IJERPH, Vol. 8, Pages 2675-2691: Polycyclic Aromatic Hydrocarbon (PAH) Exposure and DNA Adduct Semi-Quantitation in Archived Human Tissues

Polycyclic aromatic hydrocarbons (PAHs) are combustion products of organic materials, mixtures of which contain multiple known and probable human carcinogens. PAHs occur in indoor and outdoor air, as well as in char-broiled meats and fish. Human exposure to PAHs occurs by inhalation, ingestion and topical absorption, and subsequently formed metabolites are either rendered hydrophilic and excreted, or bioactivated and bound to cellular macromolecules. The formation of PAH-DNA adducts (DNA binding products), considered a necessary step in PAH-initiated carcinogenesis, has been widely studied in experimental models and has been documented in human tissues. This review describes immunohistochemistry (IHC) studies, which reveal localization of PAH-DNA adducts in human tissues, and semi-quantify PAH-DNA adduct levels using the Automated Cellular Imaging System (ACIS). These studies have shown that PAH-DNA adducts concentrate in: basal and supra-basal epithelium of the esophagus, cervix and vulva; glandular epithelium of the prostate; and cytotrophoblast cells and syncitiotrophoblast knots of the placenta. The IHC photomicrographs reveal the ubiquitous nature of PAH-DNA adduct formation in human tissues as well as PAH-DNA adduct accumulation in specific, vulnerable, cell types. This semi-quantative method for PAH-DNA adduct measurement could potentially see widespread use in molecular epidemiology studies.

IJERPH, Vol. 8, Pages 2656-2674: Anabolic Androgenic Steroids—Use and Correlates among Gym Users—An Assessment Study Using Questionnaires and Observations at Gyms in the Stockholm Region

The purpose of this study was to estimate the prevalence of anabolic androgenic steroid (AAS) use and offers to use among gym users in Stockholm County (Sweden), and to conduct a comparison of concordance in estimates of AAS and supplements at gyms between two data collection methods. A questionnaire was distributed to members at 36 training facilities and 1,752 gym users participated in the study. An observation study was conducted as covert participant observations at 64 gyms. According to the questionnaire, 3.9% of men reported life time use of AAS, 1.4% use during the past 12 months and 0.4% AAS use during past 30 days. Not only were there similar patterns found in the two methods, i.e., similar age and gender distributions for AAS use, but analyses of concordance showed that gyms with a higher prevalence of self-reported AAS-use and supplement use (questionnaire) showed a significantly higher proportion of observer-assessed AAS users. Analyses of individual predictors showed that AAS users were almost always young men, regular weight trainers and more often users of drugs and nutritional supplements. The higher prevalence of AAS use among gym users than in the general population makes the former an appropriate target group for AAS prevention. The connection between supplements, drugs and AAS use suggests that effective AAS prevention need to focus on several risk factors for AAS use. The clear resemblance in estimates between the observation and questionnaire data strengthen the credibility of the two methods.

Remote Sensing, Vol. 3, Pages 1308-1322: Towards Detecting Swath Events in TerraSAR-X Time Series to Establish NATURA 2000 Grassland Habitat Swath Management as Monitoring Parameter

Spatial monitoring tools are necessary to respond to the threat of global biodiversity loss. At the European scale, remote sensing tools for NATURA 2000 habitat monitoring have been requested by the European Commission to fulfill the obligations of the EU Habitats Directive. This paper introduces a method by which swath events in semi-natural grasslands can be detected from multi-temporal TerraSAR-X data. The investigated study sites represent rare and endangered habitats (NATURA 2000 codes 6410, 6510), located in the Döberitzer Heide nature conservation area west of Berlin. We analyzed a time series of 11 stripmap images (HH-polarization) covering the vegetation period affected by swath (June to September 2010) at a constant 11-day acquisition rate. A swath detection rule was established to extract the swath events for the NATURA 2000 habitats as well as for six contrasting pasture sites not affected by swath. All swath events observed in the field were correctly allocated. The results indicate the potential to allocate semi-natural grassland swath events to 11-day-periods using TerraSAR-X time series. Since the conservation of semi-natural grassland habitats requires compliance with specific swath management rules, the detection of swath events may thus provide new parameters for the monitoring of NATURA 2000 grassland habitats.

Polymers, Vol. 3, Pages 998-1005: The Effects of Exposure Time on the Surface Microhardness of Three Dual-Cured Dental Resin Cements

This study evaluated the exposure time of light-curing of the polymers used for cementation on microhardness test in different storage times. The polymers (specifically called resin cements) were RelyX ARC, RelyX U100, and SET. Five specimens of each group were prepared and photo-polymerized with exposure times of 20 s and 180 s, using a LED polymerization unit with wavelength of 440 ~ 480 nm and light output was consistently 1,500 mW/cm2. The Vickers hardness test was performed in a MMT-3 Microhardness Tester. Data were submitted to ANOVA and Tukey's test (α = 0.05). The values of RelyX ARC showed statistically significant difference to groups with light exposure when considering only chemical cure (p < 0.05). The groups with light exposure (20 s and 180 s) showed no significant difference between them (p > 0.05). The RelyX U100 cured only chemically showed statistically significant difference between 48 h and 7 days (p < 0.05). The SET resin cement showed no significant difference to groups without light exposure for all storage times (p > 0.05). The values of hardening of the dual-cured resin cements improved after setting by light and chemical activation demonstrating the importance of light curing.
