Molecules, Vol. 16, Pages 8874-8893: Autotoxicity and Allelopathy of 3,4-Dihydroxyacetophenone Isolated from Picea schrenkiana Needles

Posted: 24 Oct 2011 12:00 AM PDT

Bioassay-guided fractionation of the diethyl ether fraction of a water extract of Picea schrenkiana needles led to the isolation of the phenolic compound 3,4-dihydroxy- acetophenone (DHAP). The allelopathic effects of DHAP were evaluated under laboratory conditions on P. schrenkiana, rice (Oryza sativa L.), wheat (Triticum aestivum L.), radish (Raphanus sativus L.), lettuce (Latuca sativa L.), cucumber (Cucumis sativus L.) and mung bean (Phaseolus radiatus L.). DHAP significantly inhibited seed germination and seedling growth of P. schrenkiana at concentrations of 2.5 mM and 0.5 mM (p < 0.05). Soil analysis revealed that P. schrenkiana forest soils contained exceptionally high DHAP concentrations (mean = 0.51 ± 0.03 mg/g dry soil), sufficient to inhibit natural P. schrenkiana recruitment. DHAP also exhibited strong allelopathic potential. It significantly inhibited wheat and lettuce seed germination at concentrations of 1 mM and 0.5 mM (p < 0.05). The active compound also completely inhibited root growth of the six test species at high concentrations. Our results suggest a dual role of DHAP, both as an allelochemical and as an autotoxicant. The potential for a single plant needle-leached compound to influence both inter- and intra-specific interactions emphasized the complex effects that plant secondary metabolites might have on plant population and community structure.

Pharmaceuticals, Vol. 4, Pages 1381-1399: Lipothioureas as Lipids for Gene Transfection: A Review

Posted: 24 Oct 2011 12:00 AM PDT

Non-viral gene therapy requires innovative strategies to achieve higher transfection efficacy. A few years ago, our group proposed bioinspired lipids whoseinteraction with DNA was not based on ionic interactions, but on hydrogen bonds. We thusdeveloped lipids bearing a thiourea head which allowed an interaction with DNAphosphates through hydrogen bonds. After a proof of concept with a lipid bearing threethiourea functions, a molecular and cellular screening was performed by varying all partsof the lipids: the hydrophobic anchor, the spacer, the linker, and the thiourea head. Twolipothiourea-based structures were identified as highly efficient in vitro transfecting agents. The lipothioureas were shown to reduce non specific interactions with cell membranes anddeliver their DNA content intracellularly more efficiently, as compared to cationiclipoplexes. These lipids could deliver siRNA efficiently and allowed specific cell targetingin vitro. In vivo, thiourea lipoplexes presented a longer retention time in the blood and lessaccumulation in the lungs after an intravenous injection in mice. They also inducedluciferase gene expression in muscle and tumor after local administration in mice. Therefore, these novel lipoplexes represent an excellent alternative to cationic lipoplexes astransfecting agents. In this review we will focus on the structure activity studies thatpermitted the identification of the two most efficient thiourea lipids.

IJERPH, Vol. 8, Pages 4082-4101: Prospective Power Calculations for the Four Lab Study of A Multigenerational Reproductive/Developmental Toxicity Rodent Bioassay Using A Complex Mixture of Disinfection By-Products in the Low-Response Region

Posted: 24 Oct 2011 12:00 AM PDT

In complex mixture toxicology, there is growing emphasis on testing environmentally representative doses that improve the relevance of results for health risk assessment, but are typically much lower than those used in traditional toxicology studies. Traditional experimental designs with typical sample sizes may have insufficient statistical power to detect effects caused by environmentally relevant doses. Proper study design, with adequate statistical power, is critical to ensuring that experimental results are useful for environmental health risk assessment. Studies with environmentally realistic complex mixtures have practical constraints on sample concentration factor and sample volume as well as the number of animals that can be accommodated. This article describes methodology for calculation of statistical power for non-independent observations for a multigenerational rodent reproductive/developmental bioassay. The use of the methodology is illustrated using the U.S. EPA's Four Lab study in which rodents were exposed to chlorinated water concentrates containing complex mixtures of drinking water disinfection by-products. Possible experimental designs included two single-block designs and a two-block design. Considering the possible study designs and constraints, a design of two blocks of 100 females with a 40:60 ratio of control:treated animals and a significance level of 0.05 yielded maximum prospective power (~90%) to detect pup weight decreases, while providing the most power to detect increased prenatal loss.

Cancers, Vol. 3, Pages 3957-3971: Emerging Evidence for MicroRNAs as Regulators of Cancer Stem Cells

Posted: 24 Oct 2011 12:00 AM PDT

Cancer stem cells are defined as a subpopulation of cells within a tumor that are capable of self-renewal and differentiation into the heterogeneous cell lineages that comprise the tumor. Many studies indicate that cancer stem cells may be responsible for treatment failure and relapse in cancer patients. The factors that regulate cancer stem cells are not well defined. MicroRNAs (miRNAs) are small non-coding RNAs that regulate translational repression and transcript degradation. miRNAs play a critical role in embryonic and inducible pluripotent stem cell regulation and emerging evidence supports their role in cancer stem cell evolution. To date, miRNAs have been shown to act either as tumor suppressor genes or oncogenes in driving critical gene expression pathways in cancer stem cells in a wide range of human malignancies, including hematopoietic and epithelial tumors and sarcomas. miRNAs involved in cancer stem cell regulation provide attractive, novel therapeutic targets for cancer treatment. This review attempts to summarize progress to date in defining the role of miRNAs in cancer stem cells.

Quicksilver 61 - Application launcher and much more.. (Free)

Posted: 23 Oct 2011 08:59 PM PDT

Quicksilver is a unified, extensible interface for working with applications, contacts, music, and other data.

Anyone using a computer (and the Macintosh is no exception) spends a lot of time locating files and applications, and then carrying out actions on those items. The Finder is a reasonably good tool for getting a visual overview of the contents of your disk, but it isn't ideal for quickly locating and manipulating files, especially if you have a deep and complicated folder structure, or if you prefer keyboard navigation to using the mouse.

Quicksilver allows you to find what you need quickly and easily, while keeping your hands on the keyboard. For example, if you want to launch an application hidden in the depths of your file system, simply activate Quicksilver with a keystroke, type a few letters of the application's name, then hit Return or Enter to launch it. When you don't need Quicksilver, it keeps out of your way, preserving precious screen real estate, and yet you can summon it instantly with a key stroke - just like Mercury the winged messenger. Actually, Roman mythological texts are remarkably silent on whether Mercury could be summoned with a key stroke, but it would have been great if he could. Quicksilver will certainly make you feel as if you have god-like powers.

Quicksilver indexes and parses selected parts of the contents of your hard drive in the Catalog, so that it locates and displays the items matching your search really quickly. If you are a programmer or an uber-geek, you will be glad to know that you can extend and customise Quicksilver until it does exactly what you want it to. The modular structure also means that the application will never be bloated: You only load the modules that you find useful

An older, 10.3 compatible version is available: http://getqs.blacktree.com/b36/

This PSP game is the first to use two UMDs.

After that is Kirby's Return to Dreamland on the Wii. The game is a return to form for Kirby's platforming and power-stealing shenanigans. This title will also feature four-player simultaneous co-op with other Kirby clones and new copy abilities for players to experiment with.

Finally, fans of inFamous 2 can download the latest stand-alone title called inFamous 2: Festival of Blood. This side story is a "what if" scenario where main character Cole is a vampire and has to fight off other vampires in New Marais. The game features new powers and new user-generated content tools to complement the feature introduced in inFamous 2. The downloadable title will be out this Tuesday; gamers do not need a copy of inFamous 2 to play this.
