In current therapeutic strategies, bone defects are filled up by bone auto- or allografts. Since they are limited by insufficient availability and donor site morbidity, it is necessary to find an appropriate alternative of synthetic porous bone materials. Because of their osteoconductive characteristics, ceramic materials like tricalciumphosphate (TCP) are suitable to fill up bone defects. Another advantage of TCP implants is the ability of patient-specific engineering. Objective of the present in-vitro study was to analyze the migration capacity and viability of human primary osteoblasts in porous three-dimensional TCP scaffolds in a static cell culture. To obtain data of the cellular supply with nutrients and oxygen, we determined the oxygen concentration and the pH value within the 3D scaffold compared to the surrounding medium using microsensors. After eight days of cultivation we found cells on all four planes. During incubation, the oxygen concentration within the scaffold decreased by approximately 8%. Furthermore, we could not demonstrate an increasing acidification in the core of the TCP scaffold. Our results suggest that osteoblasts could migrate and survive within the macroporous TCP scaffolds. The selected size of the macropores prevents overgrowth of cells, whereby the oxygen and nutrients supply is sufficiently guaranteed.

The lack of authentic standards limits the quantitative analysis of herbal drugs in biological samples. This present work demonstrated a practicable assay of herbs and their metabolites independent of the availability of authentic standards. A liquid chromatography–electrospray ionization–mass spectrometry (LC–ESI–MS) method for the qualitative and quantitative determination of the metabolites after oral administration of Evodiae fructus and Zuojinwan preparation in rat urine has been developed. Urine samples extracted with a protein precipitation procedure were separated on a C18 column using a mixture of water (containing 0.1% formic acid) and acetonitrile (30:70, v/v) as mobile phase. The detection was performed by MS with electrospray ionization interface in positive selected reaction monitoring (SRM) mode. One urine sample after administration was selected as 'standard'. The method validation was carried out according to a conventional method that was calibrated by authentic standards. The fully validated method was applied to the pharmacokinetic study of the metabolites in rat urine. The results could provide evidence to explain the combination of Coptidis rhizoma and Evodiae fructus in terms of elimination.

Formula One in real life is finally exciting again. Sure, Vettel is winning everything in sight, but the new rules and overtaking systems have, if nothing else, at least made the races exciting to watch. That's worked out very nicely for Codemasters, who rode the crest of the wave last year with F1 2010 and are now sitting pretty to deliver a super-sequel. They were showing off the game on Xbox 360, 3DS and PlayStation Vita in London, and we played them all so there's a page about each right here. Engines on? OK, let's go. 

Molecules, Vol. 16, Pages 5807-5821: Synthesis and Spectroscopic Characterization of Two Tetrasubstituted Cationic Porphyrin Derivatives

An imidazolium tetrasubstituted cationic porphyrin derivative (the free base and its Zn(II) complex) with five-membered heterocyclic groups in the meso-positions were synthesized using microwave irradiation, and the compounds obtained characterized by 1H-NMR and mass spectrometry. We observed that under microwave irradiation the yield is similar to when the synthesis is performed under conventional heating, however, the time required to prepare the porphyrins decreases enormously. In order to investigate the electronic state of these compounds, we employed UV-Vis and fluorescence spectroscopy combined with quantum chemical calculations. The results reveal the presence, in both compounds, of a large number of electronic states involving the association between the Soret and a blue-shifted band. The Soret band in both compounds also shows a considerable solvent dependence. As for emission, these compounds present low quantum yield at room temperature and no solvent influence on the fluorescence spectra was observed.

Recently, double-gate MOSFETs (DGMOSFETs) have been shown to be more optimal for ultra-low power circuit design due to the improved subthreshold slope and the reduced leakage current compared to bulk CMOS. However, DGMOSFETs for subthreshold circuit design have not been much explored in comparison to those for strong inversion-based design. In this paper, various configurations of DGMOSFETs, such as tied/independent gates and symmetric/asymmetric gate oxide thickness are explored for ultra-low power and high efficient radio frequency identification (RFID) design. Comparison of bulk CMOS with DGMOSFETs has been conducted in ultra-low power subthreshold digital logic design and rectifier design, emphasizing the scope of the nano-scale DGMOSFET technology for future ultra-low power systems. The DGMOSFET-based subthreshold logic improves energy efficiency by more than 40% compared to the bulk CMOS-based logic at 32 nm. Among the various DGMOSFET configurations for RFID rectifiers, symmetric tied-gate DGMOSFET has the best power conversion efficiency and the lowest power consumption.

The Secret World Updated Impressions - First Dungeon Run

We get a guided tour of The Secret World's first dungeon at this year's Electronic Arts Studio Showcase.

In the online role-playing game The Secret World all your worst fears are now walking nightmares. As one of the three factions--the Illuminati, Knights Templar, or Dragon--you fight to suppress the beasts, or at least point them at your enemies. At this year's Electronic Arts Studio Showcase we got the chance to see this game in action as developer Funcom's Martin Bruusgaard, lead designer, and Joel Bylos, lead content designer, took us through the game's first dungeon in a hands-off demonstration.

Our character's first order of business was a meeting with some mysterious individuals in dark suits working for an even-more-mysterious corporation. They were monitoring a cargo ship that had recently run ashore--a ship that had also gone missing years earlier. Our hero, a bleach-blonde Southern tomboy, wasn't deterred by the less-than-ideal conditions and accepted the corporation's complimentary helicopter ride to the crash site. After touching down she met the rest of her party: a healer who specialized in blood magic, two high-damage characters using melee weapons and magical powers, and a sledgehammer-wielding tank.

The area itself was a rocky coastline beset by undead horrors and set against an overcast sky. And we're not talking about just mere zombies, but the mutated spirits of enraged Vikings as well. The Secret World pulls its foes from all corners of supernatural lore, from vampires and werewolves to the unspeakable horrors of H.P. Lovecraft. Back in the action, the first fight went off without a hitch, with the tank initiating the fight and the support characters bringing up the rear. The second encounter wasn't as simple.

At the center of the fight was a large, female Viking spirit who electrified the shallow water at our heroes' feet. Now the characters had to hop from island to island if they wanted to avoid a deadly shock. To us, it looked like the ranged characters did most of the work in finishing this encounter. Without missing a beat, the group turned from the fresh Viking corpse and ascended a cliff side. Along the way, they spotted an organic, quivering pod. Not one for mysteries, our tomboy ran up to poke at it (oh, like you wouldn't do the same) and the pod exploded. While initially unpleasant, this knowledge would come in handy later in the dungeon.

Our Funcom pilots then skipped ahead in the demo to the dungeon's first boss encounter. This was against a monstrous half-man/half-crab who had a bad habit of summoning zombies to his aid. To help even the odds our character called up her skill wheel, which was a series of concentric circles. Each circle was divided into different sections, each tied to one of The Secret World's roughly 500 skills. Players can mix and match their skills at will to build the type of character they want to play using an elegant interface reminiscent of an Apple product. Our hero switched out her melee-centric skills for some magical spells to deal with the upcoming threat.

Back in the battle, the crab-man fought in phases--alternating between attacking the group directly and hiding underwater and letting his zombie minions do the dirty work. Even with such tactics the boss was eventually overwhelmed. After sending the monster low our character spied a glowing green crate inside a shipping container. While enticing it required a code in order to be opened. A string of numbers was labeled on the side of the shipping container and we were told that, with a little out-of-game investigating, we could use those numbers to track down the needed code to open the crate.

Leaving the crate behind our pilots skipped ahead again to the dungeon's final battle. What awaited them was the Primordial Dweller, a massive creature of immense psychic power. Using its supernatural abilities, the creature tested all the tricks our heroes had been exposed to thus far. Remember those explosive pods? Now they were being hauled by kamikaze zombies who needed to be put down at a distance. It was a hard fight, but our side ultimately emerged victorious.

At this point, in a normal MMORPG, our character would have likely leveled up. However, in the Secret World, characters don't have levels. They advance in power by collecting stronger weapons and their experience points are used to purchase new skills that give them more variety. Our demonstration ended with a bang when, during their helicopter flight back, our group was attacked by a creature that looked not unlike the slumbering god, Cthulhu. The Secret World is showing a lot of promise, especially in its incorporation of alternate-reality gaming elements. It's release date, however, remains a well-guarded secret. We'll be sure to keep you updated as we learn more about this game.

Big In Japan June 27- July 3: Tales of the Abyss 3DS

Portable version of PS2 RPG pushes sales for system; Super Street Fighter IV: AE and GoldenEye 007 tailing behind.

 

While the 3DS version of Tales of the Abyss will be localized in English sometime this year, the Japanese version seems to be doing well; so much so that it's on the top spot of the recent update of Media Creates' sales chart. The RPG from Namco Bandai sold 74,173 copies on the week of June 27 through July 3. Tales of the Abyss is the eighth mothership title in the Tales series which introduces the Flex Range Linear Motion Battle system that allows freedom in movement and running across 3D space during combat.

Occupying second place is the PS3 version of Super Street Fighter IV: Arcade Edition. This version of the game sold 54,924 on its debut week, while the Xbox 360 version is in 12th place with 12,326 units sold. The second update to the fighting game introduces four new playable characters and a rebalanced character roster. The former king of the charts last week, Meruru no Atelier: Alchemist no Renkinjutsushi 3, is now at seventh place with 23,756 units sold. The 3DS version of Legend of Zelda: Ocarina of Time is still staying strong in the top ten, residing at sixth place with 24,190 units sold.

New to Japan is Test Drive Unlimited 2, the racing game that came out on March outside of the region. The PS3 version sold 15,254 units and resides in 10th place, while the Xbox 360 sold 7,765 units and is at 19th place. The Wii version of GoldenEye 007 also made its debut on the charts; the game is in fourth place with 36,174 units sold. Also interesting to note is the position drop of Ryu Ga Gotoku: Of the End, with the title now at 14th place with 11,867 units sold.

Over at the hardware side, the 3DS now takes the top slot thanks in huge part to the portable debut of Namco Bandai's RPG. The 3DS sold 30,233, barely beating the PSP and its unit sales count of 29,871. Otherwise, the hardware positions are about the same as last week's charts.
