IJMS, Vol. 13, Pages 5542-5553: A Practical Chemo-enzymatic Approach to Highly Enantio-Enriched 10-Ethyl-7,8-dihydro-?-ionone Isomers: A Method for the Synthesis of 4,5-Didehydro-a-Ionone

Posted: 09 May 2012 12:00 AM PDT

An efficient and convenient strategy for the enantioselective synthesis of enantiomerically enriched 10-ethyl-7,8-dihydro-γ-ionone isomers (R)-(+)-7, and (S)-(−)-7 are described utilizing a lipase mediated resolution protocol, and reductive elimination of the secondary allylic alcohol as the key step. The enantioselective and diastereoselective lipase kinetic acetylation of 4-hydroxy-γ-ionone derivatives 6a afforded the 4-acetyl-γ-ionone derivatives (−)-8, and the 4-hydrox-γ-ionone derivatives (+)-6a, which are suitable precursors of the desired products. Stereospecific palladium-mediated elimination of allylic acetate provides the target compounds with an excellent enantiomeric excess and yield. Additionally, the novel 4,5-didehydro-α-ionone 13 is obtained from readily prepared (2,6,6-trimethylcyclohexa-2,4-dien-1-yl) methanol 9. The structures of all newly synthesized compounds have been elucidated by 1H, 13C NMR, GC-MS, and IR spectrometry. These compounds represent a new class of odorants that may be of pivotal relevance in industrial perfumery.

IJMS, Vol. 13, Pages 5528-5541: Early Fixation of Cobalt-Chromium Based Alloy Surgical Implants to Bone Using a Tissue-engineering Approach

Posted: 09 May 2012 12:00 AM PDT

To establish the methods of demonstrating early fixation of metal implants to bone, one side of a Cobalt-Chromium (CoCr) based alloy implant surface was seeded with rabbit marrow mesenchymal cells and the other side was left unseeded. The mesenchymal cells were further cultured in the presence of ascorbic acid, β-glycerophosphate and dexamethasone, resulting in the appearance of osteoblasts and bone matrix on the implant surface. Thus, we succeeded in generating tissue-engineered bone on one side of the CoCr implant. The CoCr implants were then implanted in rabbit bone defects. Three weeks after the implantation, evaluations of mechanical test, undecalcified histological section and electron microscope analysis were performed. Histological and electron microscope images of the tissue engineered surface exhibited abundant new bone formation. However, newly formed bone tissue was difficult to detect on the side without cell seeding. In the mechanical test, the mean values of pull-out forces were 77.15 N and 44.94 N for the tissue-engineered and non-cell-seeded surfaces, respectively. These findings indicate early bone fixation of the tissue-engineered CoCr surface just three weeks after implantation.

Humanities, Vol. 1, Pages 80-103: Humanity’s Bioregional Places: Linking Space, Aesthetics, and the Ethics of Reinhabitation

Posted: 09 May 2012 12:00 AM PDT

Originally theorized as a radical environmental movement, bioregionalism connects humanity to the specificities of a place. To establish greater cohesion between environments and cultures, bioregionalism endeavors to integrate societal activities and the nuances of natural spaces known as bioregions. The criticism of bioregionalism, however, pertains to the shortcomings of circumscribing culture within ecological boundaries. In light of its criticism, bioregionalism can strengthen its theoretical basis and its potential for cultural change by engaging critically with space, aesthetics, and ethics. This engagement first involves the recognition of bioregionalism as an ethical possibility based on the fundamental spatial unit of the watershed. A watershed comprises vital regional ecological processes, bearing discrete aesthetic properties and patterns. Through the sensuous possibilities of watersheds, a bioregional aesthetic can be integrated with an ethic of reinhabitation. The relation between space, aesthetics, and ethics gives form to and sustains the experience of place, which is intrinsically related to promoting the awareness of ecological sustainability.

Sensors, Vol. 12, Pages 5966-5977: Fusion-Triggered Switching of Enzymatic Activity on an Artificial Cell Membrane

Posted: 09 May 2012 12:00 AM PDT

A nanosensory membrane device was constructed for detecting liposome fusion through changes in an enzymatic activity. Inspired by a biological signal transduction system, the device design involved functionalized liposomal membranes prepared by self-assembly of the following molecular components: a synthetic peptide lipid and a phospholipid as matrix membrane components, a Schiff's base of pyridoxal 5'-phosphate with phosphatidylethanolamine as a thermo-responsive artificial receptor, NADH-dependent L-lactate dehydrogenase as a signal amplifier, and Cu2+ ion as a signal mediator between the receptor and enzyme. The enzymatic activity of the membrane device was adjustable by changing the matrix lipid composition, reflecting the thermotropic phase transition behavior of the lipid membranes, which in turn controlled receptor binding affinity toward the enzyme-inhibiting mediator species. When an effective fusogen anionic polymer was added to these cationic liposomes, membrane fusion occurred, and the functionalized liposomal membranes responded with changes in enzymatic activity, thus serving as an effective nanosensory device for liposome fusion detection.

Buildings, Vol. 2, Pages 153-172: Impact of Vibration Time on Compressive Strength of Hardened Sandcrete Building Blocks

Posted: 09 May 2012 12:00 AM PDT

The majority of the sandcrete blocks used in the Nigerian building industry fall short of the minimum specification standards. There is evidence to suggest a wide variation in compressive strength from one block manufacturer to another and also within block samples from a single source. This problem has been attributed to poor quality control and substandard constituent materials. Also very alarming is the ignorance surrounding the usage and engineering properties of some of the widely used fine aggregate deposits. As a way forward, this paper aims to re-establish the impact of vibration time in sandcrete block production using six fine aggregate deposits found within Benin City (Midwestern Nigeria) and their various pair combinations. Some of the basic properties like silt content, grading parameters—co-efficient of uniformity (Cu), curvature co-efficient (Cc) and the fineness modulus (Fm)—of these fine aggregates were established by laboratory means. In addition, the wet and dry compressive strength of these sandcrete blocks made from these sands were established. A total of 1,080 block samples produced under very controlled conditions were used in this investigation. It was revealed that the utility value of sand can be improved when the weaker and commonly used sands were combined with those that are better, more expensive and less frequently used at different vibration periods and ratios. Findings further revealed that sand types and the sand combination approach adopted were very significant to grading parameters and strength; at a much higher vibration time the compressive strength and durability properties were also considerably improved.

Marine Drugs, Vol. 10, Pages 1044-1065: Accumulation, Biotransformation, Histopathology and Paralysis in the Pacific Calico Scallop Argopecten ventricosus by the Paralyzing Toxins of the Dinoflagellate Gymnodinium catenatum

Posted: 09 May 2012 12:00 AM PDT

The dinoflagellate Gymnodinium catenatum produces paralyzing shellfish poisons that are consumed and accumulated by bivalves. We performed short-term feeding experiments to examine ingestion, accumulation, biotransformation, histopathology, and paralysis in the juvenile Pacific calico scallop Argopecten ventricosus that consume this dinoflagellate. Depletion of algal cells was measured in closed systems. Histopathological preparations were microscopically analyzed. Paralysis was observed and the time of recovery recorded. Accumulation and possible biotransformation of toxins were measured by HPLC analysis. Feeding activity in treated scallops showed that scallops produced pseudofeces, ingestion rates decreased at 8 h; approximately 60% of the scallops were paralyzed and melanin production and hemocyte aggregation were observed in several tissues at 15 h. HPLC analysis showed that the only toxins present in the dinoflagellates and scallops were the N-sulfo-carbamoyl toxins (C1, C2); after hydrolysis, the carbamate toxins (epimers GTX2/3) were present. C1 and C2 toxins were most common in the mantle, followed by the digestive gland and stomach-complex, adductor muscle, kidney and rectum group, and finally, gills. Toxin profiles in scallop tissue were similar to the dinoflagellate; biotransformations were not present in the scallops in this short-term feeding experiment.

Biosensors, Vol. 2, Pages 171-188: Developing a Real Time Sensing System to Monitor Bacteria in Wound Dressings

Posted: 09 May 2012 12:00 AM PDT

Infection control is a key aspect of wound management strategies. Infection results in chemical imbalances and inflammation in the wound and may lead to prolonged healing times and degradation of the wound surface. Frequent changing of wound dressings may result in damage to healing tissues and an increased risk of infection. This paper presents the first results from a monitoring system that is being developed to detect presence and growth of bacteria in real time. It is based on impedance sensors that could be placed at the wound-dressing interface and potentially monitor bacterial growth in real time. As wounds can produce large volumes of exudate, the initial system reported here was developed to test for the presence of bacteria in suspension. Impedance was measured using disposable silver-silver chloride electrodes. The bacteria Staphylococcus aureus were chosen for the study as a species commonly isolated from wounds. The growth of bacteria was confirmed by plate counting methods and the impedance data were analysed for discernible differences in the impedance profiles to distinguish the absence and/or presence of bacteria. The main findings were that the impedance profiles obtained by silver-silver chloride sensors in bacterial suspensions could detect the presence of high cell densities. However, the presence of the silver-silver chloride electrodes tended to inhibit the growth of bacteria. These results indicate that there is potential to create a real time infection monitor for wounds based upon impedance sensing.
