LINE-1 (Long Interspersed Nuclear elements) and HERVs (Human Endogenous Retroviruses) are two families of retrotransposons which together account for about 28% of the human genome. Genes harbored within LINE-1 and HERV retrotransposons, particularly that encoding the reverse transcriptase (RT) enzyme, are generally expressed at low levels in differentiated cells, but their expression is up-regulated in embryonic tissues and transformed cells. Here we review evidence indicating that the LINE-1-encoded RT plays regulatory roles in early embryonic development. Indeed, antisense-mediated inhibition of expression of a highly expressed LINE-1 family in mouse zygotes caused developmental arrest at the two- or four-cell embryo stages. Development is also arrested when the embryo endogenous RT activity is pharmacologically inhibited by nevirapine, an RT inhibitor currently employed in AIDS treatment. The arrest of embryonic development is irreversible even after RT inhibition is removed and it is associated with subverted gene expression profiles. These data indicate an early requirement for LINE-1-encoded RT to support early developmental progression. Consistent with this, recent findings indicate that a reverse transcription wave is triggered in the zygote a few hours after fertilization and is propagated at least through the first two rounds of cell division. On the whole these findings suggest that reverse transcription is strictly required in early embryos as a key component of a novel RT-dependent mechanism that regulated the proper unfolding of the developmental program.

France suspends three-strikes copyright law after massive data-leak

TMG, a private contractor that administers France's HADOPI copyright system, has been hacked, resulting in a temporary suspension of HADOPI. Under HADOPI, people who use an Internet connection where one or more users have been accused of multiple acts of copyright infringement lose their Internet access for a year. TMG was in charge of storing the entertainment industry's enemies list of networks that had been used by accused infringers, and their security was basically nonexistent. The hack resulted in a dump of administrative material and IP addresses, and the head of the HADOPI agency announced that they would not gather IP addresses while they got their house in order. The UK has a plan to gather the IP addresses of networks used by accused infringers as well -- will they pick a better contractor to administer it than France did?

The problems appear to be real. Eric Walter, the head of France's HADOPI antipiracy agency that administers the "three strikes" regime, took to Twitter to tell the world that "par mesure de précaution l' #hadopi a décidé de suspendre provisoirement son interconnexion avec #TMG" [as a precautionary measure, #hadopi decided to temporarily suspend its interconnection with #TMG].
This temporary suspension of the interconnect agreement means that TMG -- the only private firm cleared to collect the IP addresses needed for HADOPI to function -- can't provide new addresses for the moment.
French tech sites like Numerama have run with the story, posting lists of questions that "need to be answered" by HADOPI and by French data-security authority CNIL.

(2'E)-1-(3-methyl-4-p-toluenesulfonyl-2-butene)-6-methyl-2,3,4,5-tetramethoxybenzene (4) is the key intermediate in the synthesis of coenzyme Q10 via a coupling reaction with solanesyl bromide. In this paper, we report a simple and effective synthesis of compound 4, starting with the readily available and inexpensive precursors p-toluenesulfonyl chloride (TsCl) and isoprene to obtain (2E)-1-p-toluenesulfonyl-2-methyl-4-hydroxy-2-butene (3) by addition, esterification and hydrolysis. Application of the Friedel-Crafts alkylation to compound 3, followed by the addition of 2,3,4,5-tetramethoxytoluene (TeMT), assembled the two parts into compound 4. The key parameters of each reaction were optimized at the same time, and the four total operations needed to produced compound 4 had a 27.9% overall yield under the optimized conditions. The structures of the compounds were characterized by 1H-NMR, IR and MS. This alternative process has the potential to be used for large-scale process.
