Cancers, Vol. 4, Pages 846-872: A microRNA Link to Glioblastoma Heterogeneity Posted: 03 Sep 2012 12:00 AM PDT Glioblastomas (GBM) are one of the most malignant adult primary brain tumors. Through decades of research using various model systems and GBM patients, we have gained considerable insights into the mechanisms regulating GBM pathogenesis, but have mostly failed to significantly improve clinical outcome. For the most part GBM heterogeneity is responsible for this lack of progress. Here, we have discussed sources of cellular and microenvironmental heterogeneity in GBMs and their potential regulation through microRNA mediated mechanisms. We have focused on the role of individual microRNAs (miRNA) through their specific targets and miRNA mediated RNA-RNA interaction networks with the potential to influence various aspects of GBM heterogeneity including tumor neo-vascularization. We believe a better understanding of such mechanisms for regulation of GBM pathogenesis will be instrumental for future therapeutic options. |

Aging in humans is generally associated with deterioration of cognitive performance and, in particular, learning and memory. Different therapeutic approaches are available to treat cognitive impairment during physiological aging and neurodegenerative or psychiatric disorders. Traditional herbal medicine and numerous plants, either directly as supplements or indirectly in the form of food, improve brain functions including memory and attention. More than a hundred herbal medicinal plants have been traditionally used for learning and memory improvement, but only a few have been tested in randomized clinical trials. Here, we will enumerate those medicinal plants that show positive effects on various cognitive functions in learning and memory clinical trials. Moreover, besides natural products that show promising effects in clinical trials, we briefly discuss medicinal plants that have promising experimental data or initial clinical data and might have potential to reach a clinical trial in the near future. |
Microsoft reveals list of Xbox Live games coming to Windows 8 Posted: 02 Sep 2012 11:58 PM PDT First set of Xbox Live games heading to Windows 8 announced; some titles to be exclusive to the OS. The first set of Xbox Live games to be available on Windows 8 has been revealed. An announcement on Major Nelson's blog listed 40 titles that will be making their way to the OS, including titles from Microsoft Studios, Rovio, Miniclip, Gameloft, ZeptoLab, Glu Mobile, and Halfbrick Studios. Some of the titles will be exclusive to Windows 8, but all will be playable on any Windows 8 PC, laptop or tablet.
Previously, Microsoft announced that Xbox Live would be available on Windows 8, rolling out with the launch of the operation system. Earlier this year, Microsoft CEO Steve Ballmer confirmed that the Kinect motion-sensing camera peripheral would be coming to the PC, saying that, "We'll support that in a formal way in the right time, and when we have an announcement to make, we'll make it." (The company has since released the Kinect software development kit for the PC.) Black Mesa Half-Life mod launching September 14 Posted: 02 Sep 2012 09:40 PM PDT First part of Source-powered community mod will be released next week; initial release will stop at Lambda Core level, offering 8-10 hours of gameplay.
The community behind the development of Half-Life mod Black Mesa (formerly Black Mesa: Source) has announced the first part of the game will finally be released on September 14 this year.
The mod's official website has been updated with a countdown timer, and Black Mesa project leader Carlos Montero posted on the site's forums confirming the game's official release. "On September 14th you will see the first release of Black Mesa," Montero said in his post. "This will include our re-envisioning of Half-Life all the way up to Lambda Core. We believe this is a great way to provide a complete-feeling 8-10 hour experience with a solid ending, make our fans happy and help us make the best overall game possible." A remake of Valve's original 1998 title Half-Life using the Source Engine, Black Mesa promises a fully reconstructed Half-Life universe built from the ground up with new models, maps, soundtrack, voice acting, and textures. The remake will include the original game's protagonists, Gordon Freeman, as well as the original cast of characters and environments. The mod project was launched in 2004 following fan disappointment over Valve's official Half-Life remake, Half-Life: Source. According to the project's site, Black Mesa will not require Half-Life: Source to play--just a copy of any Source engine game installed on Steam. Posted: 03 Sep 2012 12:00 AM PDT This paper describes an approach for the early combination of material characterization and toxicology testing in order to design carbon nanofiber (CNF) with low toxicity. The aim was to investigate how the adjustment of production parameters and purification procedures can result in a CNF product with low toxicity. Different CNF batches from a pilot plant were characterized with respect to physical properties (chemical composition, specific surface area, morphology, surface chemistry) as well as toxicity by in vitro and in vivo tests. A description of a test battery for both material characterization and toxicity is given. The results illustrate how the adjustment of production parameters and purification, thermal treatment in particular, influence the material characterization as well as the outcome of the toxic tests. The combination of the tests early during product development is a useful and efficient approach when aiming at designing CNF with low toxicity. Early quality and safety characterization, preferably in an iterative process, is expected to be efficient and promising for this purpose. The toxicity tests applied are preliminary tests of low cost and rapid execution. For further studies, effects such as lung inflammation, fibrosis and respiratory cancer are recommended for the more in-depth studies of the mature CNF product. |
Posted: 03 Sep 2012 12:00 AM PDT A new steroidal saponin, named pumilum A (1), and a new phenolic glycoside, threo-1-(4′-hydroxy-2′-methoxyphenyl)-2-(2′′,4′′-dihydroxyphenyl)-1,3-propanediol-4′-O-β-D-glucopyranoside (7) were isolated from the methanolic extract of the bulbs of Lilium pumilum DC, along with five known steroidal saponins. Their chemical structures were elucidated on the basis of detailed spectroscopic analysis, including 1D and 2D NMR techniques and chemical methods. In addition, the inhibitory activity of all the isolates on Na+/K+ ATPase was evaluated. |
Posted: 03 Sep 2012 12:00 AM PDT In the present study a universally applicable HPLC-DAD/ESI-MS/MS method was developed for carrying out the comprehensive characterization of Jitai tablets (JTT). Based on the ESI-MSn fragmentation patterns of the reference standards, a total of 101 components were identified or tentatively characterized by comparing their retention times, UV and MS spectra with those of reference standards or through the matching of empirical information with those of published components in the in-house library. The characteristic fragmentation pattern of alkaloids, phenolic acids, tanshinones, flavonoid glycosides, cyanogenic glycosides, ginsenosides, 2-(2-phenylethyl) chromones, phthalides and gingerol-related compounds were tentatively elucidated using structurally-relevant product ions. It was observed that neutral losses of C9H10O3 and C9H8O2 were the characteristic product ions of scopola alkaloids. Neutral fragment mandelonitrile was the characteristic ion of cyanogenic glycosides. To our knowledge, tropylium ion and C4H2O unit were the characteristic ions of 2-(2-phenylethyl) chromone, which resulted from the Retro-Diels-Alder (RDA) cleavage of the C ring. The results indicated that the developed analysis method could be employed as a rapid, effective technique for structural characterization of chemical constituents in TCM. This work is expected to provide comprehensive information for the quality evaluation and pharmacokinetic studies of JTT. |
Posted: 03 Sep 2012 12:00 AM PDT Commercial plant essential oils obtained from 11 Myrtaceae plant species were tested for their fumigant antifungal activity against Aspergillus ochraceus, A. flavus, and A. niger. Essential oils extracted from Leptospermum petersonii at air concentrations of 56 × 10−3 mg/mL and 28 × 10−3 mg/mL completely inhibited the growth of the three Aspergillus species. However, at an air concentration of 14 × 10−3 mg/mL, inhibition rates of L. petersonii essential oils were reduced to 20.2% and 18.8% in the case of A. flavus and A. niger, respectively. The other Myrtaceae essential oils (56 × 10−3 mg/mL) only weakly inhibited the fungi or had no detectable affect. Gas chromatography-mass spectrometry analysis identified 16 compounds in L. petersonii essential oil. The antifungal activity of the identified compounds was tested individually by using standard or synthesized compounds. Of these, neral and geranial inhibited growth by 100%, at an air concentration of 56 × 10−3 mg/mL, whereas the activity of citronellol was somewhat lover (80%). The other compounds exhibited only moderate or weak antifungal activity. The antifungal activities of blends of constituents identified in L. petersonii oil indicated that neral and geranial were the major contributors to the fumigant and antifungal activities. |

Posted: 03 Sep 2012 12:00 AM PDT Simple synthesis of modafinil derivatives and their biological activity are described. The key synthetic strategies involve substitution and coupling reactions. We determined the anti-inflammatory effects of modafinil derivatives in cultured BV2 cells by measuring the inhibition of nitrite production and expression of iNOS and COX-2 after LPS stimulation. It was found that for sulfide analogues introduction of aliphatic groups on the amide part (compounds 11a–d) resulted in lower anti-inflammatory activity compared with cyclic or aromatic moieties (compounds 11e–k). However, for the sulfoxide analogues, introduction of aliphatic moieties (compounds 12a–d) showed higher anti-inflammatory activity than cyclic or aromatic fragments (compounds 12e–k) in BV-2 microglia cells. |

Brothers in Arms: Furious 4 to be reimagined into new IP Posted: 02 Sep 2012 08:13 PM PDT Gearbox says game's new direction will see it move away from the Brothers in Arms franchise into all-new territory.
Gearbox Software president Randy Pitchford has announced the upcoming Brothers in Arms: Furious 4 will shed its Brothers in Arms legacy to be reimagined into an all-new IP.
Speaking at a PAX Prime panel in Seattle this week, (and as originally reported by Polygon), Pitchford says the original idea for the upcoming game evolved away from the core of the Brothers in Arms franchise and needed to be "unshackled" from the brand. Pitchford confirmed the upcoming title will be an all-new IP, revealing more details about the new game will be made public at Gearbox's upcoming community day in Dallas, Texas on September 15. Earlier this year, Gearbox announced the release of Brothers in Arms: Furious 4 would be delayed to 2013. At the time, Pitchford said development on the game had evolved and that gamers would be "surprised by what they see." Brothers in Arms: Furious 4 was to be the first new instalment in the Brothers in Arms franchise since 2008's well-received Brothers in Arms: Hell's Highway. The game was set to take place during World War II following a band of four soldiers chasing Hitler.
