PA1b (Pea Albumin 1, subunit b) is a peptide extract from pea seeds showing significant insecticidal activity against certain insects, such as cereal weevils (genus Sitophilus), the mosquitoes Culex pipiens and Aedes aegyptii, and certain species of aphids. PA1b has great potential for use on an industrial scale and for use in organic farming: it is extracted from a common plant; it is a peptide (and therefore suitable for transgenic applications); it can withstand many steps of extraction and purification without losing its activity; and it is present in a seed regularly consumed by humans and mammals without any known toxicity or allergenicity. The potential of this peptide to limit pest damage has stimulated research concerning its host range, its mechanism of action, its three-dimensional structure, the natural diversity of PA1b and its structure–function relationships.

The M2 channel protein on the influenza A virus membrane has become the main target of the anti-flu drugs amantadine and rimantadine. The structure of the M2 channel proteins of the H3N2 (PDB code 2RLF) and 2009-H1N1 (Genbank accession number GQ385383) viruses may help researchers to solve the drug-resistant problem of these two adamantane-based drugs and develop more powerful new drugs against influenza A virus. In the present study, we searched for new M2 channel inhibitors through a combination of different computational methodologies, including virtual screening with docking and pharmacophore modeling. Virtual screening was performed to calculate the free energies of binding between receptor M2 channel proteins and 200 new designed ligands. After that, pharmacophore analysis was used to identify the important M2 protein-inhibitor interactions and common features of top binding compounds with M2 channel proteins. Finally, the two most potential compounds were determined as novel leads to inhibit M2 channel proteins in both H3N2 and 2009-H1N1 influenza A virus.

Molecules, Vol. 16, Pages 10214-10226: Antioxidant, Anti-Glycation and Anti-Inflammatory Activities of Phenolic Constituents from Cordia sinensis

Nine compounds have been isolated from the ethyl acetate soluble fraction of C. sinensis, namely protocatechuic acid (1), trans-caffeic acid (2), methyl rosmarinate (3), rosmarinic acid (4), kaempferide-3-O-β-D-glucopyranoside (5), kaempferol-3-O-β-D-glucopyranoside (6), quercetin-3-O-β-D-glucopyranoside (7), kaempferide-3-O-α-L-rhamnopyranosyl (1→6)-β-D-glucopyranoside (8) and kaempferol-3-O-α-L-rhamno-pyranosyl (1→6)-β-D-glucopyranoside (9), all reported for the first time from this species. The structures of these compounds were deduced on the basis of spectroscopic studies, including 1D and 2D NMR techniques. Compounds 1–9 were investigated for biological activity and showed significant anti-inflammatory activity in the carrageen induced rat paw edema test. The antioxidant activities of isolated compounds 1–9 were evaluated by the DPPH radical scavenging test, and compounds 1, 2, 4 and 7–9 exhibited marked scavenging activity compared to the standard BHA. These compounds were further studied for their anti-glycation properties and some compounds showed significant anti-glycation inhibitory activity. The purity of compounds 2–5, 8 and 9 was confirmed by HPLC. The implications of these results for the chemotaxonomic studies of the genus Cordia have also been discussed.

A 'Heat treatment aqueous two phase system' was employed for the first time to purify serine protease from kesinai (Streblus asper) leaves. In this study, introduction of heat treatment procedure in serine protease purification was investigated. In addition, the effects of different molecular weights of polyethylene glycol (PEG 4000, 6000 and 8000) at concentrations of 8, 16 and 21% (w/w) as well as salts (Na-citrate, MgSO4 and K2HPO4) at concentrations of 12, 15, 18% (w/w) on serine protease partition behavior were studied. Optimum conditions for serine protease purification were achieved in the PEG-rich phase with composition of 16% PEG6000-15% MgSO4. Also, thermal treatment of kesinai leaves at 55 °C for 15 min resulted in higher purity and recovery yield compared to the non-heat treatment sample. Furthermore, this study investigated the effects of various concentrations of NaCl addition (2, 4, 6 and 8% w/w) and different pH (4, 7 and 9) on the optimization of the system to obtain high yields of the enzyme. The recovery of serine protease was significantly enhanced in the presence of 4% (w/w) of NaCl at pH 7.0. Based on this system, the purification factor was increased 14.4 fold and achieved a high yield of 96.7%.

Religions, Vol. 2, Pages 693-706: Measuring Mindfulness: A Rasch Analysis of the Freiburg Mindfulness Inventory

The objective of the study was to assess the psychometric properties of the Freiburg Mindfulness Inventory (FMI-14) using a Rasch model approach in a cross-sectional design. The scale was administered to N = 130 British patients with different psychosomatic conditions. The scale failed to show clear one-factoriality and item 13 did not fit the Rasch model. A two-factorial solution without item 13, however, appeared to fit well. The scale seemed to work equally well in different subgroups such as patients with or without mindfulness practice. However, some limitations of the validity of both the one-factorial and the two-factorial version of the scale were observed. Sizeable floor and ceiling effects limit the diagnostical use of the instrument. In summary, the study demonstrates that the two-factorial version of the FMI-13 shows acceptable approximation to Rasch requirements, but is in need of further improvement. The one-factorial solution did not fit well, and cannot be recommended for further use.

In light of increased scholarly interest in the scientific study of non-Christian religions and societies, I review sociological research on Hinduism. Specifically, I focus on Hindu congregational forms, a phenomenon noted in social scientific literature. Drawing on existing theories from the sociology of religion, this article illuminates possible social sources of Hindu congregational forms. Two preliminary sources are proposed and possible mechanisms elaborated: (1) modernization and (2) social networks. I conclude by proposing several new directions for research on Hindu congregational forms. These arguments and proposals offer directions for expanding understanding of how theories in the sociology of religion might operate beyond Christianity and the West.

Aptamers are small non-coding RNAs capable of recognizing, with high specificity and affinity, a wide variety of molecules in a manner that resembles antibodies. This class of nucleic acids is the resulting product of applying a well-established screening method known as SELEX. First developed in 1990, the SELEX process has become a powerful tool to select structured oligonucleotides for the recognition of targets, starting with small molecules, going through protein complexes until whole cells. SELEX has also evolved along with new technologies positioning itself as an alternative in the design of a new class of therapeutic agents in modern molecular medicine. This review is an historical follow-up of SELEX method over the two decades since its first appearance.

Cis-9, trans-11 conjugated linoleic acid (c9, t11 CLA) producing bacteria have attracted much attention as novel probiotics which have shown beneficial effects on host health. However, bile salts are able to inhibit bacterial growth and c9, t11 CLA production. For recovering growth and c9, t11 CLA production of Lactobacillus acidophilus F0221 in the presence of bile salts, Tween series (Tween 20, Tween 40, Tween 60 and Tween 80) were added in growth culture containing 0.3% oxgall. Results showed that the viable counts were significantly (P < 0.05) recovered to 8.58–8.75 log CFU/mL in the presence of all Tween treatments. However, recovery of c9, t11 CLA production was only demonstrated in the presence of Tween 80 (72.89 μg/mL). Stepwise increasing oxgall in a concentrations range from 0.1% to 0.9% according to human intestinal physiological environments, Tween 80 still showed significant (P < 0.05) recovery ability on growth (8.91–8.04 log CFU/mL) and c9, t11 CLA (69.22–34.27 μg/mL) production. The effect of Tween 80 on growth and production was also investigated in the presence of different types of bile salts (sodium salts of cholic acid (CA), deoxycholic acid (DCA), chendeoxycholic acid (CDCA), glycocholic acid (GCA) and taurocholic acid (TCA)). Results showed that Tween 80 could significantly (P < 0.05) recover c9, t11 CLA production in the presence of all types of bile salts, but the Tween 80 could only significantly (P < 0.05) recover viable counts of the strain in the presence of CA, DCA and CDCA. This recovery ability could be attributed to the protection of leakage of intracellular material. Additionally, although bile salts inhibited growth and c9, t11 CLA production by the growing cell, it promoted the c9, t11 CLA production by the resting cell.

Big in Japan Nov. 28 – Dec. 4: Mario Kart 7

Nintendo's latest racer beats off robots and Italian assassins for top spot; 3DS unit sales double since week of Nov. 21.

 

It was a Nintendo-filled week for Japan in terms of game sales, with big names like Mario and Zelda in the top 10 for the week of November 28 to December 4 on the latest Media Creates sales update. Mario Kart 7 raced its way to first place with 423,619 units sold, bumping the PS3 port of Kidou Senshi Gundam: Extreme VS. to second place. The latter sold 349,756 units on its first week of release.

Mario Kart 7 was recently praised for its new additions to the same formula such as kart customization, hang gliding, and rebalanced items. However, the game still has notable issues such as its online structure and the lack of mission mode. For more information, check out GameSpot's gamespace of the 3DS title.

Kidou Senshi Gundam: Extreme VS. is a PS3 port of Mobile Suit Gundam: Extreme VS for the arcade. In this 3D fighting game, players have to reduce the opposing team's points from 6000 to zero by destroying enemy Gundams.

Assassin's Creed: Revelations made its chart debut this week, coming in at fourth place with 39,947 units sold. Super Mario 3D Land for the 3DS went up to third place with 73,907 units sold, while The Legend of Zelda: Skyward Sword for the Wii went down from first to fifth place with 32,191 units sold. The PS3 version of arcade fighting title The King of Fighters XIII made its debut at 10th place with 21,525 units sold.

Meanwhile on the hardware front, the 3DS saw a huge boost in sales thanks in part to the recently-out Mario Kart title and Super Mario 3D Land. Nintendo sold 205,962 units for the week, which was close to double the sales of the system on the previous week. Sony sold 40,668 units of the PS3 and was placed second, while the company sold 39,547 units of the PSP and was placed third.

In related news, Nintendo CEO Satoru Iwata said on Nikkei that the 3DS will soon reach the 3 million sales mark thanks to the recent price drop and major releases like Super Mario 3D Land and Mario Kart 7. He also said that he expects the system to sell 4 million units before its one year anniversary.

Sega announces Samurai & Dragons for PS Vita

Action-strategy hybrid from Yakuza producer is coming to Sony's portable console; release date unconfirmed.

Gamers may recall the old SNES part-action, part-simulation title Actraiser, which allowed players to build up civilisations while requiring them to clear demon-infested areas in 2D side-scrolling glory. Sega will be putting a new spin on the hybrid concept with its newest PlayStation Vita title, announced this week in Japanese gaming publication Famitsu.

The title--called Samurai & Dragons--will be split into two parts: action and strategy. The former will require up to four players to fight their way through a dungeon, defeating dragons, mummies, and ogres, and getting loot. The strategy part of the game will involve players taking their spoils of war to build up a customisable city and train a monster army. During this part, gamers will also need to form alliances with other players, or proceed to conquer their cities with their own army.

The game's producer is Masayoshi Kikuchi, who also served as producer on the Yakuza series, director on Jet Set Radio Future, and planner for the Panzer Dragoon series. Sega's Online Entertainment Research Department, the team developing the title, said that the PS Vita was chosen for the project due to its online connectivity and potential of the device's touchscreen and motion sensors.

No release date has been announced for the title at this point in time.

IJMS, Vol. 12, Pages 9125-9137: Swelling-Activated Anion Channels Are Essential for Volume Regulation of Mouse Thymocytes

Channel-mediated trans-membrane chloride movement is a key process in the active cell volume regulation under osmotic stress in most cells. However, thymocytes were hypothesized to regulate their volume by activating a coupled K-Cl cotransport mechanism. Under the patch-clamp, we found that osmotic swelling activates two types of macroscopic anion conductance with different voltage-dependence and pharmacology. At the single-channel level, we identified two types of events: one corresponded to the maxi-anion channel, and the other one had characteristics of the volume-sensitive outwardly rectifying (VSOR) chloride channel of intermediate conductance. A VSOR inhibitor, phloretin, significantly suppressed both macroscopic VSOR-type conductance and single-channel activity of intermediate amplitude. The maxi-anion channel activity was largely suppressed by Gd3+ ions but not by phloretin. Surprisingly, [(dihydroindenyl)oxy] alkanoic acid (DIOA), a known antagonist of K-Cl cotransporter, was found to significantly suppress the activity of the VSOR-type single-channel events with no effect on the maxi-anion channels at 10 μM. The regulatory volume decrease (RVD) phase of cellular response to hypotonicity was mildly suppressed by Gd3+ ions and was completely abolished by phloretin suggesting a major impact of the VSOR chloride channel and modulatory role of the maxi-anion channel. The inhibitory effect of DIOA was also strong, and, most likely, it occurred via blocking the VSOR Cl− channels.
