Knoevenagel condensation of 2-(benzothiazol-2-ylthio) acetonitrile (2) with either furan-2-carbaldehyde or thiophene-2-carbaldehydes leads to E-isomers 4a–b exclusively, while the condensation of the compound 2 with benzaldehyde or para-substituted benzaldehydes with an electron-donating group afforded E/Z mixtures 4c–e with preferentially formation of the E-isomer. Condensation of furan-2-carbaldehyde (3a) with either 1-(benzothiazol-2'-ylthio) propan-2-one (5) or 2-(2'-oxo propylthio)-3-phenyl-quinazolin-4(3H)-one (9) leads exclusively to the Z-isomers of 6 and 10, respectively. The structures of the newly synthesized compounds were elucidated by elemental analyses, 1H-NMR and 13C-NMR spectra, COSY, HSQC, HMBC, NOE, MS and X-ray crystallographic investigations.

Catalytic hydrogenation of 3,5-bis-arylidenetetramic acids, known for their biological activity, has been developed. The chemoselective ruthenium-catalyzed reduction of the exocyclic carbon-carbon double bonds on pyrrolidine-2,4-dione ring system, containing other reducible functions, has been investigated. Depending on the substrate the yield of the hydrogenation process can reach up to 95%. The structural elucidation has been established using NMR and HRMS spectral data.

Molecules, Vol. 16, Pages 6092-6115: The Stereochemistry of Complex Polyketide Biosynthesis by Modular Polyketide Synthases

Polyketides are a diverse class of medically important natural products whose biosynthesis is catalysed by polyketide synthases (PKSs), in a fashion highly analogous to fatty acid biosynthesis. In modular PKSs, the polyketide chain is assembled by the successive condensation of activated carboxylic acid-derived units, where chain extension occurs with the intermediates remaining covalently bound to the enzyme, with the growing polyketide tethered to an acyl carrier domain (ACP). Carboxylated acyl-CoA precursors serve as activated donors that are selected by the acyltransferase domain (AT) providing extender units that are added to the growing chain by condensation catalysed by the ketosynthase domain (KS). The action of ketoreductase (KR), dehydratase (DH), and enoylreductase (ER) activities can result in unreduced, partially reduced, or fully reduced centres within the polyketide chain depending on which of these enzymes are present and active. The PKS-catalysed assembly process generates stereochemical diversity, because carbon–carbon double bonds may have either cis- or trans- geometry, and because of the chirality of centres bearing hydroxyl groups (where they are retained) and branching methyl groups (the latter arising from use of propionate extender units). This review shall cover the studies that have determined the stereochemistry in many of the reactions involved in polyketide biosynthesis by modular PKSs.

Crystals of trans-Pt2(μ-X)2X2(CO)2 (X = Br, I) have been grown and their molecular and crystalline structures have been solved by X-ray diffraction methods. In both cases the dinuclear molecules are bent, with a bending angle of 164.6° and 156.5° for the bromide and the iodide, respectively. While the structure of the bromo-derivative is reported here for the first time, a modification of trans-Pt2(μ-I)2I2(CO)2 with planar centrosymmetric molecules is known. This appears to be a rare case of a platinum(II) halo-bridged derivative structurally characterized in both bent and planar forms.

A randomized complete randomized design (RCBD) 3 by 3 experiment was designed to investigate and distinguish the relationships among production of secondary metabolites (total phenolics, TP; total flavonoids, TF), gluthatione (GSH), oxidized gluthatione (GSSG), soluble carbohydrate and antioxidant activities of the Malaysian medicinal herb Labisia pumila Blume under three levels of CO2 enrichment (400, 800 and 1,200 µmol mol−1) for 15 weeks. It was found that the treatment effects were solely contributed by interaction of CO2 levels and secondary metabolites distribution in plant parts, GSH, GSHH and antioxidant activities (peroxyl radicals (ROO), superoxide radicals (O2), hydrogen peroxide (H2O2) and hydroxyl radicals (OH). The records of secondary metabolites, glutahione, oxidized gluthathione and antioxidant activities in a descending manner came from the leaf enriched with 1,200 µmol/mol CO2 > leaf 800 µmol/mol CO2 > leaf 400 µmol/mol CO2 > stem 1,200 µmol/mol CO2 > stem 800 µmol/mol CO2 > stem 400 µmol/mol CO2 > root 1,200 µmol/mol CO2 > root 800 µmol/mol CO2 > root 400 µmol/mol CO2. Correlation analyses revealed strong significant positive coefficients of antioxidant activities with total phenolics, flavonoids, GSH and GSHH indicating that an increase in antioxidative activity of L. pumila under elevated CO2 might be up-regulated by the increase in production of total phenolics, total flavonoids, GSH, GSHH and soluble sugar. This study implied that the medicinal potential of herbal plant such as L. pumila can be enhanced under elevated CO2, which had simultaneously improved the antioxidative activity that indicated by the high oxygen radical absorbance activity against ROO, O2, H2O2, and OH radicals.

The screening of several Chinese mangrove plants for insecticidal principles showed that ethanol extract of Ceriops tagal stems and twigs possessed significant feeding deterrent activity against the red flour beetle, Tribolium castaneum (Family: Rhizophoraceae). From the ethanol extract, three feeding deterrent diterpenoids were isolated by bioassay-guided fractionation. The compounds were identified as tagalsin A, B, and H on the basis of their phytochemical and spectral data. Tagalsin A, B, and H exhibited strong feeding deterrent activity against T. castaneum adults with EC50 values of 375.3 ppm, 277.3 ppm, and 285.45 ppm, respectively.

Bleach: Soul Resurreccion Hands-On Preview

We dish out our "imprescions" on the hit anime/manga series' upcoming videogame adaptation

 

To count the number of all Bleach videogame spin-offs on one hand would be impossible. If you are referring to the English counterparts, however, there are only four of them, with the most recent one being Bleach: The 3rd Phantom for the DS. Fans hoping to check out an action game without resorting to spending lots on importing fees can look forward to the upcoming adaptation called Bleach: Soul Resurreccion, exclusively for the PS3.

For our recent playthrough, we checked out three stages: two of them starring main hero Ichigo Kurosaki and one of them with Quincy warrior Uryu Ishida. All of them involve the main heroes slashing hordes upon hordes of ghosts with different types of skullcaps called hollows. Sometimes, you'll fight a trio of giant hollows called menos grandes that bear a resemblance to the outfits of a plague doctor in the Renaissance era. Staying true to the Hueco Mundo arc of the series, you'll also be squaring off mano-a-mano against evolved Hollow lieutenants called the arrancars, with one of them mimicking a Saint Seiya reject with added blue long hair and a tail.

Controlling our main hero Ichigo is pretty easy: square and triangle are his normal and Spirit attacks respectively, while pressing circle unleashes an area-clearing attack. You can dash all across the Hueco Mundo by pressing and holding down the R2 button. Pressing the L2 button when you've killed enough enemies will unleash a character's unique power-up: in the case of Ichigo, he'll put on his Hollow mask and be all powered-up while also having access to a screen-clearing ultra attack upon pressing the L2 button again.

To defend yourself, you have to hold down R1 to block; dodging attacks via flash steps require you to push towards any direction on the left thumbstick while holding the block button. You can single out a lone target by pressing the L1 once, in which the cursor will lock onto the enemy that's within the centre-most point of the camera. All forms of spirit moves require spirit energy (located at the bottom of your health) that regenerates over time as long as you're not dashing. Exhausting the energy until depletion will increase the regeneration time, so you'll have to be careful not to drain it too much when using spirit attacks.

Uryu's main weapon is his ginrei kojaku, which is a pentacle-shaped plasma-shooting bow. Pressing Triangle in succession unleashes white arrows followed up by a wide blast that pushes Hollows away. Pressing circle lets Uryu use his ginto abilities – spells that use up liquid spirit energy from tiny vials- to clear his surrounding area of enemies. Players can look forward to controlling other Bleach mainstays like soul reapers Rukia Kuchiki, Toshiro Hitsugaya and Byakuya Kuchiki. The game also allows you to level up characters and upgrade their moves. Upon collecting the game's equivalent of experience points, they can unlock triple jumps, ability cancels and guard counter-attacks to help combat enemies.

While the combat feels satisfying upon first glance, non-fans will definitely not find much appeal to keep pressing on. There isn't a way for newbies to get in tune to the game since the bits and pieces that constitute as narratives are all over the place. The game could also benefit new players with a lexicon and dictionary ala the Codex option in Mass Effect, since most of the key abilities and skills are alien to non-fans.

The camera also got in the way while we were dashing around in confined spaces. Save for the end-of-level bosses which would require some use of dodging and running-around the wide open battlefield, the Hollow army did not put up much of a challenge and we resorted to spamming their character's respective spirit attacks while conserving their spirit energy to get ahead and not get hurt at all. Even if we got hurt, a greyed-out part of our health bar would regenerate over time, so we would probably have to try really, really hard to fail. In short, we didn't feel at the time that the aforementioned counter-attacks and upgrades were necessary in finishing a segment.

Even with the prospects of reliving a rather large and, arguably, exciting part of the Bleach storyline in videogame form, we admitted that the experience could use a lot more meat and thought. Here's hoping that the rest of the game can change our perceptions when the localized version is out in the US on August 2 and in Asia next week on July 28.

Final Fantasy Type-0 set for October in Japan

Two UMD disc action-RPG coming out this year; retails for 7,700 yen.

 

With Final Fantasy XIII already out and its sequel's release date set, it was only a matter of time until Square Enix confirmed the release of the two other Final Fantasy titles in the Fabula Nova Crystallis series: Final Fantasy Versus XIII and Final Fantasy Type-0. At long last, the company has finally revealed the release date of the latter in Japan: October 13.

The announcement that came with a brand-new trailer was made during the United States of Odaiba event, which started on July 16 and will carry on until August 31. Previously, the game was set for a summer release this year but due to "various circumstances" stemming from the April PlayStation Network outage, the game's release schedule was pushed back.

The trailer itself showcased a blend of in-game action and cutscenes, introducing possible antagonist and high-ranking commander of the Milites empire Cid Aulstyne, Suzaku's head of magic Arecia Al-Rashia, Suzaku's 174th director Khalia Chival VI, and Milites empire brigadier general Qator Bashtar. The game's theme song, dubbed "Zero", is sung by Japanese group Bump of Chicken.

The action RPG will retail for 7,700 yen and will come in two UMD discs. An English version of the game has yet to be announced. For more information, check out GameSpot's coverage on the title.

Family Guy Online open beta targeted for this year

Q&A: Fox and Roadhouse say free-to-play, microtransaction-based title will have RPG elements, show writers include Alex Carter and Andrew Goldberg.

20th Century Fox may have initially been skeptical of Family Guy's prospects, going so far as to cancel the unabashedly crude cartoon in 2001. However, the production company has changed its tune considerably in the here and now. Today, Fox officially announced its Web browser-based, free-to-play title Family Guy Online, saying that the 3D title is targeted to enter open beta this year.

Comic-Con 2011 will act as a coming out of sorts for Family Guy Online, even if its official website has been active since mid-June. However, gamers can expect a good deal more content on that website today, as Fox and developer Roadhouse Interactive have set live Family Guy Online's character creator to promote the title.

The real question, though, is what does Family Guy Online have for fans of the show?

"It's just a great way to extend the television show and allow users to engage with each other in a community in a game product that is being developed really in association with the show," Gary Rosenfeld, senior vice president, new media for Fox Consumer Product, told GameSpot in an interview. That collaboration includes help from two senior writers on the show: Alex Carter and Andrew Goldberg.

The experience begins with character creation, and Roadhouse Interactive cofounder and chief creative officer Ian Verchere told GameSpot that players will have a wealth of customization options. Verchere also noted that rather than having players assume the identity of, say, Peter Griffin, they will be able to select from four character-class archetypes that typify the different attributes of the dysfunctional family.

Fox and Roadhouse aren't entirely ready to talk about Family Guy Online's actual gameplay, but Verchere indicated that it will have elements akin to the role-playing game genre. Those mechanics include character progression and experience points. The game has both single-player and multiplayer options.

"We're not just sticking Family Guy into Champions Online or making Dungeons and Quahog or whatever," he explained. "In the same way that the show riffs on popular culture and on TV, [we've been thinking about] an intelligent way to bring Family Guy online. At its heart, you are going to earn experience and progress through the game. There are familiar structures that gamers will be able to relate to and engage with, but it is going to feel like Family Guy."

"When you come into the game and create the character, you can literally create billions of different characters," he continued. "When you go into the game, you're not going to play as you would in a console game as, say, Peter Griffin. But when you build your own character, it makes sense from a game mechanic perspective to associate with the positive and/or negative aspects of what each of the Griffins represent. If you look at how you would map out an attribute table, Peter is obviously big and slow and tank-like and Stewie is small and ranged attack. OK, there, I said 'ranged attack.'"

Of course, Family Guy Online will come heavily laden with personalities from the show--some 150 in total. "Once the users gets into the game, the user will interact with show characters," Rosenfeld said. "The show characters act as guides. They will send the user off on adventures that allow [them] to advance through the game. "

Rosenfeld also expressed optimism over Family Guy Online's ability to react to current and popular culture trends. In other words, the game will be an open-ended experience where players will be afforded new and culturally relevant quests.

"The idea behind the project, or the thing that's most exciting about it, is that we will be able to react to certain elements that the show can't react to because of the production time line," he said. "So for instance things going on in pop culture that we might want to riff on, we'll be able to react to as a result of how we set this up. Building it in Unity, we've got the flexibility to update content, add new content, pretty quick turnarounds in regular intervals."

Family Guy Online will be free-to-play, where virtual currency and in-game goods rule the day. Rosenfeld explained that the game will include microtransactions, and the studio will likely also make use of ad and sponsorship components as well as other premium content.

Rosenfeld also briefly addressed the Family Guy game that is in development at Activision, saying that Family Guy Online exists entirely apart from that project. "We want to make sure there is a Family Guy experience for fans on all the different platforms," he said. Rosenfeld declined to comment on whether that game is still on for this year.

Fox and Roadhouse plan to have a significant presence at Comic-Con this week. In addition to having Family Guy Online's character creator up and running on the show floor, Quahog mayor (and seminal Batman actor) Adam West will be on hand to sign exclusive posters for the game. Comic-Con attendees who create a character at the show will also earn an exclusive in-game item.

Cancelled Red Fly project Star Wars-related?

Resume of ex-Red Fly environment artist reveals studio was working on now-cancelled Star Wars title.

 

Earlier this year, Austin-based Red Fly Studio--the team behind the Wii version of Sega's Thor: God of Thunder--laid off 30 staff members due to the unexpected cancellation of "a large, high-profile project."

It now appears that project may have been a Star Wars title, with the online resume of an ex-Red Fly developer (discovered first on Twitter) revealing he had worked on a "next-gen Star Wars title" from January to June 2011, after the studio's completion of Thor: God of Thunder for the Wii.

Red Fly is best known for 2008's original Mushroom Men games on the Wii and DS. Its highest-profile games to date have been Wii versions of Ghostbusters: The Video Game, Star Wars: The Force Unleashed II, and Thor: God of Thunder.

Hulk Hogan, Sasha Grey line up for Saints Row: The Third

Wrestler, actress, and Daniel Dae Kim to lend their voices to THQ's upcoming open-world crime sequel.

 

THQ has wasted no time prepping the world for the release of the third installment of its open world over-the-top crime series Saints Row, recently announcing a Platinum Edition and preorder pack for the game.

Saints Row fans now have even more reason to be excited ahead of the November 15 release of Saints Row: The Third, with THQ announcing Hollywood talent Hulk Hogan, Sasha Grey, and Daniel Dae Kim will all lend their voices to key characters in the game.

Professional wrestler Hulk Hogan will step into the role of Angel De LaMuerte, a wrestler betrayed by his former tag-team partner who teams with the Saints to take over Steelport, while former porn star turned Hollywood actress Sasha Grey (The Girlfriend Experience, Entourage) will voice the character of Viola DeWynter, one half of the deadly sisterly duo masterminds of the Syndicate's financial empire.

Daniel Dae Kim (Saints Row 2, Lost, Hawaii Five-O) will reprises his role as Johnny Gat, chief lieutenant of the Third Street Saints who has now become an international celebrity, plastering his face on Saints clothing lines, energy drinks, and automobiles.

THQ has confirmed both Sasha Grey and Hulk Hogan will be attending Comic-Con in San Diego this week to sign autographs on Thursday, July 21 at the Saints Row Booth.
