This study was designed to investigate the distribution of Th17 cells in the tumor microenvironment and peripheral blood of pancreatic cancer patients, its clinical significance, and the expression profile of Th17 cell-associated cytokines. The percentage of Th17 cells detected by flow cytometry analysis (FACS) was significantly higher in 46 pancreatic tumor tissues (5.28 ± 1.65%) compared with corresponding adjacent normal tissues (2.57 ± 0.83%) (P = 0.031). In addition, the percentage of Th17 cells was significantly higher in stage III-IV tumors than stage I-II tumors (P = 0.039). The percentage of Th17 cells in peripheral blood of 20 pancreatic cancer patients (3.99 ± 1.15%) was significantly higher than 15 healthy volunteers (1.98 ± 0.57%) (P = 0.027). Immunohistochemistry (IHC) was performed to detect IL-17+ cells in 46 pancreatic tumor tissues, as well as expression of CD34 in 24 tumor tissues. IL-17 was shown to mainly locate in cytoplasm, and the frequency of IL-17+ cells in tumor tissues (39/46) was higher than control (29/46). The presence of IL-17+ cells in tumor tissues was associated with tumor, node, and metastasis (TNM) stage, and lymph node metastasis (P = 0.012 and P = 0.009) but not with patient sex, age, tumor size, and histological grade (P > 0.05). Interestingly, distribution of Th17 cells in tumor tissues was positively correlated with microvessel density (MVD) (r = 0.86, P = 0.018). Furthermore, the median survival time of patients with high and low level of IL-17+ cells frequency was 14.5 and 18.5 months respectively (P = 0.023). The serum levels of Th17 cell-associated cytokines, IL-17 and IL-23 in 20 pancreatic patients detected by enzyme-linked immunosorbent assay (ELISA) were 69.2 ± 28.5 pg/mL and 266.5 ± 98.1 pg/mL, respectively, which were significantly higher than 15 healthy volunteers (P = 0.015 and P = 0.02). Moreover, levels of IL-17 and IL-23 were significantly higher in stage III-IV tumors than stage I-II tumors (P = 0.04 and P = 0.036). This study suggests that increase in Th17 cells frequency and its related cytokines levels in pancreatic tumor tissues may indicate involvement in the invasion and metastasis of pancreatic cancer, which may thereby affect patient prognosis. Therefore, Th17 cells and related cytokines may be served as important immune indicators for predicting the prognosis of pancreatic cancer patients.

A novel magnetic nanoparticle-based electrochemical immunoassay of carcinoembryonic antigen (CEA) was designed as a model using CEA antibody-functionalized magnetic beads [DNA/Fe3O4/ZrO2; Fe3O4 (core)/ZrO2 (shell) nano particles (ZMPs)] as immunosensing probes. To design the immunoassay, the CEA antibody and O-phenylenediamine (OPD) were initially immobilized on a chitosan/nano gold composite membrane on a glassy carbon electrode (GCE/CS-nano Au), which was used for CEA recognition. Then, horseradish peroxidase (HRP)-labeled anti-CEA antibodies (HRP-CEA Ab2) were bound to the surface of the synthesized magnetic ZMP nanoparticles as signal tag. Thus, the sandwich-type immune complex could be formed between secondary antibody (Ab2) modified DNA/ZMPs nanochains tagged by HRP and GCE/CS-nano Au. Unlike conventional nanoparticle-based electrochemical immunoassays, the recognition elements of this immunoassay included both electron mediators and enzyme labels, which obviously simplifies the electrochemical measurement process. The sandwich-type immunoassay format was used for online formation of the immunocomplex of CEA captured in the detection cell with an external magnet. The electrochemical signals derived from HRP during the reduction of H2O2 with OPD as electron mediator were measured. The method displayed a high sensitivity for CEA detection in the range of 0.008–200 ng/mL, with a detection limit of 5 pg/mL (estimated at a signal-to-noise ratio of 3). The precision, reproducibility, and stability of the immunoassay were good. The use of the assay was evaluated with clinical serum samples, and the results were in excellent accordance with those obtained using the standard enzyme-linked immunosorbent assay (ELISA) method. Thus, the magnetic nanoparticle-based assay format is a promising approach for clinical applications, and it could be further developed for the detection of other biomarkers in cancer diagnosis.

In order to characterize the molecular events in the carcinogenesis of esophageal cancer and to identify biomarkers for the early detection of the disease, matched precancerous and cancerous tissues resected from 34 esophageal cancer patients in Chongqing of southern China were compared for the extent of loss of heterozygosity (LOH). Sixteen microsatellite markers on nine chromosome regions were used for the PCR-based LOH analysis. The overall frequency of LOH at the 16 microsatellite loci was significantly increased as the pathological status of the resection specimens changed from low-grade dysplasia (LGD) to high-grade dysplasia (HGD) and squamous cell carcinoma (SCC) (P < 0.001), indicating that tumorigenesis of the esophageal squamous epithelia is a progressive process involving accumulative changes of LOH. A total of eight markers showed LOH in the LGD samples, suggesting that these loci may be involved in the early-stage tumorigenesis of esophageal squamous cell carcinoma (ESCC) and that LOH analysis at these loci may help improve the early detection of this disease. In addition, heterozygosity was regained at four loci in the SCC samples of four patients compared with the HGD samples, suggesting the possibility of genetic heterogeneity in the tumorigenesis of esophageal cancer.

Data centric storages for sensor networks have been proposed to efficiently process multi-dimensional range queries as well as exact matches. Usually, a sensor network does not process only one type of the query, but processes various types of queries such as range queries, exact matches and skyline queries. Therefore, a sensor network based on a data centric storage for range queries and exact matches should process skyline queries efficiently. However, existing algorithms for skyline queries have not considered the features of data centric storages. Some of the data centric storages store similar data in sensor nodes that are placed on geographically similar locations. Consequently, all data are ordered in a sensor network. In this paper, we propose a new skyline query processing algorithm that exploits the above features of data centric storages.

Margaret Thatcher reimbursed for more than £500K in expenses by UK taxpayers

Margaret Thatcher leads Britain's former prime-ministers in claiming expenses back from the taxpayer. She's been reimbursed more than £500,000 in the past five years.

Figures revealed by the Cabinet Office minister, Francis Maude, in response to a written parliamentary question by the Conservative MP Philip Hollobone, show that Thatcher has received £535,000 from the state since 2006, and John Major, who set up the allowance in 1991, has received £490,000. Tony Blair has claimed since 2007 and received £273,000. The figures reveal he received £169,076 in 2008-9, more than his salary in office.

The public duties cost allowance is administered by the Cabinet Office and claims must be supported by documentary evidence. Thatcher, who has suffered ill health which limits her engagements, still attends some public events, including an address by the Pope in the UK. According to figures released last year the maximum allowance claimable doubled from £47,568 in 1997-98 to £100,205 in 2008-9. Defending the allowance's value for money, Maude said: "The public duties cost allowance is kept under review."

Recently, the range of available Radio Frequency Identification (RFID) tags has been widened to include smart RFID tags which can monitor their varying surroundings. One of the most important factors for better performance of smart RFID system is accurate measurement from various sensors. In the multi-sensing environment, some noisy signals are obtained because of the changing surroundings. We propose in this paper an improved Kalman filter method to reduce noise and obtain correct data. Performance of Kalman filter is determined by a measurement and system noise covariance which are usually called the R and Q variables in the Kalman filter algorithm. Choosing a correct R and Q variable is one of the most important design factors for better performance of the Kalman filter. For this reason, we proposed an improved Kalman filter to advance an ability of noise reduction of the Kalman filter. The measurement noise covariance was only considered because the system architecture is simple and can be adjusted by the neural network. With this method, more accurate data can be obtained with smart RFID tags. In a simulation the proposed improved Kalman filter has 40.1%, 60.4% and 87.5% less Mean Squared Error (MSE) than the conventional Kalman filter method for a temperature sensor, humidity sensor and oxygen sensor, respectively. The performance of the proposed method was also verified with some experiments.

In order to perform sensing tasks, most current Intelligent Transportation Systems (ITS) rely on expensive sensors, which offer only limited functionality. A more recent trend consists of using Wireless Sensor Networks (WSN) for such purpose, which reduces the required investment and enables the development of new collaborative and intelligent applications that further contribute to improve both driving safety and traffic efficiency. This paper surveys the application of WSNs to such ITS scenarios, tackling the main issues that may arise when developing these systems. The paper is divided into sections which address different matters including vehicle detection and classification as well as the selection of appropriate communication protocols, network architecture, topology and some important design parameters. In addition, in line with the multiplicity of different technologies that take part in ITS, it does not consider WSNs just as stand-alone systems, but also as key components of heterogeneous systems cooperating along with other technologies employed in vehicular scenarios.

Mobile autonomous systems are very important for marine scientific investigation and military applications. Many algorithms have been studied to deal with the computational efficiency problem required for large scale Simultaneous Localization and Mapping (SLAM) and its related accuracy and consistency. Among these methods, submap-based SLAM is a more effective one. By combining the strength of two popular mapping algorithms, the Rao-Blackwellised particle filter (RBPF) and extended information filter (EIF), this paper presents a Combined SLAM—an efficient submap-based solution to the SLAM problem in a large scale environment. RBPF-SLAM is used to produce local maps, which are periodically fused into an EIF-SLAM algorithm. RBPF-SLAM can avoid linearization of the robot model during operating and provide a robust data association, while EIF-SLAM can improve the whole computational speed, and avoid the tendency of RBPF-SLAM to be over-confident. In order to further improve the computational speed in a real time environment, a binary-tree-based decision-making strategy is introduced. Simulation experiments show that the proposed Combined SLAM algorithm significantly outperforms currently existing algorithms in terms of accuracy and consistency, as well as the computing efficiency. Finally, the Combined SLAM algorithm is experimentally validated in a real environment by using the Victoria Park dataset.

Assassin's Creed: Revelations Updated Hands-On Preview

We make time our plaything, take the scenic view of the city, and go all detective to uncover the past in our latest hands-on.

 

Get ready to do the time warp with Assassin's Creed: Revelations. While the game may appear to be more of the same rooftop acrobatics, hidden blade stabbery, and subterfuge that has made the series a hit three times over, things are going to become even more Inception-inspired, as this time Desmond, controlling Ezio, spends his time locating the five keys to unlock Altair's Library of Masyaf. Successfully tracking down these remnants allows Ezio to travel through time and relive pivotal moments in Altair's life from age 26 all the way past 60 years. Warning, preview contains some potential storyline spoilers.

Our previous peeks at the game have showcased some of the new item creation that is possible, such as bombcrafting, which allows you to turn three rudimentary, found items into (among others) proximity mines, smoke grenades, or bombs filled with gold coins to create impromptu diversions. We've also been given a taste of the new tower defence mode that will crop up at intervals during the narrative, providing you with the chance to hang up your wall-climbing boots for a while and sit for a spell. As you play General, you will place archers, gunners, and Brotherhood leaders on rooftops, build upgradable barricades, and call in mortar-style cannon attacks to demolish aggressors.

In our most recent hands-on time with the game, we took both Ezio and Altair for a jog.

After successfully locating and securing one of the five keys used to find the lost library of Altair Ibn La'Ahad, we were transported back to the Crusades to wrap up some unfinished business. 26-year-old Altair had just slain Al Mualim, Grand Master of the Syrian Assassin Brotherhood, for his betrayal, corrupted by the apple at the centre of the original game. Abbas was the only onlooker to express concern with what had occurred, but made no attempt to stop us as we picked up and cradled the leader's body, carrying it down a grassy slope towards a prepared funeral pyre. As the flames licked at the sky, Abbas returned, pushing us off the raised area towards the crowd below. Because crowd surfing wouldn't be invented for another 600-odd years (Thanks, Benjamin Franklin!) we hit the ground, and watched as our attacker took possession of the apple, fleeing with it to a nearby tower.

As power is known to do, it quickly did its magic on Abbas, putting a vice-like grip on him and radiating energy that knocked us backwards, loosening our hand holds as we attempted to climb scaffolding to retrieve the artefact. Once we were successfully at the top we found him, begging for forgiveness with outstretched hands. We retook control without delay.

Back in the Ezio timeline, Tarik, leader of the Janissaries, had been wheeling and dealing. With no faith in Prince Suleiman, heir to the throne, he bided his time, preparing an army to attack Constantinople. Our job was to infiltrate the arsenal area located in the Forum of Ox with a hope of finding out exactly what sort of forces were being amassed. The amount of armour and training provided to the Janissary guards made them tough targets to take on solo at the best of times. For this reason, we opted for a slightly more deceptive approach, colluding and bribing leaders in the marketplace to stir up a dash of civilian unrest. With guards' backs turned to us as rioters trashed the place, we were able to choose our moment to strike, plunging swords and wrist blades into them from behind to take them down more easily. Granted, it wasn't all that honourable, but it was effective. While the crowd managed to hold its own reasonably well against the better-equipped forces, you won't be able to sit idly by and let them do all the work. Sacrificing too many civilian lives will cause you to fail the event.

With slain guards swimming in pools of their own blood, we slipped through the gates of the arsenal. Inside, we used Ezio's upgraded Eagle Sense ability to locate and stalk Manuel's slimy ethereal trail, which had been left painted to the cobblestones underfoot. Finally inside the building, we opened up large wooden crates where muskets were being stored. Unfortunately, even pro assassins aren't always as stealthy as they think they are, and we were spotted in the act, forcing us to run rather than stay and fight. Ezio's free running had us leaping over gaps between suspended wooden slats, scaling the edges of a ship and using the game's many conveniently placed ziplines and the beneficial forces of gravity to zoom to safety, ditching our less-nimble pursuers in the process.

With a clean set of heels, we found and entered an underground cavern filled with a maze of damaged walkways and perilous falls. As we snaked our way through the finger-tip-sized grabs using the prowess of a spider monkey, we came across a group of armed guards loading barrels of gunpowder into a boat. A free-run and some gymnastics later, we got a look at the sequence's multiple available paths. Rather than failing you and requiring a restart for mistiming a jump or for grabbing instead of maintaining momentum to swing, the action stayed fast and frenetic, pushing us down other avenues. Feet barely touched the small pillars of wood poking out of dark, swirling water before we were off again. The scene ended with a zipline execution, zooming above our targets before plunging blades into skulls as we landed with a thud. The boat was destroyed in the action, but we made it to dry land, and came across another of the pieces of Altair's memories.

Now much older, his face wearing the creases of a life hard fought, Altair sought an explanation for the death of one of his sons. His wife Maria pleaded for him not to seek a violent resolution, and as we approached a looming tower, we were stopped by old pal, Abass, and one of his cronies. Abass explained with a venom-tipped tongue that he was the one who had called for the murder, and before you could say "peaceful adult discussion", blades were unsheathed, and Maria was stabbed and killed in the conflict.

With no time to mourn, we needed to locate and protect Altair's living son. As we hurried out of the city on the escort quest, blades swished with attackers, dashes were made down hills, and the pair of us leapt from building to building with ease. Capable of defending himself, against multiple simultaneous attackers, it was clear that he was his father's son.

Everyone has a few skeletons in the closet, and with three concurrent timelines intersecting to cover The Crusades, late 15th-century Renaissance, and present day, expect this to be a significant adventure of discovery. Xbox 360 and PlayStation 3 versions of Assassin's Creed: Revelations will launch globally on November 15, while the PC release is planned for December this year. Stay tuned for our full review soon.

The crystal structures of four homologous series of rod-like molecules are reviewed, two of which form hydrogen bonds and two with a symmetric chemical constitution. Many of the compounds investigated turn into liquid crystalline phases upon temperature increase. It is of valuable interest to know possible conformations and possible packing arrangements as prerequisites to model liquid crystalline structures. The hydrogen bonds of homologous series of pure 4-(ω-hydroxyalkyloxy)-4′-hydroxybiphenyl (HnHBP, n the alkyloxy tail length) are realized through head to tail arrangements of the hydroxyl groups and crystallize except one compound in chiral space groups without the molecules containing any asymmetric carbon. The hydrogen bonds of the homologous series of 4-substituted benzoic acids with various lengths of the tail provide dimers through strong polar bonding of adjacent carboxyl groups and thus provide the stiff part of a mesogenic unit prerequisite for liquid crystalline phases. The homologous series of dialkanoyloxybiphenyls (BP-n, n = 1, 19), of which nine compounds could be crystallized, show liquid crystalline behavior for longer alkane chain lengths, despite the high mobility of the alkane chain ends already detectable in the crystal phase. A single molecule, half a molecule or two half molecules form the asymmetric unit in a centrosymmetric space group. The homologous series of 1,4-terephthalidene-bis-N-(4′-n-alkylaniline) (TBAA-n) exhibit a large variety of packing arrangements in the crystalline state, with or without relying on the symmetry center within the molecules.
