nessViewer offers multi-functionality by combining media player, media center, media server, media editor and preview tool. Its many functions include editing, arrangement and presentation of various media (pictures, movies, DVDs, PDFs). The integrated media center provides direct access to local media and many internet media providers. Switching to a specific media software is no longer necessary.

A main feature of nessViewer is the "Media Show" which allows it to arrange and save playlists for (later) presentations. In addition to the access to iPhoto or local volumes, the media show enables the user to integrate internet media without having to download them (Clipfish, DailyMotion, YouTube and others).

The media center offers a lot of internet media: Arte, BBC, DailyMotion, German television, Hulu, live broadcasting, Miro video podcasts, Spiegel online, YouTube and others. Of course, local media can be accessed too (Aperature, iPhoto, iTunes, folders and mounted volumes) and EyeTV movies can be started (even without the EyeTV application). Through the functionality of "virtual playlist", plenty of different media can be easily selected and continuously presented afterwards.

Big in Japan April 9-15: Kingdom Hearts, Super Robot Taisen

Namco Bandai and Square Enix's cash cows secure pole positions; Xbox 360 sales lost Kinect Star Wars boost.

 

While last week appeared slow for Japan software and hardware sales, two prominent titles made their debut in the top five spots according to Media Create's latest weekly sales update. Namco Bandai's recent mecha-filled strategy RPG title, Dai-2-Ji Super Robot Taisen Z Saisei-hen, held first place for the week of April 9 to April 15 with 34,403 units sold.

Square Enix's latest Kingdom Hearts title for the 3DS ended up just below the Super Robot Taisen entry, with Kingdom Hearts 3D: Dream Drop Distance selling 20,977 units. For more information on the RPG, check out GameSpot's recent coverage.

Super Mario 3D Land clawed its way back up the list after hovering lower on the top ten. Nintendo's 3DS platformer sold 18,119 units, and was followed by the PSP version of Pro Baseball Spirits 2012 from Konami and Kid Icarus: Uprising from Nintendo. The former sold 17,037 units while the latter for the 3DS moved 15,228 copies.

All hardware sales suffered this week. Sales of Sony's PlayStation Vita handheld dropped to 8,250 units from the previous Media Creates sales update, while the Xbox 360 lost the momentum it gained from Kinect Star Wars, falling to 1,373 units sold, down from the previous period's 3,764 units.

Capcom inviting fan tributes for Street Fighter 25th anniversary

Art and music contributions to be used in "official capacity" for near future.

 

Amidst the recent Street Fighter X Tekken DLC kerfuffle, Capcom recently announced on its community blog that it plans to celebrate Street Fighter's 25th anniversary with the help of its players. The company announced that fans will be able to participate in the upcoming festivities by contributing original art and music from the fighting universe.

Capcom will use these fan submissions in an "official capacity" in the near future, but hasn't yet revealed its own plans for the anniversary. Artists who wish to pay tribute can submit traditional hand-drawn artwork, digital artwork, or a photograph of physical creations such as statues and costumes.

Those interested in creating a musical tribute will need to submit remixes of existing Street Fighter music, or fully original tunes inspired by the series. Entries close at 11:59pm PST on May 4. For more information, head to the art submission and music submission guideline pages.

Capcom's latest release in the long running franchise is the recent Street Fighter X Tekken for Xbox 360 and PS3. Check out GameSpot's coverage of the upcoming Vita version.

Sony unveils new God of War game

Publisher releases teaser trailer for PS3-exclusive God of War: Ascension; title appears to be a prequel; no other details released.

 

Fans of Sony's God of War series have much cause for celebration today with confirmation that Kratos will be returning to the PS3 for one more adventure. Sony released a teaser trailer showcasing the next entry in the franchise, to be titled God of War: Ascension.

While Sony released no other information on the new game, the teaser trailer points to a possible prequel: images of a bloody, shadowed Kratos bound in chains are presented with a voice that speaks of "a time before Kratos became the monster known as the Ghost of Sparta".

In the final moments of the trailer, Kratos breaks free from his chains while a voice proclaims: "A time when he would break his blood oath to an unjust god, a time when he would no longer be bound in blood." The trailer ends with an image of the PS3.

In mystical and religious context, the word "ascension" refers to those who ascend to heaven without dying; the word can also be used to describe spiritual enlightenment.

Sony first hinted at an upcoming teaser campaign for a new God of War game earlier this month, posting an image of Kratos on the official PlayStation Facebook page. In February, retailer BT Games posted a product listing for God of War IV, and a month before that, multimedia composer Timothy Williams indicated he was working on a fourth entry in the franchise.

Last year, job listings at Sony Santa Monica called for people to join the God of War development team to create multiplayer services for the franchise, including matchmaking infrastructure and leaderboards. While the new trailer indicates the title will be headed to the PS3, last month Sony Santa Monica senior director of product development Shannon Studstill said the franchise might be headed to the PlayStation Vita or mobile platforms.

The God of War franchise's last outing was God of War: Ghost of Sparta for the PSP in 2010, whose events took place in-between the original God of War and God of War II. God of War Origins Collection, featuring God of War: Chains of Olympus and Ghost of Sparta, was released in September last year for the PS3 in HD. The last entry in the main series was God of War III, released in 2010 on the PS3, whose story seemingly concluded the main franchise.

Transparent conducting oxides (TCOs) are electrical conductive materials with comparably low absorption of electromagnetic waves within the visible region of the spectrum. They are usually prepared with thin film technologies and used in opto-electrical apparatus such as solar cells, displays, opto-electrical interfaces and circuitries. Here, based on a modern database-system, aspects of up-to-date material selections and applications for transparent conducting oxides are sketched, and references for detailed information are given. As n-type TCOs are of special importance for thin film solar cell production, indium-tin oxide (ITO) and the reasonably priced aluminum-doped zinc oxide (ZnO:Al), are discussed with view on preparation, characterization and special occurrences. For completion, the recently frequently mentioned typical p-type delafossite TCOs are described as well, providing a variety of references, as a detailed discussion is not reasonable within an overview publication.

The employment of monoclonal antibodies (Mabs) to identify disease-associated biomarkers in clinical samples represents the underlying principle for many diagnostic tests. To date, these have been principally developed for protein targets with few reported applications for lipids due to their hydrophobicity and poor immunogenicity. Oxysterols represent a family of lipids implicated in diverse human diseases where Mab-based detection assays could have a profound effect on their utility as clinical biomarkers. These are usually identified in patients' samples by mass- spectrometry based approaches. Here, we describe an antibody phage-library based screening methodology for generating a recombinant monoclonal antibody (RAb) targeting the oxysterol-15-ketocholestane (15-KA), a lipid implicated in multiple sclerosis and Autoimmune Encephalomyelitis (EAE). The antibody is highly specific for 15-KA and shows little or no binding activity for other closely related oxysterols. We employ RAb2E9 to address the controversy over whether 15-KA is a true biomarker for MS/EAE and show that 15-KA is undetectable in serum taken from mice with EAE using antibody based detection methodologies; a finding confirmed by mass-spectrometry analysis. This study demonstrates the technical feasibility of using phage display to isolate highly specific antibodies against poorly immunogenic, small molecule lipids.

IJMS, Vol. 13, Pages 4920-4936: Structural and Functional Characterization of Two Alternative Splicing Variants of Mouse Endothelial Cell-Specific Chemotaxis Regulator (ECSCR)

Endothelial cells (ECs) that line the lumen of blood vessels are important players in blood vessel formation, and EC migration is a key component of the angiogenic process. Thus, identification of genes that are specifically or preferentially expressed in vascular ECs and in-depth understanding of their biological functions may lead to discovery of new therapeutic targets. We have previously reported molecular characterization of human endothelial cell-specific molecule 2 (ECSM2)/endothelial cell-specific chemotaxis regulator (ECSCR). In the present study, we cloned two mouse full-length cDNAs by RT-PCR, which encode two putative ECSCR isoform precursors with considerable homology to the human ECSCR. Nucleotide sequence and exon-intron junction analyses suggested that they are alternative splicing variants (ECSCR isoform-1 and -2), differing from each other in the first and second exons. Quantitative RT-PCR results revealed that isoform-2 is the predominant form, which was most abundant in heart, lung, and muscles, and moderately abundant in uterus and testis. In contrast, the expression of isoform-1 seemed to be more enriched in testis. To further explore their potential cellular functions, we expressed GFP- and FLAG-tagged ECSCR isoforms, respectively, in an ECSCR deficient cell line (HEK293). Interestingly, the actual sizes of either ECSCR-GFP or -FLAG fusion proteins detected by immunoblotting are much larger than their predicted sizes, suggesting that both isoforms are glycoproteins. Fluorescence microscopy revealed that both ECSCR isoforms are localized at the cell surface, which is consistent with the structural prediction. Finally, we performed cell migration assays using mouse endothelial MS1 cells overexpressing GFP alone, isoform-1-GFP, and isoform-2-GFP, respectively. Our results showed that both isoforms significantly inhibited vascular epidermal growth factor (VEGF)-induced cell migration. Taken together, we have provided several lines of experimental evidence that two mouse ECSCR splicing variants/isoform precursors exist. They are differentially expressed in a variety of tissue types and likely involved in modulation of vascular EC migration. We have also defined the gene structure of mouse ECSCR using bioinformatics tools, which provides new information towards a better understanding of alternative splicing of ECSCR.

IJMS, Vol. 13, Pages 4906-4919: Novel Drug Delivery System Based on Docetaxel-Loaded Nanocapsules as a Therapeutic Strategy Against Breast Cancer Cells

In the field of cancer therapy, lipid nanocapsules based on a core-shell structure are promising vehicles for the delivery of hydrophobic drugs such as docetaxel. The main aim of this work was to evaluate whether docetaxel-loaded lipid nanocapsules improved the anti-tumor effect of free docetaxel in breast cancer cells. Three docetaxel-loaded lipid nanocapsules were synthesized by solvent displacement method. Cytotoxic assays were evaluated in breast carcinoma (MCF-7) cells treated by the sulforhodamine B colorimetric method. Cell cycle was studied by flow cytometry and Annexin V-FITC, and apoptosis was evaluated by using propidium iodide assays. The anti-proliferative effect of docetaxel appeared much earlier when the drug was encapsulated in lipid nanoparticles than when it was free. Docetaxel-loaded lipid nanocapsules significantly enhanced the decrease in IC50 rate, and the treated cells evidenced apoptosis and a premature progression of the cell cycle from G(1) to G(2)-M phase. The chemotherapeutic effect of free docetaxel on breast cancer cells is improved by its encapsulation in lipid nanocapsules. This approach has the potential to overcome some major limitations of conventional chemotherapy and may be a promising strategy for future applications in breast cancer therapy.
