nuclear power plants.

As Xeni wrote on Tuesday, the MPAA isn't pleased about sites like this one going dark to protest SOPA and PIPA. Former Senator Chris Dodd, Chairman and CEO of the Motion Picture Association of America called it "an abuse of power given the freedoms these companies enjoy in the marketplace today."

Well, he should know.

After all, he is the CEO of the organization responsible for inserting those unskippable FBI warnings (which are highly prejudiced and factually incorrect, advising, for example, that DVDs can't be rented, even though the law says they can) before every commercial DVD. He's the CEO of the organization that inserts those insulting PSAs in front of every movie chiding those of us who buy our DVDs because someone else decided to download the same movie for free.

And he's the CEO of the organization responsible for the section of the DMCA that makes it illegal to build a DVD player that can skip these mandatory, partisan, commercially advantageous messages.

So he knows a thing or two about "abuse of power given the freedoms these companies enjoy in the marketplace today."

Cancers, Vol. 4, Pages 31-38: Surgical Treatment of Intra-Abdominal Desmoid Tumors Resulting In Short Bowel Syndrome

Advanced intra-abdominal desmoids tumors present with severe symptoms, complications or rapid growth, which lead to adverse outcomes. Our aim was to evaluate the treatment and outcome of patients with advanced intra-abdominal desmoids tumors, and develop guidelines for surgical management of these patients. We reviewed the clinical courses of 21 adult patients with advanced stage intra-abdominal desmoid tumors who presented to an intestinal rehabilitation and transplantation program. Patients with massive intestinal resection presented in two groups. The first group had a short small intestinal remnant after resection ( < 60 cm). These patients were poor rehabilitation candidates and eventually met criteria for transplant. The second had longer intestinal remnants and were more successfully rehabilitated and have not had complications that would lead to transplantation. Advanced intra-abdominal desmoid tumors have outcomes after resection that merit aggressive resection and planned intestinal rehabilitation and intestinal transplantation as indicated.

New derivatives of 7-aminocephalosporanic acid 1–8 were synthesized by acylation of the 7-amino group of the cephem nucleus with various arylidinimino-1,3,4-thiadiazole-thio(or dithio)-acetic acid intermediates 3a–d and 5a–d, respectively, so the acyl side chains of these new cephalosporins contained a sulfide or disulfide bond. This unique combination of a Schiff base with the sulfide or disulfide bonds in the acyl side chain afforded new cephalosporins of reasonable potencies, some of which were found to possess moderate activities against the tested microorganisms. Their chemical structures were characterized by ¹H-NMR, IR spectroscopy and elemental microanalysis. Preliminary in vitro antimicrobial activities of the prepared cephalosporins were investigated using a panel of selected microorganisms. Results indicated that the newly synthesized cephalosporins containing disulfide bonds (compounds 5–8) exhibited better activities against Staphylococcus aureus and Escherichia coli. The cephalosporins cross-linked by a sulfide bond (compounds 1–4) showed a slight change in antimicrobial activities when compared with that of the reference cephalosporin (cephalexin).

Twenty one phenylpropanoids (including eugenol and safrole) and synthetic analogues, thirteen of them new compounds, were evaluated for antifungal properties, first with non-targeted assays against a panel of human opportunistic pathogenic fungi. Some structure-activity relationships could be observed, mainly related to the influence of an allyl substituent at C-4, an OH group at C-1 and an OCH3 at C-2 or the presence of one or two NO2 groups in different positions of the benzene ring. All active compounds were tested in a second panel of clinical isolates of C. albicans and non-albicans Candida spp., Cryptococcus neoformans and dermatophytes. The eugenol derivative 4-allyl-2-methoxy-5-nitrophenol (2) was the most active structure against all strains tested, and therefore it was submitted to targeted assays. These studies showed that the antifungal activity of 2 was not reversed in the presence of an osmotic support such as sorbitol, suggesting that it does not act by inhibiting the fungal cell wall synthesis or assembly. On the other hand, the Ergosterol Assay showed that 2 did not bind to the main sterol of the fungal membrane up to 250 µg mL−1. In contrast, a 22% of fungal membrane damage was observed at concentrations = 1 × MIC and 71% at 4× MIC, when 2 was tested in the Cellular Leakage assay. The comparison of log P and MICs for all compounds revealed that the antifungal activity of the eugenol analogues would not to be related to lipophilicity.

Marine Drugs, Vol. 10, Pages 234-241: Antibacterial Bisabolane-Type Sesquiterpenoids from the Sponge-Derived Fungus Aspergillus sp.

Four new bisabolane-type sesquiterpenoids, aspergiterpenoid A (1), (−)-sydonol (2), (−)-sydonic acid (3), and (−)-5-(hydroxymethyl)-2-(2′,6′,6′-trimethyltetrahydro-2H- pyran-2-yl)phenol (4) together with one known fungal metabolite (5) were isolated from the fermentation broth of a marine-derived fungus Aspergillus sp., which was isolated from the sponge Xestospongia testudinaria collected from the South China Sea. Four of them (1–4) are optically active compounds. Their structures and absolute configurations were elucidated by using NMR spectroscopic techniques and mass spectrometric analysis, and by comparing their optical rotations with those related known analogues. Compounds 1–5 showed selective antibacterial activity against eight bacterial strains with the MIC (minimum inhibiting concentrations) values between 1.25 and 20.0 µM. The cytotoxic, antifouling, and acetylcholinesterase inhibitory activities of these compounds were also examined.

The brominated pyrrole-imidazole Ageladine A was used for live imaging of the jellyfish (jellies) Nausithoe werneri, the sea anemone Metridium senile and the flatworm Macrostomum lignano. The fluorescence properties of Ageladine A allow for estimation of pH values in tissue and organs in living animals. The results showed that Nausithoe werneri had the most acidic areas in the tentacles and close to the mouth (pH 4–6.5), Metridium senile harbours aggregates of high acidity in the tentacles (pH 5) and in Macrostomum lignano, the rhabdoids, the gonads and areas close to the mouth were the most acidic with values down to pH 5.

In this study, we investigated the effects of temozolomide (TMZ)/Poly (lactide-co-glycolide)(PLGA)/nano-hydroxyapatite microspheres on the behavior of U87 glioma cells. The microspheres were fabricated by the "Solid/Water/Oil" method, and they were characterized by using X-Ray diffraction, scanning electron microscopy and differential scanning calorimetry. The proliferation, apoptosis and invasion of glioma cells were evaluated by MTT, flow cytometry assay and Transwell assay. The presence of the key invasive gene, αVβ3 integrin, was detected by the RT-PCR and Western blot method. It was found that the temozolomide/PLGA/nano-hydroxyapatite microspheres have a significantly diminished initial burst of drug release, compared to the TMZ laden PLGA microspheres. Our results suggest they can significantly inhibit the proliferation and invasion of glioma cells, and induce their apoptosis. Additionally, αVβ3 integrin was also reduced by the microspheres. These data suggest that by inhibiting the biological behavior of glioma cells in vitro, the newly designed temozolomide/PLGA/nano-hydroxyapatite microspheres, as controlled drug release carriers, have promising potential in treating glioma.

Collective motion of cell sheets plays a role not only in development and repair, but also in devastating diseases such as cancer. However, unlike single-cell motility, collective motion of cell sheets involves complex cell-cell communication during migration; therefore, its mechanism is largely unknown. To elucidate propagation of signaling transduced by cell-cell interaction, we designed a hydrogel substrate that can cause local mechanical stretching of cell sheets. Poly (N-isopropyl acrylamide) (PNIPAAm) hydrogel is a temperature-responsive polymer gel whose volume changes isotropically in response to temperature changes below 37 °C. We designed a combined hydrogel substrate consisting of collagen-immobilized PNIPAAm as the local stimulation side and polyacrylamide (PAAm) as the non-stimulation side to assess propagation of mechanical transduction. Mardin-Darby canine kidney (MDCK) cells adhered to the collagen-immobilized PNIPAAm gel increased it area and were flattened as the gel swelled with temperature decrease. E-cadherin in these cells became undetectable in some domains, and actin stress fibers were more clearly observed at the cell base. In contrast, E-cadherin in cells adhered to the collagen-immobilized PAAm side was equally stained as that in cells adhered to the collagen-immobilized PAAm side even after temperature decrease. ERK1/2 MAPK activation of cells on the non-stimulated substrate occurred after partial stretching of the cell sheet suggesting the propagation of signaling. These results indicate that a change in the balance of mechanical tension induced by partial stretching of cell sheets leads to activation and propagation of the cell signaling.

This paper investigates the water molecule from a variety of viewpoints. Water can involve different isotopes of Hydrogen and Oxygen, it can form differently shaped isomer molecules, and, when frozen, it occupies space differently than most other substances do. The tool for conducting the investigation of all this is called 'Algebraic Chemistry'. This tool is a quantitative model for predicting the energy budget for all sorts of changes between different ionization states of atoms that are involved in chemical reactions and in changes of physical state. The model is based on consistent patterns seen in empirical data about ionization potentials, together with rational scaling laws that can interpolate and extrapolate for situations where no data are available. The results of the investigation of the water molecule include comments, both positive and negative, about technologies involving heavy water, poly water, Brown's gas, and cold fusion.
