More than 200 generic drugs were screened to identify the inhibitory activity on melanogenesis in mouse B16 melanoma cells. Cinnarizine and trazodone were identified as melanogenesis inhibitors. The inhibitory effects of the two drugs on cell survival, melanogenesis, and tyrosinase activity were investigated. The results showed that both cinnarizine and trazodone inhibited melanogenesis in B16 cells by a dose-dependent manner at the non-cytotoxic concentrations. Based on the results of the present study, seeking new melanogenesis inhibitors from generic drugs is an alternative approach to developing new depigmenting agents in cosmeceuticals. Moreover, cinnarizine and trazodone were proven to be good candidates as skin-whitening agents for treatment of skin hyperpigmentation.

Molecular docking is an important method for the research of protein-protein interaction and recognition. A protein can be considered as a network when the residues are treated as its nodes. With the contact energy between residues as link weight, a weighted residue network is constructed in this paper. Two weighted parameters (strength and weighted average nearest neighbors' degree) are introduced into this model at the same time. The stability of a protein is characterized by its strength. The global topological properties of the protein-protein complex are reflected by the weighted average nearest neighbors' degree. Based on this weighted network model and these two parameters, a new docking scoring function is proposed in this paper. The scoring and ranking for 42 systems' bound and unbounded docking results are performed with this new scoring function. Comparing the results obtained from this new scoring function with that from the pair potentials scoring function, we found that this new scoring function has a similar performance to the pair potentials on some items, and this new scoring function can get a better success rate. The calculation of this new scoring function is easy, and the result of its scoring and ranking is acceptable. This work can help us better understand the mechanisms of protein-protein interactions and recognition.

This work includes specific basic characterization of synthesized glycine-based Ionic Liquid (IL) [QuatGly-OEt][EtOSO3] by NMR, elementary analysis and water content. Thermophysical properties such as density, ρ, viscosity, η, refractive index, n, and conductivity, κ, for the binary mixture of [QuatGly-OEt][EtOSO3] with poly(ethylene glycol) (PEG) [Mw = 200] are measured over the whole composition range. The temperature dependence of density and dynamic viscosity for neat [QuatGly-OEt][EtOSO3] and its binary mixture can be described by an empirical polynomial equation and by the Vogel-Tammann-Fucher (VTF) equation, respectively. The thermal expansion coefficient of the ILs is ascertained using the experimental density results, and the excess volume expansivity is evaluated. The negative values of excess molar volume for the mixture indicate the ion-dipole interactions and packing between IL and PEG oligomer. The results of binary excess property () and deviations (Δη, ∆xn, ∆Фn, ∆xR, and ∆ФR) are discussed in terms of molecular interactions and molecular structures in the binary mixture.

A simple and sensitive HPLC method has been developed in combination with fingerprint analysis and simultaneous determination of five markers, namely gallic acid, corilagin, methyl brevifolincarboxylate, ellagic acid and rutin for evaluation and quality control of Geranium carolinianum L. water extract. Extraction methods were optimized by comparing the hydrolysis efficiency of geraniin, a major tannin of the herb, resulting in the method of extraction with water under reflux. Water extracts were analyzed by HPLC, with a mobile phase of 0.1% aqueous phosphoric acid (v/v) and acetonitrile in a gradient program within 65 min. Compounds were detected at 274 nm UV wavelength. For fingerprint analysis, 17 peaks were selected as the characteristic peaks to evaluate the similarities of different samples collected from the suburb of Nanjing. The correlation coefficients of similarity were greater than 0.993. In quantitative analysis, the five selected markers showed good regression (R > 0.9991) within test ranges, and the average recoveries were between 97.2–101.7% and their RSD values were less than 4.50%. The total contents of the five markers varied from 44.28 to 71.84 mg/g. The method can be very useful for further development of G. carolinianum L. extracts and preparations.

This paper raises the design of an ultrasonic array for obstacle detection based on Phased Array (PA) techniques, which steers the acoustic beam through the environment by electronics rather than mechanical means. The transmission of every element in the array has been encoded, according to Code Division for Multiple Access (CDMA), which allows multiple beams to be transmitted simultaneously. All these features together enable a parallel scanning system which does not only improve the image rate but also achieves longer inspection distances in comparison with conventional PA techniques.

Chronic wounds typically have excessive levels of matrix metalloproteinases (MMPs) and proinflammatory cytokines that impair healing. Reducing these detrimental proteins may be key to healing chronic wounds. Proprietary protease blends were formulated specifically to degrade excessive amounts of proinflammatory factors that could prevent wound healing. Applications of protease-containing wound dressings to acute and chronic wounds have been observed clinically to resolve inflammation and appear to aid healing. The purpose of this study was to test in vitro a deliberate blend of proteases for the ability to deactivate or activate known proteins associated with inflammation or healing. Purified human target proteins were incubated with test and control solutions and samples removed at various time points. Blinded samples were tested using a novel infrared protein multiplex sandwich-ELISA-type array technique. Many proinflammatory proteins such as MMPs, cytokines and chemokines were susceptible to degradation. Many proteins such as growth factors, cytokines and TIMP1 were resistant to degradation. Not all proinflammatory proteins were deactivated. Family protein structure did not appear to affect susceptibility to degradation or deactivation. These results suggest that specific protease containing wound dressings appear to reduce multiple detrimental components which may disrupt their deleterious effects on the wound bed and microenvironment. By improving the wound microenvironment through the use of definitive proteases, these novel wound dressings may help transition wounds into the subsequent phase of healing.

In the discussions for the future DSM-5, the Substance-Related Disorders Work Group has been addressing "addiction-like" behavioral disorders such as "Internet addiction" to possibly be considered as potential additions for the diagnostic system. Most research aiming to specify and define the concept of Internet addiction (or: Excessive/Compulsive/Problematic Internet Use—PIU), takes its point of departure in conventional terminology for addiction, based in established DSM indicators. Still, it is obvious that the divide between characteristics of addiction and dimensions of new lifestyles built on technological progress is problematic and far from unambiguous. Some of these research areas are developing from the neurobiological doctrine of addiction as not being tied to specific substances. The concept of "behavioral addictions", based on biological mechanisms such as the reward systems of the brain, has been launched. The problems connected to this development are in this study discussed and reflected with data from a Swedish survey on Internet use (n = 1,147). Most Swedes (85%) do use the Internet to some degree. The prevalence of excessive use parallels other similar countries. Respondents in our study spend (mean value) 9.8 hours per week online at home, only 5 percent spend more than 30 hours per week. There are both positive and negative social effects at hand. Many respondents have more social contacts due to the use of Internet, but there is a decline in face-to-face contacts. About 40% of the respondents indicate some experience of at least one problem related to Internet use, but only 1.8% marked the presence of all problems addressed. Most significant predictors for problem indicators, except for age, relate to "time" and time consuming activities such as gaming, other activities online or computer skills.

This paper presents the findings of the impact of atmospheric effects when applied on satellite images intended for supporting archaeological research. The study used eleven multispectral Landsat TM/ETM+ images from 2009 until 2010, acquired over archaeological and agricultural areas. The modified Darkest Pixel (DP) atmospheric correction algorithm was applied, as it is considered one of the most simple and effective atmospheric corrections algorithm. The NDVI equation was applied and its values were evaluated before and after the application of atmospheric correction to satellite images, to estimate its possible effects. The results highlighted that atmospheric correction has a significant impact on the NDVI values. This was especially true in seasons where the vegetation has grown. Although the absolute impact on NDVI, after applying the DP, was small (0.06), it was considered important if multi-temporal time series images need to be evaluated and cross-compared. The NDVI differences, before and after atmospheric correction, were assessed using student's t-test and the statistical differences were found to be significant. It was shown that relative NDVI difference can be as much as 50%, if atmosphere effects are ignored. Finally, the results had proven that atmospheric corrections can enhance the interpretation of satellite images (especially in cases where optical thickness of water vapour is minimized ≈ 0). This fact can assist in the detection and identification of archaeological crop marks. Therefore, removal of atmospheric effects, for archaeological purposes, was found to be of great importance in improving the image enhancement and NDVI values.

GT 5 Spec II arriving in Japan Feb 2012

Sony formally announces arrival of Spec II in Japan; pack to include game and all previous DLC; no formal announcement on European or US release.

 

After five years in development, Polyphony Digital's PlayStation exclusive racer Gran Turismo 5 shipped 6.37 million units worldwide following its highly-anticipated release last November. Publisher Sony has since supported the game, releasing a "Spec 2.0" update and downloadable content pack earlier this year.

Now, Japanese gaming site Andria Sang is reporting that Sony Japan has formally announced the Gran Turismo 5 Spec II pack, which will include the updated version of Gran Turismo 5 with all the content from the previous update, as well as a product code for downloading the DLC content released earlier this year.

The site is reporting the Spec II pack will hit store shelves in Japan in February 2012, and will cost é4,980 ($63.99).

The Gran Turismo 5 "Spec 2.0" update offers new interiors for all cars in the game, reduced load times, the ability to save during a lengthy race, and the option to adjust weather conditions.

Additionally, the update includes revisions to the game's Photo and Shuffle modes, offers full support for Logitech's GT5 and GT7 racing wheels, and gives gamers the 2011 versions of 11 NASCAR vehicles.

The DLC offered in the Spec II pack includes new vehicles and tracks.

Sony reportedly told Eurogamer that it has no plans to release the pack in Europe at this time. The publisher has also made no other mention of the pack hitting the US or any other territories.

Square Enix announces Chousoku Henkei Gyrozetter

Card-based game to be released in arcades and on the 3DS; manga and anime spin-off in the works.

The last card game Square Enix published for arcades was the 2008 fantasy-based Lord of Vermillion series. Now, the Final Fantasy-publisher plans to take the card concept towards a new futuristic IP. The new title, called Chousoku Henkei Gyrozetter, will be released in arcades and on the 3DS in Japan in 2012.

In the game, players control vehicles that can transform into robots. Users will have to use cards to initiate attacks and special moves against enemy robots, while alternating between vehicle and robot mode.

In the arcade version, the arcade cabinet will transform according to the mode the player is in. For instance, if a player is in a driving segment, they will have to use the steering wheel on the cabinet. If in robot mode, gamers will have access to special levers that appear from the machine.

In addition, the publisher is teaming up with Japanese car makers Mitsubishi, Nissan, and Mazda to bring licensed cars to the game. The game will be developed by Rocket Studio, renowned for the Dragon Quest: Monsters Battle Road arcade series. At this point, there aren't any concrete details on the 3DS version of the game.

Furthermore, Square Enix has plans to launch a manga and TV anime series for the franchise. The former will be out in Japan's Sueisha's Saikyou Jump magazine starting December 3, while the latter is currently in production by A-1 Pictures studio.

Gyrozetter will be out in arcades on 2012 in Japan.
