| Posted: 20 Aug 2012 12:00 AM PDT The use of methamphetamine (MA) is increasing in the U.S. and elsewhere around the world. MA's capacity to cause addiction significantly exceeds other psychostimulant drugs, and its use negatively impacts learning and memory. Recently, attempts have been made to interfere with the presumed mechanism(s) underlying the establishment of drug-induced memory consolidation. The majority of these studies have employed matrix metalloproteinase (MMP) inhibitors to disrupt MMP-induced extracellular matrix molecule dependent synaptic reconfiguration, or GABA receptor agonists. The present investigation utilized an angiotensin IV (AngIV) analogue, Divalinal-AngIV (divalinal), to disrupt acquisition of MA-induced dependence in rats as measured using the conditioned place preference paradigm. Results indicate that both acute and chronic intracerebroventricular infusion of divalinal prior to each daily subcutaneous injection of MA prevented acquisition. However, divalinal was unable to prevent MA-induced reinstatement after prior acquisition followed by extinction trials. These results indicate that prevention of MA dependence can be accomplished by blockade of the brain AT4 receptor subtype. On the other hand, once MA-induced memory consolidation is in place divalinal appears to be ineffective. Mechanistic studies indicated that divalinal is a potent inhibitor of the hepatocyte growth factor (HGF)/c-Met receptor system, and thus it appears that a functional HGF/c-Met system is required for the acquisition of MA-mediated conditioned place preference. |

| Posted: 20 Aug 2012 12:00 AM PDT Longleaf pine has been classified as very shade intolerant but leaf physiological plasticity to light is not well understood, especially given longleaf pine's persistent seedling grass stage. We examined leaf morphological and physiological responses to light in one-year-old grass-stage seedlings and young trees ranging in height from 4.6 m to 6.3 m to test the hypothesis that young longleaf pine would demonstrate leaf phenotypic plasticity to light environment. Seedlings were grown in a greenhouse under ambient levels of photosynthetically active radiation (PAR) or a 50% reduction in ambient PAR and whole branches of trees were shaded to provide a 50% reduction in ambient PAR. In seedlings, shading reduced leaf mass per unit area (LMA), the light compensation point, and leaf dark respiration (RD), and increased the ratio of light-saturated photosynthesis to RD and chlorophyll b and total chlorophyll expressed per unit leaf dry weight. In trees, shading reduced LMA, increased chlorophyll a, chlorophyll b and total chlorophyll on a leaf dry weight basis, and increased allocation of total foliar nitrogen to chlorophyll nitrogen. Changes in leaf morphological and physiological traits indicate a degree of shade tolerance that may have implications for even and uneven-aged management of longleaf pine. |
| Posted: 20 Aug 2012 12:00 AM PDT A series of pyrido[2,3-d]pyrimidine derivatives were designed and synthesized based on known CC chemokine receptor 4 (CCR4) antagonists. The activities of all the newly synthesized compounds were evaluated using a chemotaxis inhibition assay. Compound 6b was proven to be a potent CCR4 antagonist that can block cell chemotaxis induced by macrophage-derived chemokine (MDC), thymus and activation regulated chemokine (TARC), and CKLF1, the natural ligands of CCR4. In addition, compound 6b is more effective than budesonide in the murine rhinitis model. The intravenous injection LD50 of compound 6b is 175 mg/kg and the oral LD50 is greater than 2,000 mg/kg. |
| Posted: 20 Aug 2012 12:00 AM PDT Many scientific studies have shown that laminarin has anti-tumor effects, but the anti-tumor mechanism was unclear. The purpose of this study was to investigate the effect of laminarin on the induction of apoptosis in human colon cancer LOVO cells and the molecular mechanism involved. LOVO cells were treated with different concentrations of laminarin at different times. Morphology observations were performed to determine the effects of laminarin on apoptosis of LOVO cells. Flow cytometry (FCM) was used to detect the level of intracellular reactive oxygen species (ROS) and pH. Laser scanning confocal microscope (LSCM) was used to analyze intracellular calcium ion concentration, mitochondrion permeability transition pore (MPTP) and mitochondrial membrane potential (MMP). Western blotd were performed to analyze the expressions of Cyt-C, Caspase-9 and -3. The results showed the apoptosis morphology, which showed cell protuberance, concentrated cytoplasm and apoptotic bodies, was obvious after 72 h treatment. Laminarin treatment for 24 h increased the intracellular level of ROS and Ca2+; decreased pH value; activated intracellular MPTP and decreased MMP in dose-dependent manners. It also induced the release of Cyt-C and the activation of Caspase-9 and -3. In conclusion, laminarin induces LOVO cell apoptosis through a mitochondrial pathway, suggesting that it could be a potent agent for cancer prevention and treatment. |

| Posted: 20 Aug 2012 12:00 AM PDT Two new alkaloids, aconicarmine (1) and aconicaramide (5), were isolated from the EtOH extract of the lateral roots of Aconitum carmichaelii, together with five known compounds: fuziline (2), neoline (3), N-ethylhokbusine B (4), 5-hydroxymethylpyrrole-2-carbaldehyde (6), and oleracein E (7). Their structures were elucidated by physical and NMR analysis. Pyrrole alkaloids were isolated from A. carmichaelii for the first time. In the in vitro assays, compounds 2 and 3 showed activity against pentobarbital sodium-induced cardiomyocytes damage by obviously recovering beating rhythm and increasing the cell viability, while compounds 5 and 7 showed moderate antibacterial activity. |

| OnLive to continue as newly-formed company Posted: 19 Aug 2012 07:52 PM PDT Newly-formed company will operate under the OnLive name using all of the previous company's assets; services, apps, and current initiatives to continue uninterrupted. OnLive have clarified recent industry reports regarding the sale of its assets to another company. In a public statement released today designed, the company confirmed that after a series of "difficult financial decisions" it had sold all of its assets (including its technology and intellectual property) to a newly-formed company that will continue to operate under the OnLive name. An affiliate of Lauder Partners has been confirmed as the first investor in the newly-structured company.
The decision was made after OnLive's board of directors decided to pursue a restructuring under an "Assignment for the Benefit of Creditors", which led to the sale of OnLive's assets to the newly-formed company. According to OnLive, the nature of this transaction did not allow the company to transfer its shares or its staff; however, the company says almost half of OnLive's staff were given employment offers by the new company at their current salaries immediately upon after transfer. The non-hired staff will also be allegedly given offers of consulting work in return for options in the new company. "Upon closing additional funding, the company plans to hire more staff, both former OnLive employees as well as new employees," the company said in its statement. Speaking on already-existing reports about OnLive's sale of assets, a company spokesperson noted that only company executives had access to the information regarding the transaction, and that "much of what we are seeing is fiction". OnLive also used the public statement to reassure users, saying all OnLive games, products, services, devices, apps, current partnerships and current initiatives are expected to continue without interruption, with all customer purchases allegedly remaining intact. The company says it also expects to make "major announcements" regarding the launch of new products and services in the coming weeks and months. "The asset acquisition, although a heartbreaking transition for everyone involved with OnLive, allows the company…s core innovation and ongoing offerings--the product of over a decade of hard work transforming the OnLive vision into reality--to survive and continue to evolve." Below is an FAQ included by OnLive in its public statement: Q. Will users see any change in the OnLive Game or Desktop Services? What about their purchases? A. Users should see no change in the OnLive Game or Desktop Services. All of their purchases remain intact and available.
