Context: Further Selected Essays on Productivity, Creativity, Parenting, and Politics in the 21st Century

Posted: 03 Oct 2011 03:19 AM PDT

Context: Further Selected Essays on Productivity, Creativity, Parenting, and Politics in the 21st Century, my second essay collection, is now officially available from Tachyon Books, and in finer bookstores everywhere. It features an introduction by the estimable Tim O'Reilly, as well as a walloping 44 essays that were previously published in various magazines, newspapers and websites. As with my other books, the whole text is available as a Creative Commons BY-NC-SA download for your remixing pleasure. I'm also in search of libraries and school that would like free copies of the book sent to them by donors.

From Tim's introduction:

Edwin Schlossberg once said "The skill of writing is to create a context in which other people can think." And oh, how we need that skill today!

In times of transition and upheaval, we are literally "off the map" of past experience that is our normal guide to what to expect and how to think about it. It's at times like these that we need context-setters to shape how we understand and think about the changes facing us.

It was clear from the first that Cory Doctorow is one of the great context-setters of our generation, helping us all to understand the implications of the technology being unleashed around us. We are fortunate that unlike many who practice this trade, who look backward at recent changes, or forward only a year or two, Cory uses the power of story to frame what is going on in larger terms.

Sensors, Vol. 11, Pages 9450-9466: Biosensor Applications in the Field of Antibiotic Research—A Review of Recent Developments

Posted: 03 Oct 2011 12:00 AM PDT

Antibacterials are among of the most important medications used in health care. However, their efficacy is increasingly impeded by a tremendous and globally spread bacterial resistance phenomenon. This bacterial resistance is accelerated by inadequate application of antibacterial drugs in humans, the widespread veterinary use of antibacterials, and antibacterial occurrence in the environment and food. Further, there is a lack of development of innovative novel drugs. Therefore, the search for novel antibacterials has to be intensified and the spread of antibacterials in the environment has to be restricted. Due to the fundamental progress in biosensor development and promising applications in the antibiotic field, this review gives for the first time an overview on the use and prospects of biosensor applications in that area. A number of reports have applied biosensors of different design and techniques to search for antibacterials in environmental and foodstuff matrices. These studies are discussed with respect to the analytical values and compared to conventional techniques. Furthermore, biosensor applications to elucidate the mode of action of antimicrobial drugs in vitro have been described. These studies were critically introduced referring to the informational value of those simulations. In summary, biosensors will be illustrated as an innovative and promising, although not yet comprehensively applied, technique in the antibacterial field.

Sensors, Vol. 11, Pages 9442-9449: Self-Assembled Films of Dendrimers and Metallophthalocyanines as FET-Based Glucose Biosensors

Posted: 03 Oct 2011 12:00 AM PDT

Separative extended gate field effect transistor (SEGFET) type devices have been used as an ion sensor or biosensor as an alternative to traditional ion sensitive field effect transistors (ISFETs) due to their robustness, ease of fabrication, low cost and possibility of FET isolation from the chemical environment. The layer-by-layer technique allows the combination of different materials with suitable properties for enzyme immobilization on simple platforms such as the extended gate of SEGFET devices enabling the fabrication of biosensors. Here, glucose biosensors based on dendrimers and metallophthalocyanines (MPcs) in the form of layer-by-layer (LbL) films, assembled on indium tin oxide (ITO) as separative extended gate material, has been produced. NH3+ groups in the dendrimer allow electrostatic interactions or covalent bonds with the enzyme (glucose oxidase). Relevant parameters such as optimum pH, buffer concentration and presence of serum bovine albumin (BSA) in the immobilization process were analyzed. The relationship between the output voltage and glucose concentration shows that upon detection of a specific analyte, the sub-products of the enzymatic reaction change the pH locally, affecting the output signal of the FET transducer. In addition, dendritic layers offer a nanoporous environment, which may be permeable to H+ ions, improving the sensibility as modified electrodes for glucose biosensing.

Sensors, Vol. 11, Pages 9426-9441: Human Thrombin Detection Through a Sandwich Aptamer Microarray: Interaction Analysis in Solution and in Solid Phase

Posted: 03 Oct 2011 12:00 AM PDT

We have developed an aptamer-based microarray for human thrombin detection exploiting two non-overlapping DNA thrombin aptamers recognizing different exosites of the target protein. The 15-mer aptamer (TBA1) binds the fibrinogen-binding site, whereas the 29-mer aptamer (TBA2) binds the heparin binding domain. Extensive analysis on the complex formation between human thrombin and modified aptamers was performed by Electrophoresis Mobility Shift Assay (EMSA), in order to verify in solution whether the chemical modifications introduced would affect aptamers/protein recognition. The validated system was then applied to the aptamer microarray, using the solid phase system devised by the solution studies. Finally, the best procedure for Sandwich Aptamer Microarray (SAM) and the specificity of the sandwich formation for the developed aptasensor for human thrombin were optimized.

Where's My Water?

Posted: 02 Oct 2011 03:23 PM PDT

[Platform: Mobile (iPhone, iPod Touch)] Alligators aren't usually known for being picky about their personal hygiene. Swampy, the smiling 'gator who lives deep in the sewers, is quite a bit different. While the rest of the alligators are tearing up everything in the sewers, Swampy is getting ready to take a bath. It seems his shower isn't quite working, though, so you'll have to suss out the source of the problem. In Where's My Water?, a smartly-built physics-based puzzle game from Disney Mobile, its your job to cut holes in the soil to direct the water down to Swampy's tub. And with things like movable walls, puddles of acid, and lively bits of mold, that's much easier said than done.

Molecules, Vol. 16, Pages 8402-8409: Synthesis of ent-Kaurane Diterpene Monoglycosides

Posted: 03 Oct 2011 12:00 AM PDT

Synthesis of two ent-kaurane diterpene glycosides, steviol 19-O-β-D-glucopyranosiduronic acid (steviol glucuronide, 5), and 13-hydroxy ent-kaur-16-en-19-oic acid-β-D-glucopyranosyl ester (7) has been achieved from a common starting material, steviol, using phase transfer catalyst. Also, synthesis of an additional 17-nor-ent-kaurane glycoside, namely 13-methyl-16-oxo-17-nor-ent-kauran-19-oic acid-β-D-glucopyranosyl ester (10) was performed using the starting material isosteviol and similar synthetic methodology. Synthesis of all three steviol glycosides was performed using straightforward chemistry and their structures were characterized on the basis of 1D and 2D NMR as well as mass spectral (MS) data.

Coatings, Vol. 1, Pages 88-107: In Situ Fabrication of AlN Coating by Reactive Plasma Spraying of Al/AlN Powder

Posted: 03 Oct 2011 12:00 AM PDT

Reactive plasma spraying is a promising technology for the in situ formation of aluminum nitride (AlN) coatings. Recently, it became possible to fabricate cubic-AlN-(c-AlN) based coatings through reactive plasma spraying of Al powder in an ambient atmosphere. However, it was difficult to fabricate a coating with high AlN content and suitable thickness due to the coalescence of the Al particles. In this study, the influence of using AlN additive (h-AlN) to increase the AlN content of the coating and improve the reaction process was investigated. The simple mixing of Al and AlN powders was not suitable for fabricating AlN coatings through reactive plasma spraying. However, it was possible to prepare a homogenously mixed, agglomerated and dispersed Al/AlN mixture (which enabled in-flight interaction between the powder and the surrounding plasma) by wet-mixing in a planetary mill. Increasing the AlN content in the mixture prevented coalescence and increased the nitride content gradually. Using 30 to 40 wt% AlN was sufficient to fabricate a thick (more than 200 µm) AlN coating with high hardness (approximately 1000 Hv). The AlN additive prevented the coalescence of Al metal and enhanced post-deposition nitriding through N2 plasma irradiation by allowing the nitriding species in the plasma to impinge on a larger Al surface area. Using AlN as a feedstock additive was found to be a suitable method for fabricating AlN coatings by reactive plasma spraying. Moreover, the fabricated coatings consist of hexagonal (h-AlN), c-AlN (rock-salt and zinc-blend phases) and certain oxides: aluminum oxynitride (Al5O6N), cubic sphalerite Al23O27N5 (ALON) and Al2O3. The zinc-blend c-AlN and ALON phases were attributed to the transformation of the h-AlN feedstock during the reactive plasma spraying. Thus, the zinc-blend c-AlN and ALON phases were not included in the feedstock and were not formed through nitriding of the Al.
