Molecules, Vol. 17, Pages 12851-12867: In Vitro Antioxidant Effects of Aloe barbadensis Miller Extracts and the Potential Role of These Extracts as Antidiabetic and Antilipidemic Agents on Streptozotocin-Induced Type 2 Diabetic Model Rats

In this study, the total phenolic and flavonoid contents, the 2,2-diphenyl-1-picryl hydrazyl (DPPH) radical scavenging ability and the ferric reducing power (FRAP) of Aloe vera were measured to determine the antioxidant activity of this species. The in vivo antidiabetic effects of the plant were also investigated using streptozotocin-induced type 2 diabetic model rats that were divided into five groups based on the treatment received: (1) water (WC); (2) glibenclamide; (3) concentrated gel extract (Gel-C); (4) ethanol (80%) gel extract (Gel-Et); and (5) ethanol (80%) skin extract of Aloe vera (Skin-Et). Skin-Et, which contained the highest level of total phenolics (62.37 ± 1.34 mggallic acid/kg) and flavonoids (20.83 ± 0.77 mg/kg), exhibited the highest scavenging activity (85.01 ± 0.52%) and the greatest reducing power (185.98 ± 0.41 µM), indicating that the skin contained the highest level of antioxidants. The oral consumption of Gel-Et for 4 weeks a caused significant reduction in the fasting serum glucose levels of the rats. The rats in the Gel-C-, Gel-Et- and Skin-Et-treated groups experienced a reduction in their total cholesterol levels by 11%, 17% and 25%, respectively and a reduction in their LDL cholesterol levels by 45%, 3% and 69%, respectively. The in vivo experimental antioxidant parameter MDA is strongly correlated with the in vitro antioxidant parameters of flavonoids and polyphenols, namely the DPPH and FRAP values (r = 0.94, 0.92, 0.93, 0.90), thus confirming the antioxidant potential of the Aloe vera extracts.

Tilapia frame protein was hydrolyzed by different proteases, including properase E, pepsin, trypsin, flavourzyme, neutrase, gc106 and papain, to obtain antioxidant peptides. The tilapia frame protein hydrolysate (TFPH) obtained by trypsin exhibited the highest degree of hydrolysis and antioxidant activity. Three series of peptides (TFPH1, TFPH 2 and TFPH 3) were obtained by ultrafiltration of TFPH through molecular weight cut-off membranes of 5, 3 and 1 kDa, respectively, and their IC50 values on scavenging 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide anion radical (•O2), hydrogen peroxides (H2O2) and hydroxyl radical (•OH) activities were determined and compared with glutathione (GSH). The results showed that TFPH1 had the highest antioxidant activity. TFPH1 was further purified using ion exchange chromatography, gel filtration chromatography, and reversed phase high performance liquid chromatography (RP-HPLC). Finally, two antioxidant peptides were identified and the amino acid sequences were identified as Asp-Cys-Gly-Tyr (456.12 Da) and Asn-Tyr-Asp-Glu-Tyr (702.26 Da), respectively. The IC50 values of two peptides on hydroxyl radical scavenging activity were 27.6 and 38.4 μg/mL, respectively.

The Kabachnik–Fields (phospha-Mannich) reaction involving the condensation of primary or secondary amines, oxo compounds (aldehydes and ketones) and >P(O)H species, especially dialkyl phosphites, represents a good choice for the synthesis of α-aminophosphonates that are of significant importance due to their biological activity. In general, these three-component reactions may take place via an imine or an α-hydroxy-phosphonate intermediate. The monitoring of a few Kabachnik–Fields reactions by in situ Fourier transform IR spectroscopy has indicated the involvement of the imine intermediate that was also justified by theoretical calculations. The Kabachnik–Fields reaction was extended to >P(O)H species, comprising cyclic phosphites, acyclic and cyclic H-phosphinates, as well as secondary phosphine oxides. On the other hand, heterocyclic amines were also used to prepare new α-amino phosphonic, phosphinic and phosphine oxide derivatives. In most cases, the synthesis under solvent-free microwave (MW) conditions is the method of choice. It was proved that, in the cases studied by us, there was no need for the use of any catalyst. Moreover, it can be said that sophisticated and environmentally unfriendly catalysts suggested are completely unnecessary under MW conditions. Finally, the double Kabachnik–Fields reaction has made available bis(phosphonomethyl)amines, bis(phosphinoxidomethyl)amines and related species. The bis(phosphinoxidomethyl)amines serve as precursors for bisphosphines that furnish ring platinum complexes on reaction with dichlorodibenzonitriloplatinum.

Deoxynivalenol (DON), mainly produced by Fusarium fungi, and also commonly called vomitoxin, is a trichothecene mycotoxin. It is one of the most abundant trichothecenes which contaminate cereals consumed by farm animals and humans. The extent of cereal contamination is strongly associated with rainfall and moisture at the time of flowering and with grain storage conditions. DON consumption may result in intoxication, the severity of which is dose-dependent and may lead to different symptoms including anorexia, vomiting, reduced weight gain, neuroendocrine changes, immunological effects, diarrhea, leukocytosis, hemorrhage or circulatory shock. During the last two decades, many studies have described DON toxicity using diverse animal species as a model. While the action of the toxin on peripheral organs and tissues is well documented, data illustrating its effect on the brain are significantly less abundant. Yet, DON is known to affect the central nervous system. Recent studies have provided new evidence and detail regarding the action of the toxin on the brain. The purpose of the present review is to summarize critical studies illustrating this central action of the toxin and to suggest research perspectives in this field.

Scorpion toxins have been central to the investigation and understanding of the physiological role of potassium (K+) channels and their expansive function in membrane biophysics. As highly specific probes, toxins have revealed a great deal about channel structure and the correlation between mutations, altered regulation and a number of human pathologies. Radio- and fluorescently-labeled toxin isoforms have contributed to localization studies of channel subtypes in expressing cells, and have been further used in competitive displacement assays for the identification of additional novel ligands for use in research and medicine. Chimeric toxins have been designed from multiple peptide scaffolds to probe channel isoform specificity, while advanced epitope chimerization has aided in the development of novel molecular therapeutics. Peptide backbone cyclization has been utilized to enhance therapeutic efficiency by augmenting serum stability and toxin half-life in vivo as a number of K+-channel isoforms have been identified with essential roles in disease states ranging from HIV, T-cell mediated autoimmune disease and hypertension to various cardiac arrhythmias and Malaria. Bioengineered scorpion toxins have been monumental to the evolution of channel science, and are now serving as templates for the development of invaluable experimental molecular therapeutics.

Dengue viruses and other members of the Flaviviridae family are emerging human pathogens. Dengue is transmitted to humans by Aedes aegypti female mosquitoes. Following infection through the bite, cells of the hematopoietic lineage, like dendritic cells, are the first targets of dengue virus infection. Dendritic cells (DCs) are key antigen presenting cells, sensing pathogens, processing and presenting the antigens to T lymphocytes, and triggering an adaptive immune response. Infection of DCs by dengue virus may induce apoptosis, impairing their ability to present antigens to T cells, and thereby contributing to dengue pathogenesis. This review focuses on general mechanisms by which dengue virus triggers apoptosis, and possible influence of DC-apoptosis on dengue disease severity.

The emergence of a highly pathogenic avian influenza virus H5N1 has increased the potential for a new pandemic to occur. This event highlights the necessity for developing a new generation of influenza vaccines to counteract influenza disease. These vaccines must be manufactured for mass immunization of humans in a timely manner. Poultry should be included in this policy, since persistent infected flocks are the major source of avian influenza for human infections. Recombinant adenoviral vectored H5N1 vaccines are an attractive alternative to the currently licensed influenza vaccines. This class of vaccines induces a broadly protective immunity against antigenically distinct H5N1, can be manufactured rapidly, and may allow mass immunization of human and poultry. Recombinant adenoviral vectors derived from both human and non-human adenoviruses are currently being investigated and appear promising both in nonclinical and clinical studies. This review will highlight the current status of various adenoviral vectored H5N1 vaccines and will outline novel approaches for the future.

Cell penetrating peptides (CPPs) can deliver cell-impermeable therapeutic cargos into cells. In particular, CPP-cargo conjugates tend to accumulate inside cells by endocytosis. However, they often remain trapped inside endocytic organelles and fail to reach the cytosolic space of cells efficiently. In this review, the evidence for CPP-mediated endosomal escape is discussed. In addition, several strategies that have been utilized to enhance the endosomal escape of CPP-cargos are described. The recent development of branched systems that display multiple copies of a CPP is presented. The use of viral or synthetic peptides that can disrupt the endosomal membrane upon activation by the low pH of endosomes is also discussed. Finally, we survey how CPPs labeled with chromophores can be used in combination with light to stimulate endosomal lysis. The mechanisms and challenges associated with these intracellular delivery methodologies are discussed.

Both geniposide (Ge) and borneol (Bo) are bioactive substances derived from traditional Chinese medicine. Injections containing co-compound of Gardenia-Borneol are widely used for stroke treatment in China, such as “Xingnaojing” multi-component injection. As more and more adverse reactions (especially drug allergy) were reported, it is urgent to find more effective and safer routes of administration for such kinds of medicines. In this paper, bioavailabilities and brain-target effects of geniposide in Gardenia-Borneol co-compound through different administration routes in mice were investigated. Geniposide concentrations in plasma and in brain of mice were determined by reversed-phase high-performance liquid chromatography. The pharmacokinetics parameters of intranasal (i.n.) and intragastric (i.g.) administration were compared with intravenous (i.v.) administration. The bioavailabilities of Ge were 85.38% and 28.76% for i.n. and i.g. while Tmax were 1 min and 30 min. Cmax were 21.881 ± 5.398, 1.914 ± 0.327 and 42.410 ± 6.268 μg/mL for i.n., i.g. and i.v., respectively. The AUC of Ge in brain were 32413.6 ± 4573.9, 6440.1 ± 863.7 and 37270.5 ± 4160.6 ng/g·min for i.n., i.g. and i.v., respectively. The drug target indexes (DTI) were 1.02 and 0.60 for i.n. and i.g. The results demonstrated that geniposide could be absorbed promptly and thoroughly by i.n. administration in mice and basically transported into the brain though blood vessel passways.

Geosciences, Vol. 2, Pages 260-276: Examining Local Climate Variability in the Late Pennsylvanian Through Paleosols: An Example from the Lower Conemaugh Group of Southeastern Ohio, USA

Three temporally close stratigraphic sections were excavated in Glenshaw Formation of Athens County, Ohio. The described units are Upper Pennsylvanian (Gzhelian, 305–302 Ma) and located in the distal portion of the Appalachian foreland basin. Mudstone units interpreted as paleosols were identified across all three sections. Detailed field and micromorphological studies lead to the recognition of two separate paleosols within the profile. The profile consists of a composite paleosol composed of two cumulative paleosols. The lower paleosol is interpreted as a calcic Vertisol which formed in a seasonally dry environment whereas the upper paleosol is interpreted as a gleyed Inceptisol which formed in a seasonally wet environment. The change in paleosol types is the result of increased precipitation which led to saturation of the soil and surface ponding. Pedogenic carbonate nodules are a common feature throughout the entire profile as are stress cutans. A coalesced carbonate horizon (Bk) was observed approximate 120 cm from the top of the profile in all three sections. This carbonate horizon formed in the Vertisol and later served as a barrier which limited the downward movement of surface water. This limited the gleization of the bottom portion of the overprinted Vertisol resulting in a diffuse boundary with the overlying Inceptisol and producing a composite paleosol.

In this study, a novel 96-well microplate swivel system (M96SS) was built for high-throughput screening of microalgal strains for CO2 fixation. Cell growth under different CO2 supply conditions (0.2, 0.4, 0.8, and 1.2 g L−1 d−1), residual nitrate, and pH value of Chlorella sp. SJTU-3, Chlorella pyrenoidosa SJTU-2, and Scenedesmus obliquus SJTU-3 were examined in the M96SS and traditional flask cultures. The dynamic data showed there was a good agreement between the systems. Two critical problems in miniature culture systems (intra-well mixing and evaporation loss) were improved by sealed vertical mixing of the M96SS. A sample screen of six microalgal species (Chlorella sp. SJTU-3, Chlorella pyrenoidosa SJTU-2, Selenastrum capricornutum, Scenedesmus obliquus SJTU-3, Chlamydomonas sajao, Dunaliella primolecta) was carried out in flasks and the M96SS. Chlamydomonas sajao appeared to be a robust performer (highest cell density: 1.437 g L−1) in anaerobic pond water with 0.8, and 1.2 g L−1 d−1 CO2. The reliability and efficiency of the M96SS were verified through a comparison of traditional flask culture, M96SS, Lukavský's system, and a microplate shaker.

Evaluation is increasingly important in decision-making processes for the sustainable planning and design of port plans. It acts as a support for plan preparation, for making values, interests and needs explicit, and for exploring the components of the decision-making process itself. Evaluation can be likened to an “implicit tool” that can integrate approaches, methodologies and models, adapting to the many needs revealed during the decision-making process. New sustainability challenges call for new approaches to creating frameworks for the analysis and evaluation of plans and projects that allow the integration of multidimensional goals and values. Utilizing some selected case studies of port plans in six Italian cities, this paper explores how environmental assessment can become a tool for dialog and interaction among different fields of expertise to support dynamic learning processes, knowledge management and the creation of shared choices, using suitable approaches and tools. In this view, Integrated Spatial Assessment (ISA) can be useful in supporting decision-making processes on different scales and institutional levels to stimulate dialog between technical and political evaluations, referring to complex values that are part of conflicting and changing realities in which it has become imperative to operate according to sustainability principles.

This article discusses the contention between the state and local Guji people on issues of development and conservation of a Protected Area—Nech Sar National Park in southern Ethiopia. The park, which covers over 514 square kilometers, is a contested space between different actors, not only for its economic values, but it is also an arena of contestation over development and conservation perspectives. Since its inception as a national park in 1974, it has been administered with strict protectionist conservation approach, and later in 1990s, the ‘modernist’ development program was introduced in the form of ecotourism. On the contrary, the Guji people had strong determination for conservation embedded deep in their worldviews and beliefs. By tracing the genesis of the philosophies behind protected areas in Africa, particularly how it was adopted by the Ethiopian state and its implications, I argue that contrasts in environmental cosmologies between the western and indigenous perspectives have ultimately resulted in unsustainable resource management and also disrupted local livelihood conditions. Despite its existence as an independent country, Ethiopia also experienced similar conservation models that were imported to colonial Africa. In this article, I argue that conservation, particularly in the form of protected areas, is a form of hegemonic control over territories, people and their spaces (historical, economic, cultural and political spaces).

In transmitting image/video data over Video Sensor Networks (VSNs), energy consumption must be minimized while maintaining high image/video quality. Although image/video compression is well known for its efficiency and usefulness in VSNs, the excessive costs associated with encoding computation and complexity still hinder its adoption for practical use. However, it is anticipated that high-performance handheld multi-core devices will be used as VSN processing nodes in the near future. In this paper, we propose a way to improve the energy efficiency of image and video compression with multi-core processors while maintaining the image/video quality. We improve the compression efficiency at the algorithmic level or derive the optimal parameters for the combination of a machine and compression based on the tradeoff between the energy consumption and the image/video quality. Based on experimental results, we confirm that the proposed approach can improve the energy efficiency of the straightforward approach by a factor of 2~5 without compromising image/video quality.

Given that comparative theology is aimed at learning from the insights of other religious traditions, the comparative theologian’s confessional perspective must be engaged and subject to possible transformation through the discovery of truth in those traditions. Despite Francis Clooney’s and James Fredericks’ attempts to distance comparative theology from the theology of religions, its truth-seeking dimension makes participation in the theology of religions unavoidable. Crucial to integrating what is learned, moreover, is a willingness to allow presuppositions about the other to be challenged and to make revisions if necessary. Keith Ward exhibits this willingness but, on this basis, distinguishes comparative theology from confessional theology, thus obscuring the legitimacy of revision from a committed religious standpoint. Where comparative theologians are willing and able to integrate all that is learned through their study of other traditions, comparative theology can be conceived of as both a confessional enterprise and a contribution to what Wilfred Cantwell Smith called ‘World Theology’—that is, the ongoing attempt to give intellectual expression to the faith of us all.

Experimentally measured mechanical properties of hollow sphere steel foam are the subject of this paper. The characterization of the hollow sphere foam encompasses compressive yield stress and densification strain, compressive plastic Poisson’s ratio, and compressive unloading modulus, as well as tensile elastic modulus, tensile unloading modulus, tensile yield stress, and tensile fracture strain. Shear properties are also included. These tests provide sufficient information to allow calibration of a macroscopic, continuum constitutive model. Calibrated foam plasticity parameters are tabulated, and unique feature of foam plasticity are explained. Also, initial development of mesoscale simulations, which explicitly model voids and sintered hollow spheres, is reported. This work is part of a larger effort to help the development of steel foam as a material with relevance to civil engineering applications.

Insects, Vol. 3, Pages 1143-1155: Individual Variability of Nosema ceranae Infections in Apis mellifera Colonies

Since 2006, beekeepers have reported increased losses of Apis mellifera colonies, and one factor that has been potentially implicated in these losses is the microsporidian Nosema ceranae. Since N. ceranae is a fairly recently discovered parasite, there is little knowledge of the variation in infection levels among individual workers within a colony. In this study we examined the levels of infection in individual bees from five colonies over three seasons using both spore counting and quantitative real-time PCR. The results show considerable intra-colony variation in infection intensity among individual workers with a higher percentage of low-level infections detected by PCR than by spore counting. Colonies generally had the highest percentage of infected bees in early summer (June) and the lowest levels in the fall (September). Nosema apis was detected in only 16/705 bees (2.3%) and always as a low-level co-infection with N. ceranae. The results also indicate that intra-colony variation in infection levels could influence the accuracy of Nosema diagnosis.
