DEB025 (alisporivir) is a synthetic cyclosporine with inhibitory activity against human immunodeficiency virus type-1 (HIV-1) and hepatitis C virus (HCV). It binds to cyclophilin A (CypA) and blocks essential functions of CypA in the viral replication cycles of both viruses. DEB025 inhibits clinical HIV-1 isolates in vitro and decreases HIV-1 virus load in the majority of patients. HIV-1 isolates being naturally resistant to DEB025 have been detected in vitro and in nonresponder patients. By sequence analysis of their capsid protein (CA) region, two amino acid polymorphisms that correlated with DEB025 resistance were identified: H87Q and I91N, both located in the CypA-binding loop of the CA protein of HIV-1. The H87Q change was by far more abundant than I91N. Additional polymorphisms in the CypA-binding loop (positions 86, 91 and 96), as well as in the N-terminal loop of CA were detected in resistant isolates and are assumed to contribute to the degree of resistance. These amino acid changes may modulate the conformation of the CypA-binding loop of CA in such a way that binding and/or isomerase function of CypA are no longer necessary for virus replication. The resistant HIV-1 isolates thus are CypA-independent.

The existence of di-glucosylated derivative of T-2 toxin in plant (corn powder) was confirmed for the first time in addition to that of HT-2 toxin. These masked mycotoxins (mycotoxin glucosides) were identified as T-2 toxin-di-glucoside (T2GlcGlc) and HT-2 toxin-di-glucoside (HT2GlcGlc) based on accurate mass measurements of characteristic ions and fragmentation patterns using high-resolution liquid chromatography-Orbitrap mass spectrometric (LC-Orbitrap MS) analysis. Although the absolute structure of T2GlcGlc was not clarified, two glucose molecules were suggested to be conjugated at 3-OH position in tandem when considering the structure of T-2 toxin. On the other hand, the specification of the structure seems to be more complicated in the case of HT2GlcGlc, since HT-2 toxin has two possible positions (at 3-OH and 4-OH) to be glusocylated. In addition, 15-monoacetoxyscirpenol-glucoside (MASGlc) was also detected in the identical sample.

Using 100 years of fire perimeter maps, we investigate the existence of geographical patterns in fire orientation across California. We computed fire perimeter orientation, at the watershed level, using principal component analysis. Circular statistics were used to test for the existence of preferential fire perimeter orientations. Where perimeters displayed preferential orientation, we searched for evidence of orographic channeling by comparing mean fire orientation with watershed orientation. Results show that in California, 49% of the burnt area is associated with watersheds, where fires displayed preferential orientation. From these, 25% of the burnt area is aligned along the NE/SW orientation and 18% in the E/W orientation. In 27 out of 86 watersheds with preferential fire alignment, there is also correspondence between mean fire orientation and watershed orientation. Topographic influence on fire spread and dominant wind patterns during the fire season can account for the consistency in fire perimeter orientation in these regions. Our findings highlight the historical pattern of fire perimeter orientation and identify watersheds with potential orographic channeling.

Agronomy, Vol. 3, Pages 248-255: Sustainable Production of Japanese Eggplants in a Piedmont Soil in Rotation with Winter Cover Crops

Eggplant is a popular vegetable consumed all over the world. Cover cropping is an efficient way of recycling nutrients and reducing inorganic fertilizer requirements to maintain the sustainability of the soil without affecting productivity and profitability. Eggplants (Solanum melongena) (Japanese varieties Hansel and Kamo) were grown in a Piedmont soil with two main treatments, cover crop (CC) and no cover crop (NC), and four sub-fertilizer treatments (T1: 0-0-0, T2: 56-28-112, T3: 84-56-168, and T4: 168-112-224 N-P-K kg/ha), using four replications. The Hansel variety eggplant yield was significantly higher than the Kamo variety. Eggplant yields from CC treatments for both varieties were significantly higher (p &lt; 0.001) than the yields from NC treatments. No significant difference was observed in the yields between T1 and T2 treatments, but the yields from T3 were significantly higher than T1 and T2 and yields from T4 were significantly higher than T3 yields. N released through mineralization of cover crop mixture ranged from 13.33 g/kg at the beginning of the growing season and increased to 18.32 g/kg at the end of the growing season. These results suggest that Japanese eggplants can be successfully grown in the Piedmont area of North Carolina in rotation with cover crops for higher yields.

BioShock Infinite cost $200 million to produce and market – Report

Analysts estimate game development to have cost $100 million; additional $100 million spent on marketing and promotion.

 

According to a New York Times article citing unnamed analysts, BioShock Infinite may have cost its parent company Take-Two Interactive $100 million to develop, with a further $100 million spent to promote and market the game to the public.

BioShock Infinite is the third instalment in the series and has been in development at Irrational Games for four years, with a 200 person team working on the project. The game follows on from 2007's BioShock, and 2010's BioShock 2.

The cost of developing games has risen across the board in recent years. Last month, using a "hypothetical profitability model", Sterne Agee analyst Arvind Bhatia predicted that Grand Theft Auto V–s creation may cost in excess of $137.5 million based on a 250-person team during a five-year development period. An additional $69 million to $109.3 million is expected to be spent in marketing costs to raise awareness for the game.

By comparison, during its six years of production--across four continents and through the hands of more than 800 developers--2011's Star Wars: The Old Republic reportedly cost publisher EA $200 million dollars in development costs alone.

This study compared two methods of assaying the 25-hydroxylated metabolites of cholecalciferol (vitamin D3) and ergocalciferol (vitamin D2). A fully automated electrochemiluminescence assay from Roche Diagnostics and an HPLC based method from Chromsystems were used to measure vitamin D levels in surplus sera from 96 individuals, where the majority has the D2 form of the vitamin. Deming regression, concordance rate, correlation and Altman Bland agreement were performed. Seventy two subjects (75%) had a D2 concentration &gt;10 nmol/L while the remaining twenty four subjects had vitamin D2 concentration of less than 10 nmol/L by HPLC. Overall, the Roche Diagnostics method showed a negative bias of −2.59 ± 4.11 nmol/L on the e602 as compared to the HPLC with a concordance rate of 84%. The concordance rate was 91% in samples with D2 of less than 10 nmol/L and 82% in those with D2 concentration &gt;10 nmol/L. The overall correlation had an r value of 0.77. The r value was higher in samples with D2 levels of less than 10 nmol/L, r = 0.96, as compared to those with D2 values of greater than 10 nmol/L, r = 0.74. The observed bias had little impact on clinical decision and therefore is clinically acceptable.

Several models have been proposed to explain the dark energy that is causing universe expansion to accelerate. Here the acceleration predicted by the Holographic Dark Information Energy (HDIE) model is compared to the acceleration that would be produced by a cosmological constant. While identical to a cosmological constant at low redshifts, z &lt; 1, the HDIE model results in smaller Hubble parameter values at higher redshifts, z &gt; 1, reaching a maximum difference of 2.6 ± 0.5% around z ~ 1.7. The next generation of dark energy measurements, both those scheduled to be made in space (ESA's Euclid and NASA's WFIRST missions) and those to be made on the ground (BigBOSS, LSST and Dark Energy Survey), should be capable of determining whether such a difference exists or not. In addition a computer simulation thought experiment is used to show that the algorithmic entropy of the universe always increases because the extra states produced by the accelerating expansion compensate for the loss of entropy from star formation.

In plasmas, Debye screening structures the possible correlations between particles. We identify a phase space minimum h* in non-equilibrium space plasmas that connects the energy of particles in a Debye sphere to an equivalent wave frequency. In particular, while there is no a priori reason to expect a single value of h* across plasmas, we find a very similar value of h* ≈ (7.5 ± 2.4)×10−22 J·s using four independent methods: (1) Ulysses solar wind measurements, (2) space plasmas that typically reside in stationary states out of thermal equilibrium and spanning a broad range of physical properties, (3) an entropic limit emerging from statistical mechanics, (4) waiting-time distributions of explosive events in space plasmas. Finding a quasi-constant value for the phase space minimum in a variety of different plasmas, similar to the classical Planck constant but 12 orders of magnitude larger may be revealing a new type of quantization in many plasmas and correlated systems more generally.

In this paper we propose an approach to the estimation and simulation of loss distributions based on Maximum Entropy (ME), a non-parametric technique that maximizes the Shannon entropy of the data under moment constraints. Special cases of the ME density correspond to standard distributions; therefore, this methodology is very general as it nests most classical parametric approaches. Sampling the ME distribution is essential in many contexts, such as loss models constructed via compound distributions. Given the difficulties in carrying out exact simulation,we propose an innovative algorithm, obtained by means of an extension of Adaptive Importance Sampling (AIS), for the approximate simulation of the ME distribution. Several numerical experiments confirm that the AIS-based simulation technique works well, and an application to insurance data gives further insights in the usefulness of the method for modelling, estimating and simulating loss distributions.

In this paper, the optimal composition of a paeonol temperature-sensitive in situ gel composed of poloxamer 407 (P407) was determined, and a preliminary study of its effect on allergic rhinitis was performed. The optimal composition of the paeonol temperature-sensitive in situ gel included 2% paeonol inclusion, 22% P407, 2% poloxamer 188 (P188) and 2% PEG6000, as assessed by thermodynamic and rheological studies. The toad palate model was employed to study the toxicity of the paeonol temperature-sensitive in situ gel on the nasal mucosa. The result of this experiment showed low toxicity to cilia, which allows the gel to be used for nasal administration. The Franz diffusion cell method was used to study the in vitro release of paeonol and suggested that the in vitro release was in line with the Higuchi equation. This result suggests that the paeonol could be absorbed into the body through mucous membranes and had some characteristics of a sustained effect. Finally, the guinea pig model of ovalbumin sensitized allergic rhinitis was used to evaluate the preliminary efficacy of the gel, with the paeonol temperature-sensitive in situ gel showing a significant effect on the guinea pig model of sensitized allergic rhinitis (AR).
