AUCKLAND, New Zealand – September 1, 2010 – Independent games studio Grinding Gear Games today announced Path of Exile, a free-to-play PC online action Role-Playing Game (RPG) set in a dark fantasy world. The game will be available in early 2011.

Playing as one of six character classes, players find themselves banished for their past misdeeds to the dark fantasy world of Wraeclast. There, they will encounter hundreds of species of opponents laden with loot and mysterious artefacts as they explore the abandoned continent.

Players will be able to purchase in-game perks, such as extra victory animations, clothing and special effects. To protect the integrity of the game for all players, Grinding Gear Games has decided not to sell abilities or experience points – ensuring a level-playing field for everyone in game.

"We listened to the fan communities of several popular online action RPGs as we designed Path of Exile and are focused on replayability, depth and accessibility," said Chris Wilson, Grinding Gear Games' Founder. "We've designed the game that we would want to play ourselves, and we know hardcore Diablo, Titan Quest and Dungeon Siege players will find the style familiar and addictive."

Wilson believes there has been a shortage of dark fantasy worlds in recent RPGs. "While the art style is a return to the classic dark fantasy that many hardcore players grew up with, we're confident the difficulty curve is accessible enough for anyone. There are no rainbows and unicorns in our game, but there is plenty of blood and rare items to collect."

Some fluoro-substituted liquid crystals mixed with arachidic acid in monolayers formed at air-liquid (Langmuir films) and air-solid substrate (Langmuir-Blodgett films) interfaces were investigated. Molecular organization in Langmuir films was determined on the basis of the analysis of the shape of the surface pressure-mean molecular area isotherm and observations made by means of a Brewster angle microscope. It was found that in the compression process the liquid crystal molecules are pushed out towards the top of the first monolayer being in direct contact with the subphase. Langmuir films were transferred onto the quartz substrates at various surface pressures and mono- and multilayered Langmuir-Blodgett films were obtained. The films were characterized using electronic absorption measurements. The conditions for obtaining the homeotropic orientation of the liquid crystal molecules were determined.

Methylsulfonylmethane (or dimethyl sulfone), a naturally produced and vitally important organosulfur compound in living organisms, was irradiated with gamma rays, and the produced radicals were investigated using electron paramagnetic resonance spectroscopy at different temperatures. The structure and behavior of the radical changed when the temperatures varied. The hyperfine splitting of the CH3 group was small, and the 33S splitting was relatively high between 80 and −50 °C. When the temperature was between −50 and −160 °C, the 33S splitting became small and the CH3 splitting was higher. However, the group kept rotating; therefore, only the isotropic splitting values were measured, and the g-values were anisotropic. When the temperature decreased below −180 °C, the CH3 group stopped rotating, and the hydrogen splitting values became nonequivalent due to an inhomogeneous electron distribution. The observed structures can be explained by referring to both the experimental and theoretically calculated values reported.

Dietary antioxidants play an important role against oxidation, an underlying mechanism in the incidence of chronic diseases. Greens+ is a commercially available preparation containing a variety of plant-derived ingredients. The aim of the current study was to evaluate the antioxidant potential of the methanolic extract of greens+ powder using in vitro and in vivo techniques. In vitro studies were conducted using a liposome model system to simulate biological cell membranes. Total antioxidant potential and polyphenol content of the herbal preparation was measured. For in vivo analysis, 10 healthy human subjects consumed either three or six teaspoons of greens+ per day for four weeks. Blood samples were analyzed at baseline and at the conclusion of the treatment period for total antioxidant capacity, polyphenol content, protein, lipid and LDL oxidation, and the level of glutathione peroxidase. Results showed that greens+ supplementation was well tolerated and increased serum antioxidant potential at higher levels of intake in a dose-dependent manner. HPLC analysis showed the presence of quercetin, apigenin, kaempferol and luteolin in the supplement. Plasma analysis indicated the presence of kaempferol only. A statistically significant (p < 0.05) reduction in protein and lipid oxidation was observed. Based on its antioxidant properties, the results suggest that greens+ might play a role in reducing the risk of chronic diseases involving a burden of oxidative damage.

A series of new bis(heterocycles) featuring thieno[2,3-b]thiophene rings was synthesized in a combinatorial manner. Intramolecular cyclization of enaminone derivatives with appropriate N-nucleophiles afforded the target compounds. All compounds were characterized by 1H-, 13C-NMR, GCMS, IR, and UV-Vis spectrometry. These compounds represent a new class of sulfur- and nitrogen-containing heterocycles that should also be of interest as new materials.

In the present study, the anti-influenza A (H2N2) virus activity of patchouli alcohol was studied in vitro, in vivo and in silico. The CC50 of patchouli alcohol was above 20 µM. Patchouli alcohol could inhibit influenza virus with an IC50 of 4.03 ± 0.23 µM. MTT assay showed that the inhibition by patchouli alcohol appears strongly after penetration of the virus into the cell. In the influenza mouse model, patchouli alcohol showed obvious protection against the viral infection at a dose of 5 mg/kg/day. Flexible docking and molecular dynamic simulations indicated that patchouli alcohol was bound to the neuraminidase protein of influenza virus, with an interaction energy of –40.38 kcal mol–1. The invariant key active-site residues Asp151, Arg152, Glu119, Glu276 and Tyr406 played important roles during the binding process. Based on spatial and energetic criteria, patchouli alcohol interfered with the NA functions. Results presented here suggest that patchouli alcohol possesses anti-influenza A (H2N2) virus properties, and therefore is a potential source of anti-influenza agents for the pharmaceutical industry.

Phenylacetonitrile, (E)-b-ocimene, linalool, (E)-4,8-dimethyl-1,3,7-nonatriene and (E,E)-a-farnesene were identified as Japanese beetle, Popillia japonica, feeding-induced volatiles from the leaves of the giant knotweed, Fallopia sachalinensis, but not by mechanical damage. Volatile emission was also induced by treatment with a cellular signaling molecule, methyl jasmonate. These results suggest that volatiles will be synthesized de novo by a biotic elicitor from P. japonica oral secretion.
