Diagnostics, Vol. 3, Pages 155-169: A Disposable Microfluidic Virus Concentration Device Based on Evaporation and Interfacial Tension

We report a disposable and highly effective polymeric microfluidic viral sample concentration device capable of increasing the concentration of virus in a human nasopharyngeal specimen more than one order of magnitude in less than 30 min without the use of a centrifuge. The device is fabricated using 3D maskless xurography method using commercially available polymeric materials, which require no cleanroom operations. The disposable components can be fabricated and assembled in five minutes. The device can concentrate a few milliliters (mL) of influenza virus in solution from tissue culture or clinical nasopharyngeal swab specimens, via reduction of the fluid volume, to tens of microliters (mL). The performance of the device was evaluated by nucleic acid extraction from the concentrated samples, followed by a real-time quantitative polymerase chain reaction (qRT-PCR). The viral RNA concentration in each sample was increased on average over 10-fold for both cultured and patient specimens compared to the starting samples, with recovery efficiencies above 60% for all input concentrations. Highly concentrated samples in small fluid volumes can increase the downstream process speed of on-chip nucleic acid extraction, and result in improvements in the sensitivity of many diagnostic platforms that interrogate small sample volumes.

The aim of the present editorial is to briefly summarize the current scientific and technological accomplishments in the field of organic electronic biosensors as described in the articles published in this Special Issue. By definition, a biosensor is a robust analytical device that combines a biological recognition element (e.g., antibodies, enzymes, cells) with a transducer. Organic electronic bio-devices are considered as potentially reliable substitutes of conventional and rather expensive analytical techniques employed for several applications such as medical diagnosis, food safety and environment pollution monitoring. Some insights into the selection and immobilization of recognition elements, signal amplification, fabrication techniques and analytical performance of biosensing devices will be presented.

Water is both a key and limited resource in the Okavango Catchment of Southern Africa. It is vital for the ecosystem and the three riparian states Angola, Botswana and Namibia who use the water of the catchment for multiple purposes including pastoralism, farming and tourism. Socioeconomic changes, primarily strong population growth and increasing development demands pose significant challenges for the Okavango Catchment and its Integrated Water Resource Management (IWRM). In this paper, we first review the socioeconomic background and the current and projected water situation. Against this background, we analyze the dependence of the riparian states and the local livelihoods on the Okavango Catchment. Third, we discuss the implications of socioeconomic changes and increased water demand for the IWRM in the catchment. We review the scientific literature and relevant reports. Further we utilize (geo-spatial) analysis of socioeconomic, livelihood and hydrological data, supplemented by a field visit to Namibia and Botswana. Our findings suggest that strong population growth and the stabilization of Angola are likely to increase the pressure to develop the region along the Okavango. The central challenge for IWRM is hence to enable Angola to meet its development needs without limiting livelihood and economic prospects in Botswana and Namibia.

Merkel cell carcinoma (MCC) is an aggressive neuroendocrine skin cancer. The Merkel cell polyomavirus (MCPyV) is implicated in its pathogenesis. Immune mechanisms are also implicated. Patients who are immunosuppressed have an increased risk. There is evidence that high intratumoral T-cell counts and immune transcripts are associated with favorable survival. Spontaneous regressions implicate immune effector mechanisms. Immunogenicity is also supported by observation of autoimmune paraneoplastic syndromes. Case reports suggest that immune modulation, including reduction of immune suppression, can result in tumor regression. The relationships between MCPyV infection, the immune response, and clinical outcome, however, remain poorly understood. Circulating antibodies against MCPyV antigens are present in most individuals. MCPyV-reactive T cells have been detected in both MCC patients and control subjects. High intratumoral T-cell counts are also associated with favorable survival in MCPyV-negative MCC. That the immune system plays a central role in preventing and controlling MCC is supported by several observations. MCCs often develop, however, despite the presence of humoral and cellular immune responses. A better understanding on how MCPyV and MCC evade the immune response will be necessary to develop effective immunotherapies.

In Washington, over fifty percent of the wheat produced under rainfed conditions receives less than 300 mm of annual precipitation. Hence, a winter wheat-summer fallow cropping system has been established to obtain adequate moisture for winter wheat production. Current tilled fallow systems receive significant soil erosion through both wind and water. As a result, no-till chemical fallow systems are being adopted to mitigate erosion concerns. The objective of this study was to evaluate current Pacific Northwest cultivars under no-till chemical fallow and tilled fallow systems to identify cultivars adapted to a late-planted no-till system. Twenty-one cultivars were planted in a split-plot design with fallow type as the main plot and genotype as the sub-plot. Four replications were planted at two locations over three years. Data was collected on heading date, grain yield and grain volume weight. Analysis of variance was conducted on data from each year and location. Results were significant for all traits. Cultivars in the late-planted no-till system yielded an average of 39% less than the tilled fallow system. It is evident that cultivars vary in their adaptability and yield stability across production systems. Chukar and Eltan displayed the highest levels of yield stability, and growers who wish to plant winter wheat in a late-planted no-till system may benefit from choosing these cultivars.

This paper presents an energy-efficient solar energy harvesting and sensing microsystem that harvests solar energy from a micro-power photovoltaic module for autonomous operation of a gas sensor. A fully integrated solar energy harvester stores the harvested energy in a rechargeable NiMH microbattery. Hydrogen concentration and temperature are measured and converted to a digital value with 12-bit resolution using a fully integrated sensor interface circuit, and a wireless transceiver is used to transmit the measurement results to a base station. As the harvested solar energy varies considerably in different lighting conditions, in order to guarantee autonomous operation of the sensor, the proposed area- and energy-efficient circuit scales the power consumption and performance of the sensor. The power management circuit dynamically decreases the operating frequency of digital circuits and bias currents of analog circuits in the sensor interface circuit and increases the idle time of the transceiver under reduced light intensity. The proposed microsystem has been implemented in a 0.18 µm complementary metal-oxide-semiconductor (CMOS) process and occupies a core area of only 0.25 mm2. This circuit features a low power consumption of 2.1 µW when operating at its highest performance. It operates with low power supply voltage in the 0.8V to 1.6 V range.

A smoke plume originating from the massive wildfires near Moscow was clearly detected in northern Finland on 30 July 2010. Measurements made with remote sensing instruments demonstrated how the biomass burning aerosols affected the chemical and optical characteristics of the atmosphere in regions hundreds of kilometers away from the actual fires. In this study, we used MODIS, AIRS, CALIOP, PFR, ceilometers, FTS and Brewer data to quantify the properties of the transported smoke plume. In addition, in situ measurements of aerosol concentration (DMPS), absorption (aethalometer) and scattering (nephelometer) are presented. We found that due to the smoke plume in northern Finland, the daily averaged optical thickness of aerosols increased fourfold, and MODIS retrieved AOD as high as 4.5 for the thickest part of the plume. FTS measurements showed that CO concentration increased by 100% during the plume. CALIOP and ceilometer measurements revealed that the smoke plume was located close to the surface, below 3 km, and that the plume was not homogeneously mixed. In addition, in situ measurements showed that the scattering and absorption coefficients were almost 20 times larger in the smoke plume than on average, and that the number of particles larger than 320 nm increased 14-fold. Moreover, a comparison with in situ measurements recorded in eastern Finland on the previous day showed that the transport from eastern to northern Finland decreased the scattering coefficient, black carbon concentration, and total number concentration 0.5%/h, 1.5%/h and 2.0%/h, respectively.

In the spring of 2006, four human cases of parapoxvirus infections in Missouri residents were reported to the Centers for Disease Control and Prevention (CDC), two of which were initially diagnosed as cutaneous anthrax. This investigation was conducted to determine the level of recognition of zoonotic parapoxvirus infections and prevention measures, the degree to which veterinarians may be consulted on human infections and what forces were behind this perceived increase in reported infections. Interviews were conducted and clinical and environmental sampling was performed. Swab and scab specimens were analyzed by real-time polymerase chain reaction (PCR), whereas serum specimens were evaluated for parapoxvirus antibodies. Three case patients were found to have fed ill juvenile animals without using gloves. Forty-six percent of veterinarians reported having been consulted regarding suspected human orf infections. Orf virus DNA was detected from five of 25 asymptomatic sheep. Analysis of extracellular envelope gene sequences indicated that sheep and goat isolates clustered in a species-preferential fashion. Parapoxvirus infections are common in Missouri ruminants and their handlers. Infected persons often do not seek medical care; some may seek advice from veterinarians rather than physicians. The initial perception of increased incidence in Missouri may have arisen from a reporting artifact stemming from heightened concern about anthrax. Asymptomatic parapoxvirus infections in livestock may be common and further investigation warranted.

Potatoes are regarded as a significant antioxidant source in human nutrition. However, different types of environmental stress may affect the level of antioxidants in their tuber tissue. In this study, two purple breeding clones and the yellow fleshed cultivar (cv.) Agave were grown in the glasshouse under control with drought stress conditions for two consecutive years. After harvest, the tubers were analysed for concentrations of antioxidants measured as ascorbic acid equivalent (ACE) and trolox equivalent (TXE) in fresh tissue and after wounding. In addition, the peroxidase enzyme (POD) activities and total amounts of anthocyanins (Ac) were assayed. Drought stress caused a significant decrease in tuber yield but had no significant effect on Ac, POD, ACE and TXE. Wounding stress significantly induced the POD activity in control and drought stressed tubers of all genotypes. Also the ACE and TXE were notably increased by wounding in cv. Agave. This was less pronounced in the purple clones which in general displayed a higher level of antioxidants. The results revealed significant differences between genotypes and that the effect of drought stress on the level of antioxidants is smaller than that of wounding stress.

Climate change is already altering the landscape at high latitudes. Permafrost is thawing, the growing season is starting earlier, and, as a result, certain regions in the Arctic may be subjected to an increased incidence of freeze-thaw events. The potential release of carbon and nutrients from soil microbial cells that have been lysed by freeze-thaw transitions could have significant impacts on the overall carbon balance of arctic ecosystems, and therefore on atmospheric CO2 concentrations. However, the impact of repeated freezing and thawing with the consequent growth and recrystallization of ice on microbial communities is still not well understood. Soil samples from three distinct sites, representing Canadian geographical low arctic, mid-arctic and high arctic soils were collected from Daring Lake, Alexandra Fjord and Cambridge Bay sampling sites, respectively. Laboratory-based experiments subjected the soils to multiple freeze-thaw cycles for 14 days based on field observations (0 °C to −10 °C for 12 h and −10 °C to 0 °C for 12 h) and the impact on the communities was assessed by phospholipid fatty acid (PLFA) methyl ester analysis and 16S ribosomal RNA gene sequencing. Both data sets indicated differences in composition and relative abundance between the three sites, as expected. However, there was also a strong variation within the two high latitude sites in the effects of the freeze-thaw treatment on individual PLFA and 16S-based phylotypes. These site-based heterogeneities suggest that the impact of climate change on soil microbial communities may not be predictable a priori; minor differential susceptibilities to freeze-thaw stress could lead to a "butterfly effect" as described by chaos theory, resulting in subsequent substantive differences in microbial assemblages. This perspectives article suggests that this is an unwelcome finding since it will make future predictions for the impact of on-going climate change on soil microbial communities in arctic regions all but impossible.

Almost all transfection protocols for mammalian cells use a drug resistance gene for the selection of transfected cells. However, it always requires the characterization of each isolated clone regarding transgene expression, which is time-consuming and labor-intensive. In the current study, we developed a novel method to selectively isolate clones with high transgene expression without drug selection. Porcine embryonic fibroblasts were transfected with pCEIEnd, an expression vector that simultaneously expresses enhanced green fluorescent protein (EGFP) and endo-b-galactosidase C(EndoGalC; an enzyme capable of digesting cell surface a-Gal epitope) upon transfection. After transfection, the surviving cells were briefly treated with IB4SAP (a-Gal epitope-specific BS-I-B4 lectin conjugated with a toxin saporin). The treated cells were then allowed to grow in normal medium, during which only cells strongly expressing EndoGalC and EGFP would survive because of the absence of a-Gal epitopes on their cell surface. Almost all the surviving colonies after IB4SAP treatment were in fact negative for BS-I-B4 staining, and also strongly expressed EGFP. This system would be particularly valuable for researchers who wish to perform large-scale production of therapeutically important recombinant proteins.

A stripped no-core optical fiber with a 125 µm diameter was transformed into a symmetric and unbroken optical fiber that tapers slightly to a 45-µm-diameter waist. The laser light can be easily launched into the no-core optical fiber. The enhanced evanescent wave of the slightly tapered no-core optical fiber can attract nearby 5-µm-diameter polystyrene microparticles onto the surface of the tapered multimode optical fiber within fast flowing fluid and propel the trapped particles in the direction of the light propagation to longer delivery range than is possible using a slightly tapered telecom single-mode optical fiber.

This paper proposes a novel and autonomous weighing system for wild animals. It allows evaluating changes in the body weight of animals in their natural environment without causing stress. The proposed system comprises a smart scale designed to estimate individual body weights and their temporal evolution in a bird colony. The system is based on computational intelligence, and offers valuable large amount of data to evaluate the relationship between long-term changes in the behavior of individuals and global change. The real deployment of this system has been for monitoring a breeding colony of lesser kestrels (Falco naumanni) in southern Spain. The results show that it is possible to monitor individual weight changes during the breeding season and to compare the weight evolution in males and females.

This paper presents a multipurpose and low cost sensor for temperature control over the wine fermentation process, in order to steadily communicate data through wireless modules in real time to a viticulturist's mobile or fixed device. The advantage of our prototype is due to the fact that it will be used by small winemakers in the "Ribera del Duero" area, and as it is a cheaper sensor and easy to use for the control and monitoring of the grape fermentation process, it will probably be used by other business men with the same necessities in the region. The microcontroller MSP430G2553 is among the components that make up the sensor, that are integrated onto a motherboard. It communicates with the RN-42 Bluetooth module through an UART interface. After verifying that all elements are working correctly, the parts are assembled to form the final prototype. This device has been tested in a winery in the region, fulfilling the initial project specifications.
