Molecules, Vol. 17, Pages 3475-3483: Synthesis and Antimicrobial Activity of N′-Heteroarylidene-1-adamantylcarbohydrazides and (±)-2-(1-Adamantyl)-4-acetyl-5-[5-(4-substituted phenyl-3-isoxazolyl)]-1,3,4-oxadiazolines

The reaction of adamantane-1-carbohydrazide (1) with heterocyclic aldehydes, namely 5-(4-chlorophenyl)isoxazole-3-carboxaldehyde (2a), 5-(4-methylphenyl)isoxazole-3-carboxaldehyde (2b), 5-(4-methoxyphenyl)isoxazole-3-carboxaldehyde (2c), 1H-imidazole-2-carboxaldehyde and 2-butyl-4-chloro-1H-imidazole-5-carboxaldehyde, in ethanol, yielded the corresponding N′-heteroarylidene-1-adamantylcarbohydrazides 3a, 3b, 3c, 4 and 5, respectively, in good yields. The 4-acetyl-1,3,4-oxadiazoline analogues 6a‑c were prepared in 48–55% yields by heating their corresponding N′-heteroarylidene-1-adamantylcarbohydrazides 3a–c with acetic anhydride for two hours. Compounds 3a–c, 4, 5 and 6a–c were tested for in vitro activities against a panel of Gram-positive and Gram-negative bacteria and the yeast-like pathogenic fungus Candida albicans. Compounds 4 and 5 displayed potent broad-spectrum antimicrobial activity, while compounds 3a–c showed good activity against the Gram-positive bacteria.

Experimental design method was used for HPLC determination of irbesartan and hydrochlorothiazide in combined dosage forms. The traditional approach for optimization of experiments is time-consuming, involves a large number of runs and does not allow establishing the multiple interacting parameters. The main advantages of the experimental design method include the simultaneous screening of a larger number of factors affecting response and the estimation of possible interactions. On the basis of preliminary experiments, three factors-independent variables were selected as inputs (methanol content, pH of the mobile phase and temperature) and as dependent variables, five responses (resolution, symmetry of irbesartan peak, symmetry of hydrochlorothiazide peak, retention factor of irbesartan and retention factor of hydrochlorothiazide) were chosen. A full 23 factorial design, where factors were examined at two different levels ("low" and "high") was used to determine which factors had an effect on the studied response. Afterwards, experimental design was used to optimize these influent parameters in the previously selected experimental domain. The novelty of our method lies in the optimization step accomplished by Derringer¢s desirability function. After optimizing the experimental conditions a separation was conducted on a Supelcosil C18 (150 mm × 4.6 mm, 5 mm particle size) column with a mobile phase consisting of methanol-tetrahydrofuran-acetate buffer 47:10:43 v/v/v, pH 6.5 and a column temperature of 25 °C. The developed method was successfully applied to the simultaneous separation of these drug-active compounds in their commercial pharmaceutical dosage forms.

IJMS, Vol. 13, Pages 3598-3617: Improvement of Carbon Tetrachloride-Induced Acute Hepatic Failure by Transplantation of Induced Pluripotent Stem Cells without Reprogramming Factor c-Myc

The only curative treatment for hepatic failure is liver transplantation. Unfortunately, this treatment has several major limitations, as for example donor organ shortage. A previous report demonstrated that transplantation of induced pluripotent stem cells without reprogramming factor c-Myc (3-genes iPSCs) attenuates thioacetamide-induced hepatic failure with minimal incidence of tumorigenicity. In this study, we investigated whether 3-genes iPSC transplantation is capable of rescuing carbon tetrachloride (CCl4)-induced fulminant hepatic failure and hepatic encephalopathy in mice. Firstly, we demonstrated that 3-genes iPSCs possess the capacity to differentiate into hepatocyte-like cells (iPSC-Heps) that exhibit biological functions and express various hepatic specific markers. 3-genes iPSCs also exhibited several antioxidant enzymes that prevented CCl4-induced reactive oxygen species production and cell death. Intraperitoneal transplantation of either 3-genes iPSCs or 3-genes iPSC-Heps significantly reduced hepatic necrotic areas, improved hepatic functions, and survival rate in CCl4-treated mice. CCl4-induced hepatic encephalopathy was also improved by 3-genes iPSC transplantation. Hoechst staining confirmed the successful engraftment of both 3-genes iPSCs and 3-genes iPSC-Heps, indicating the homing properties of these cells. The most pronounced hepatoprotective effect of iPSCs appeared to originate from the highest antioxidant activity of 3-gene iPSCs among all transplanted cells. In summary, our findings demonstrated that 3-genes iPSCs serve as an available cell source for the treatment of an experimental model of acute liver diseases.

Radix Angelica Sinensis (RAS) and Rhizome Ligusticum (RLC) combination is a popular herb pair commonly used in clinics for treatment of blood deficiency syndrome in China. The aim of this study is to compare the pharmacokinetic properties of ferulic acid (FA), a main bioactive constituent in both RAS and RLC, between normal and blood deficiency syndrome animals, and to investigate the influence of compatibility of RAS and RLC on the pharmacokinetic of FA. The blood deficiency rats were induced by injecting 2% Acetyl phenylhydrazine (APH) on the first day, every other day, to a total of five times, at the dosage of 100, 50, 50, 30, 30 mg/kg body mass, respectively. Quantification of FA in rat plasma was achieved by using a simple and rapid HPLC method. Plasma samples were collected at different time points to construct pharmacokinetic profiles by plotting drug concentration versus time, and estimate pharmacokinetic parameters. Between normal and blood deficiency model groups, both AUC(0–t) and Cmax of FA in blood deficiency rats after RAS-RLC extract administration increased significantly (P < 0.05), while clearance (CL) decreased significantly. Among three blood deficiency model groups, t1/2α, Vd, AUC(0–t) and AUC(0–∞) all increased significantly in the RAS-RLC extract group compared with the RAS group. The results indicated that FA was absorbed better and eliminated slower in blood deficiency rats; RLC could significantly prolong the half-life of distribution, increase the volume of distribution and the absorption amount of FA of RAS in blood deficiency rats, which may be due to the synergic action when RAS and RLC were used together to treat blood deficiency syndrome.

Autophagy is a highly regulated intracellular process involved in the turnover of most cellular constituents and in the maintenance of cellular homeostasis. In this study, we show that the activity of autophagy increases in H2O2 or RasV12-induced senescent fibroblasts. Inhibiting autophagy promotes cell apoptosis in senescent cells, suggesting that autophagy activation plays a cytoprotective role. Furthermore, our data indicate that the increase of autophagy in senescent cells is linked to the activation of transcription factor FoxO3A, which blocks ATP generation by transcriptionally up-regulating the expression of PDK4, an inhibitor of pyruvate dehydrogenase complex, thus leading to AMPK activation and mTOR inhibition. These findings suggest a novel mechanism by which FoxO3A factors can activate autophagy via metabolic alteration.

This first part of the study introduces an elementary concept of information. Our interest for newness, our curiosity in the new, will be considered as a main building block of information, and of reality itself. A typical definition of information (the reduction of uncertainty) needs to be fundamentally inverted: Information is a compositional activity, including the inconsistent, the paradox, the contradiction and the incoherent meaning. This study expands on the analysis of the composition of new structure (new macrophysical laws), and the analysis of the causality and causal state of such structures ("causally active symbols"). The classical, scientific-objective, passive understanding of information gives meaning to the fact that modern information technology does not by itself lead to an increase of human values. However, our social and moral stance is an informational one, and our informational, active conscious process holds the power to mediate and to enforce this process towards an enriched life. The indicator for such enrichment is given to us by information, and the knowledge about this process will feed us with energy to move towards an active spirit of ethics, and towards the information society. Part I of this study expands on the fundament basis and on our intrinsic responsibility to release the forces that are based on the active dimension of information. Those forces are required in order to reveal the so-called information society from its metaphorical character (Part II).

Forests, Vol. 3, Pages 95-113: Five Year Field Evaluation of Prosopis alba Clones on pH 9–10 Soils in Argentina Selected for Growth in the Greenhouse at Seawater Salinities (45 dS m−1)

Prosopis alba seedlings, that grew at the 45 dS m−1 salinity level in a previous study of growth and survival of Argentine and Peruvian Prosopis, were propagated by rooting cuttings and established in a seed orchard/long term evaluation trial on soils with low salinity (EC 5.1–7.5 dS m−1) but high pH (8.9 to 10.2). A pH gradient occurred in the field with values ranging from pH 9.4 in block 1 to pH 10.3 in block 5. After five years growth, almost all of the clones had a mean height greater than 4 m and one clone was more than 5 m. Ten of the 21 tested clones had significantly greater biomass growth than the three seed propagated check varieties. The broad-sense (i.e., clone mean) heritability was estimated to be 0.45 for biomass, 0.53 for diameter and 0.59 for height indicating that strong genetic gains should be possible by selecting and vegetatively propagating the best genotypes. In the block with the highest pH values, two clones that appear to be P. alba × P. ruscifolia hybrids (i.e., P. vinallilo) had the greatest biomass. Correlations between growth during the last two months in the high salinity hydroponic greenhouse selection system and growth in the field were significant (R2 = 0.262) and positive, although the relationship was negative for putative P. vinallilo clones (R2 = 0.938). The several fold increase in biomass of some of the clones over the three check varieties, suggests that the greenhouse screen was successful in identifying superior salt tolerant clones. Apparently whether the greenhouse seedlings had lesser (~1 cm) to greater (~3 cm) height growth was not as important as just having a healthy live apical meristem. The observed salt tolerance of the putative P. vinalillo clones may prove useful as rootstocks for recently described high pod producing P. alba clones.
