Nine new microsatellite markers (SSR) were isolated from Salvia officinalis L. A total of 125 alleles, with 8 to 21 alleles per locus, were detected in a natural population from the east Adriatic coast. The observed heterozygosity, expected heterozygosity, and polymorphic information content ranged from 0.46 to 0.83, 0.73 to 0.93 and 0.70 to 0.92, respectively. New microsatellite markers, as well as previously published markers, were tested for cross-amplification in Salvia brachyodon Vandas, a narrow endemic species known to be present in only two localities on the Balkan Peninsula. Out of 30 microsatellite markers tested on the natural S. brachyodon population, 15 were successfully amplified. To obtain evidence of recent bottleneck events in the populations of both species, observed genetic diversity (HE) was compared to the expected genetic diversity at mutation-drift equilibrium (HEQ) and calculated from the observed number of alleles using a two-phased mutation model (TPM). Recent bottleneck events were detected only in the S. brachyodon population. This result suggests the need to reconsider the current threat category of this endemic species.

Little is known about the interactions of single nucleotide polymorphisms (SNPs) and overweight/obesity on blood pressure levels. The present study was undertaken to detect 10 lipid-related gene SNPs and their interactions with overweight/obesity on blood pressure levels. Genotyping of ATP-binding cassette transporter A1 (ABCA-1) V825I, acyl-CoA:cholesterol acyltransferase-1 (ACAT-1) rs1044925, low density lipoprotein receptor (LDL-R) AvaII hepatic lipase gene (LIPC) −250G > A, endothelial lipase gene (LIPG) 584C > T, methylenetetrahydrofolate reductase (MTHFR) 677C > T, the E3 ubiquitin ligase myosin regulatory light chain-interacting protein (MYLIP) rs3757354, proprotein convertase subtilisin-like kexin type 9 (PCSK9) E670G, peroxisome proliferator-activated receptor delta (PPARD) +294T > C, and Scavenger receptor class B type 1 (SCARB1) rs5888 was performed in 978 normal weight and 751 overweight/obese subjects. The interactions were detected by factorial regression analysis. The genotypes of ACAT-1 AC, LIPC GA and AA, and SCARB1 TT; LDL-R A-A- and LIPC GA; and SCARB1 TT were interacted with overweight/obesity to increase systolic, diastolic blood pressure (SBP, DBP) and pulse pressure (PP) levels; respectively. The genotypes of ACAT-1 CC; ACAT-1 AA and CC were interacted with overweight/obesity to decrease SBP, PP levels (p < 0.01–0.001); respectively. The differences in blood pressure levels between normal weight and overweight/obese subjects might partly result from different interactions of several SNPs and overweight/obesity.

The 40 million hectare southern Australian winter cropping region suffers from widespread infestation by Lolium rigidum (commonly known as annual or rigid ryegrass), a Mediterranean species initially introduced as a pasture plant. Along with its high competitiveness within crops, rapid adaptability and widespread resistance to herbicides, the dormancy of its seeds means that L. rigidum is the primary weed in southern Australian agriculture. With the individuals within a L. rigidum population exhibiting varying levels of seed dormancy, germination can be staggered across the crop-growing season, making complete weed removal virtually impossible, and ensuring that the weed seed bank is constantly replenished. By understanding the processes involved in induction and release of dormancy in L. rigidum seeds, it may be possible to develop strategies to more effectively manage this pest without further stretching herbicide resources. This review examines L. rigidum seed dormancy and germination from a weed-management perspective and explains how the seed bank can be depleted by control strategies encompassing all stages in the lifecycle of a seed, from development to germination.

Metabolites, Vol. 2, Pages 632-647: A Topological Characterization of Medium-Dependent Essential Metabolic Reactions

Metabolism has frequently been analyzed from a network perspective. A major question is how network properties correlate with biological features like growth rates, flux patterns and enzyme essentiality. Using methods from graph theory as well as established topological categories of metabolic systems, we analyze the essentiality of metabolic reactions depending on the growth medium and identify the topological footprint of these reactions. We find that the typical topological context of a medium-dependent essential reaction is systematically different from that of a globally essential reaction. In particular, we observe systematic differences in the distribution of medium-dependent essential reactions across three-node subgraphs (the network motif signature of medium-dependent essential reactions) compared to globally essential or globally redundant reactions. In this way, we provide evidence that the analysis of metabolic systems on the few-node subgraph scale is meaningful for explaining dynamic patterns. This topological characterization of medium-dependent essentiality provides a better understanding of the interplay between reaction deletions and environmental conditions.

Eight novel cyclic bis-1,3-dialkylpyridiniums, as well as two known compounds from the cyclostellettamine class, were isolated from the sponge Haliclona sp. from Korea. Structures of these novel compounds were determined using combined NMR and FAB-MS/MS analyses. Several of these compounds exhibited moderate cytotoxic and antibacterial activities against A549 cell-line and Gram-positive strains, respectively. The structure-activity relationships of cyclostellettamines are discussed based on their bioactivities.

Marine Drugs, Vol. 10, Pages 2111-2125: Dose-Response on the Chemopreventive Effects of Sarcophine-Diol on UVB-Induced Skin Tumor Development in SKH-1 Hairless Mice

Sarcophine-diol (SD) is a lactone ring-opened analogue of sarcophine. It has shown chemopreventive effects on chemically-induced skin tumor development in female CD-1 mice, as well as in a UVB-induced skin tumor development model in hairless SKH-1 mice at a dose of 30 μg SD applied topically and 180 mJ/cm2 UVB. The objective of this study was to determine the dose-response on the chemopreventive effects of SD on SKH-1 hairless mice when exposed to a UVB radiation dose of 30 mJ/cm2. This UVB dose better represents chronic human skin exposure to sunlight leading to skin cancer than previous studies applying much higher UVB doses. Carcinogenesis was initiated and promoted by UVB radiation. Female hairless SKH-1 mice were divided into five groups. The control group was topically treated with 200 μL of acetone (vehicle), and the SD treatment groups were topically treated with SD (30 μg, 45 μg, and 60 μg dissolved in 200 μL of acetone) 1 h before UVB radiation (30 mJ/cm2). The last group of animals received 60 μg SD/200 μL acetone without UVB exposure. These treatments were continued for 27 weeks. Tumor multiplicity and tumor volumes were recorded on a weekly basis for 27 weeks. Weight gain and any signs of toxicity were also closely monitored. Histological characteristics and the proliferating cell nuclear antigen (PCNA) were evaluated in the mice skin collected at the end of the experiment. The dose-response study proved a modest increase in chemopreventive effects with the increase in SD dose. SD reduced the number of cells positively stained with PCNA proliferation marker in mice skin. The study also showed that SD application without UVB exposure has no effect on the structure of skin. The results from this study suggest that broader range doses of SD are necessary to improve the chemopreventive effects.

In the continued search for novel bioactive substances from the Taiwanese octocoral Briareum excavatum collected at Orchid Island, three new briarane-type diterpenoids, briacavatolides D–F (1–3) were isolated from the acetone extract. The structures of these compounds were elucidated by extensive NMR spectroscopic analysis and physical data. The anti-HCMV (human cytomegalovirus) activity of 1–3 and their cytotoxicity against selected cancer cell lines were evaluated.

A combination of on-line HPLC-NMR and off-line chemical investigations has resulted in the identification of the previously reported polyhalogenated monoterpene plocamenone, together with the new structural analogue isoplocamenone from the crude extract of the marine alga Plocamium angustum. On-flow and stop-flow HPLC-NMR analyses (including the acquisition of WET 2D NMR spectra) rapidly assisted in the identification of the major component plocamenone and in the partial identification of its unstable double bond isomer isoplocamenone. Conventional off-line isolation and structural characterization techniques were employed to unequivocally confirm both structures, leading to a structural revision for plocamenone, as well as to obtain sufficient quantities for biological testing.

We have investigated several building stone materials, including minerals and rocks, using continuous flow hyperpolarized xenon (CF-HP) NMR spectroscopy to probe the surface composition and porosity. Chemical shift and line width values are consistent with petrographic information. Rare upfield shifts were measured and attributed to the presence of transition metal cations on the surface. The evolution of freshly cleaved rocks exposed to the atmosphere was also characterized. The CF-HP 129Xe NMR technique is non-destructive and it could complement currently used techniques, like porosimetry and microscopy, providing additional information on the chemical nature of the rock surface and its evolution.

A batch experiment was conducted to compare the Cu scavenging capacity between two different red mud types: the first one was a highly basic red mud derived from a combined sintering and Bayer process, and the second one was a seawater-neutralized red mud derived from the Bayer process. The first red mud contained substantial amounts of CaCO3, which, in combination with the high OH− activity, favored the immobilization of water-borne Cu through massive formation of atacamite. In comparison, the seawater-neutralized red mud had a lower pH and was dominated by boehmite, which was likely to play a significant role in Cu adsorption. Overall, it appears that Cu was more tightly retained by the CaCO3-dominated red mud than the boehmite-dominated red mud. It is concluded that the heterogeneity of red mud has marked influences on its capacity to immobilize water-borne Cu and maintain the long-term stability of the immobilized Cu species. The research findings obtained from this study have implications for the development of Cu immobilization technology by using appropriate waste materials generated from the aluminium industry.

An experimental study of plain concrete specimens of water-cement ratio 0.55, subjected to 0, 15, 25, 40, 50 and 75 cycles of freeze-thaw was completed. The dynamic modulus of elasticity (DME), weight loss, compressive strength, tensile strength, flexural strength, cleavage strength and stress-strain relationships of plain concrete specimens suffering from freeze-thaw cycles were measured. The experimental results showed that the strength decreased as the freeze-thaw cycles were repeated. A concise mathematic formula between DME, weight loss, mechanical properties and number of freeze-thaw cycles was also established. The influences of freeze-thaw cycles on the DME, weight loss and mechanical properties were analyzed. The experimental results serve as a reference for the maintenance, design and life prediction of dams, hydraulic structures, offshore structures, concrete roads and bridges in cold regions.

Multivariate hydrologic frequency analysis has been widely studied using: (1) commonly known joint distributions or copula functions with the assumption of univariate variables being independently identically distributed (I.I.D.) random variables; or (2) directly applying the entropy theory-based framework. However, for the I.I.D. univariate random variable assumption, the univariate variable may be considered as independently distributed, but it may not be identically distributed; and secondly, the commonly applied Pearson's coefficient of correlation (g) is not able to capture the nonlinear dependence structure that usually exists. Thus, this study attempts to combine the copula theory with the entropy theory for bivariate rainfall and runoff analysis. The entropy theory is applied to derive the univariate rainfall and runoff distributions. It permits the incorporation of given or known information, codified in the form of constraints and results in a universal solution of univariate probability distributions. The copula theory is applied to determine the joint rainfall-runoff distribution. Application of the copula theory results in: (i) the detection of the nonlinear dependence between the correlated random variables-rainfall and runoff, and (ii) capturing the tail dependence for risk analysis through joint return period and conditional return period of rainfall and runoff. The methodology is validated using annual daily maximum rainfall and the corresponding daily runoff (discharge) data collected from watersheds near Riesel, Texas (small agricultural experimental watersheds) and Cuyahoga River watershed, Ohio.
